Denne metoden kan bidra til å svare på viktige spørsmål i hepatotropiske infeksjoner og leversykdom felt. Aspirer alle medier pumpet ut av reservoarsiden av platen. Tilsett 1,4 milliliter hepatocytt såing medium til hver brønn. Deretter returnerer du platen til dokken for å bringe det totale volumet per godt opp til 1,6 milliliter.
Først varmer hepatocytter tiningsmediet og hepatocyte såing medium til 37 grader Celsius. Deretter tine ett hetteglass med primære menneskelige hepatocytter i henhold til leverandørens instruksjoner. I et glassrørskap, resuspend cellene i en milliliter hepatocyte seeding medium.

Overfør platen til dokkingstasjonen i en fuktet inkubator ved 37 grader Celsius og med 5%CO2. Start strømmen i nedoverretningen med en strømningshastighet på én mikroliter per sekund i åtte timer. Etter dette reverserer du strømmen til oppadgående retning med en strømningshastighet på ett mikroliter per sekund i åtte timer.
Overfør deretter platen til vaskedokken, og aspirerer alt medium fra brønnene. Tilsett 400 mikroliter hepatocyttvedlikeholdsmedium til hver brønn. Og start strømmen i nedoverretningen med en strømningshastighet på en mikroliter per sekund i 3 1/2 minutter.
Deretter aspirerer du alle medier fra reservoaret, og tilsett 1,4 milliliter hepatocyttvedlikeholdsmedium. Overfør platen til dokkingstasjonen i en fuktet inkubator ved 37 grader Celsius og 5%CO2. Start strømningen i oppadgående retning med en strømningshastighet på ett mikroliter per sekund i 48 timer.
Etter 48 timer vask platen ved å overføre den til vaskedokken. Aspirer alle medier fra brønnene. Og tilsett 400 mikroliter vedlikeholdsmedium.
Og start strømmen i nedoverretningen på ett mikroliter per sekund i 3,5 minutter. Deretter aspirerer alle medier som vises i reservoarsiden av brønnene. Tilsett 1,4 milliliter hepatocyttvedlikeholdsmedium.
Overfør deretter platen tilbake til dokkingstasjonen i den fuktede inkubatoren. Start strømningen i oppadgående retning med en strømningshastighet på ett mikroliter per sekund i 48 timer. Bytt medium ved hjelp av denne vaske-og-erstatt prosessen hver 48.
Primære menneskelige hepatocytter er vanligvis bare stabile i en begrenset periode når du bruker konvensjonelle kultursystemer. Men ved hjelp av protokollen som er beskrevet her, kan de vedlikeholdes funksjonelt i lengre perioder. Human albumin utskilles av funksjonelle hepatocytter, og anses å være den beste markøren for evaluering av leverfunksjonalitet.
Albumin er sett på som stabilt og høyt uttrykt av 3D-kulturer til minst dag 40 post-seeding. For co-kulturer evalueres Kupffer-cellenes funksjonalitet og levedyktighet ved å måle sekresjonen av spesifikke cytokiner. Som sett her indikerer de målte nivåene av IL-6 og TNF alfa at cellene var funksjonelt vedlikeholdt og levedyktige.
I tillegg til å beholde sin fysiologiske cellulær metabolisme, ble disse kulturene eksepsjonelt utsatt for HBV-infeksjon. HBV DNA og andre virale markører er lett påviselig fra dag to etter infeksjon. Mens konvensjonelle kulturer krever inokulasjon med minst 500 HBV genomekvivalenter supplert med DMSO og PEG, er disse 3D-kulturene infisert med så lite som 05 usupplementerte genomekvivalenter.
I tillegg til utskillede markører for virusinfeksjon, hentes hepatocyteholdige stillas fra disse kulturene. Immunofluorescence mikroskopi avslører at disse stillasene inneholder virale antigener. Mens du prøver denne prosedyren, er det viktig å huske å være forsiktig når du håndterer hepatocytter før seeding for å unngå celledød.
I tillegg er det viktig å sikre at platene er riktig montert og alle strømningskanalene tillater riktig mediesirkulasjon før såing av hepatocytter. Etter denne prosedyren kan andre metoder som immunofluorescence utføres for å svare på flere spørsmål. Inkludert plasseringen og frekvensen av infiserte celler i vevet.
Den fysiologiske tilstanden til hepatocytter kan også vurderes ved farging for albumin eller leverstrukturer, inkludert tett kobling protein ZO-1. Etter denne utviklingen baner denne teknikken vei for forskere innen virologi for å utforske hepatitt B-infeksjon og immunresponser i et fysiologisk 3D-modellsystem. Ikke glem at arbeid med hepatitt B-virus kan være farlig, og forholdsregler som tidligere vaksinasjon, bruk av passende PPE, og arbeid under passende biosikkerhetsretningslinjer bør alltid tas mens du utfører denne prosedyren.
Målet med denne protokollen er å gi en trinnvis veiledning for å utføre eksperimenter 3D "lever på-chip" infeksjon med hepatitt B-viruset.
