この方法は、シグナル伝達経路が異なる刺激や薬物によってどのように影響を受けるかなど、細胞生物学や免疫学の分野における重要な質問に答えるのに役立ちます。この手法の主な利点は、単一セルシグナル伝達解析を中~高スループット方式で実行できることです。補充されたRPMI 16/40培地で細胞を再懸濁した後、必要量の細胞懸濁液を96ウェルVボトムプレートのウェルに移す。プレートを予熱した摂氏37度の水浴に移し、そこで10分間休ませます。その後、新しい96ウェルプレートの各ウェルに固定バッファの60マイクロリットルを1つ追加し、固定プレートを設定します。このプレートを摂氏37度の水浴に入れます。50マイクロリットルのコントロールサンプルをプレートに移し、上下にピペットで混ぜます。必要に応じて、ミリリットルの抗IgMあたり10マイクロリットルを細胞に加え、シミュレーションタイムコースを開始し、上下にピペットで混合します。50マイクロリットルのサンプルを各時点でフィックスプレートに移し、各サンプルを加えるにつれて混合する。最後のサンプルを加えた後、10分間水浴中に固定プレートを残します。まず、細胞をPBSで3回洗浄する。5分間Gの500倍のプレートを遠心し、上清を捨てます。次に、バーコーディング試薬を用いてフレッシュな96ウェルVボトムプレートを調製し、各々のサンプルを染色するのに必要なウェルの数を各ウェルに各バーコード試薬の5マイクロリットルをピペット化して、各サンプルを染色するのに必要なウェルの数を充填する。固定セルプレートでは、各ウェルの細胞を190マイクロリットルのPBSで再懸濁する。次に、各ウェルの細胞をバーコードプレート内の対応するウェルに移し、各ウェルをよく混合する。プレートは暗闇の中で20分間室温で休ませます。5分間Gの500倍のプレートを遠心し、上清を捨てます。この後、フロー洗浄で各ウェルの細胞を2回洗います。その後、細胞の各ウェルに190マイクロリットルのフローウォッシュを加えます。バーコード化されたサンプルをすべて15ミリリットルチューブに結合し、各補償制御を別の1.7ミリリットルチューブに移します。500回Gで5分間遠心分離機を、上清を捨てる。開始するには、パーマバッファー3の2ミリリットルを15ミリリットルチューブに移します。マイナス20度で保管して、使用すると氷が冷たくなります。準備ができたら、セルの凝集を避けるために渦を打ちながら、バーコード化された細胞集団を含む15ミリリットルのチューブに1.5ミリリットルの氷冷パーマバッファーを加えます。同じ方法で各補償制御に氷冷パーマバッファーの100マイクロリットルを追加します。すぐに最低30分間マイナス80度で冷凍庫にセルを移します。抗原染色を開始する準備ができたら、冷凍庫から細胞を取り除き、氷の箱に移します。過剰なフロー洗浄で細胞を3回洗います。5分間500倍Gと摂氏4度で細胞を遠心分離します。上清を捨てて、流量洗浄の中でバーコード化された細胞集団を再中断し、例えばリン酸化抗体染色当たり25マイクロリットルの細胞懸濁液があるということである。200マイクロリットルのフロー洗浄で補償制御を再中断します。染色用の抗体の調製を開始するには、フロー洗浄で希釈した10マイクロリットルのリン酸特異的抗体を、新鮮な96ウェルVボトムプレートの各ウェルに加えます。表面マーカー15マイクロリットルを加え、フローウォッシュで希釈し、各ウェルに25マイクロリットルのセルサスペンションを加え、井戸あたり50マイクロリットルの最終体積を確保します。細胞は暗闇の中で室温で30分間休ませます。この後、染色した細胞を流れ洗いで2回洗います。5分間Gの500倍で遠心分離機。上清を捨て、フロー洗浄の150マイクロリットルで細胞を再中断します。次に、テキストプロトコルで概説されているようにフローサイトメトリー解析を行います。提示されたプロトコルでは、3つのバーコード染料を組み合わせることにより3次元バーコードが行われる。個々のサンプルは、次に各バーコード試薬対側散乱領域の後続の格子によって分解される。B細胞受容体の下流にある20のシグナル伝達分子の基底および刺激誘導リン酸化レベルは、次いで、CLL患者からのB細胞および様々な条件下での正常な制御において特徴づけられる。stat3ホスホチロシン705は、大幅に上昇する規制が見られる。細胞は、B細胞受容体経路を介して誘導されるシグナル伝達収差を同定するために、最大30分間抗IgMで刺激される。変異していないIGVH患者からのCLL細胞は抗IgM刺激に対する感受性を増加させるが、AKT蛍光体セリン473に対して統計的に有意である。次に細胞はPI 3キナーゼデルタ阻害剤イデラリシブに曝され、異常なAKT pS473シグナルが逆転できるかどうかを試験します。ここに示すように、異常レベルは、CLL細胞における異常シグナル伝達を正常化するためにキナーゼ阻害剤を適用できることを示す濃度依存的な方法での治療時に有意に低下する。この手順を試みる前に、バーコード試薬と抗体をティンテリングし、選択した表面マーカーが固定および透過性試薬と互換性があることを確認することが重要です。細胞は、蛍光体分析のために懸濁液である必要があるが、それでもプロトコルは、全体のトリプジネーションのような単一細胞懸濁液を得るために手順に従って異常細胞または組織に適応させることができる。
