Phospho-flowcytometri er en kraftfull teknikk å måle protein fosforylering nivå i enkeltceller. Siden metoden er avhengig av farging med antistoffer, begrenses phospho flowcytometri antistoff-tilgjengelighet. Phospho-flowcytometri brukes til å analysere protein fosforylering nivå encellede oppløsning. Det overordnede målet med metoden er å tilordne mobilnettet signalnettverk mønstre angitte vilkår. Ved å utnytte flowcytometri multiparameter kapasitet, kan flere signalveier analyseres samtidig i forskjellige undergrupper av en heterogen celle befolkningen som perifert blod. Disse trekkene tilbyr fordeler over andre antistoff-baserte teknologier som immunohistochemistry, enzym knyttet immunosorbent analysen (ELISA), protein matrise og omvendt fase protein matrise (RPPA)1. Phospho flowcytometri kan kombineres med fluorescerende celle barcoding (FCB), som betyr at enkeltcelle prøver er merket med unike signaturer av fluorescerende fargestoffer slik at de kan være blandet sammen, beiset og analyseres som en enkelt prøve2. Dette reduserer antistoff forbruket øker data robusthet gjennom kombinasjonen av kontroll og behandlet prøver og forbedrer hastigheten på oppkjøpet. Kombinert FCB befolkningen kan deretter delt inn i mindre prøver og farget med opp til 35 forskjellige phospho-spesifikke antistoffer, avhengig av starter materiale. Store profilering eksperimenter kan dermed kjøres med standard cytometer maskinvare. Phospho-flowcytometri brukes på profil signalnettverk trasé i pasientprøvene fra flere Hematologisk kreft inkludert kronisk lymfatisk leukemi (CLL)3,4,5, akutt myelogen leukemi (AML) 6 og non-Hodgkin lymfomer7. Phospho flowcytometri er dermed en effektiv tilnærming til å karakterisere signalnettverk avvik, identifisere og validere biomarkers og vurdere pharmacodynamics.

phospho flow cytometry with fluorescent cell barcoding for single cell signaling analysis and biomarker discovery

Avvikende celle signalisering spiller en sentral rolle i hudkreft og progresjon. Mest romanen målrettet terapi er faktisk rettet mot proteiner og protein funksjoner, og cellen signalnettverk avvik kan fungere som biomarkers å angi personlige behandlingstilbud. I motsetning til DNA og RNA analyser, kan endringer i protein aktivitet mer effektivt vurdere mekanismene bak narkotika følsomhet og motstand. Phospho-flowcytometri er en kraftfull teknikk som måler protein fosforylering hendelser på cellenivå, en viktig funksjon som skiller denne metoden fra andre antistoff tilnærminger. Metoden tillater samtidig analyse av flere signalnettverk proteiner. Denne metoden kan bidra til å svare på viktige spørsmål innen cellebiologi og immunologi som hvordan er signalveier påvirket av forskjellige stimuli eller narkotika? Den største fordelen med denne teknikken er at den lar deg utføre enkeltcellesignalanalyse i middels til høy gjennomstrømning. Etter re-suspendere cellene i supplert RPMI 16/40 medium, overføre den nødvendige mengden celle suspensjon til brønnene av en 96 brønn V-bunnplate.Overfør platen til et forvarmet 37 graders Celsius vannbad og la den hvile der i 10 minutter. Tilsett deretter 60 mikroliter med fast buffer en til hver brønn av en ny 96 brønnplate for å sette opp festeplaten. Plasser denne platen i 37 grader Celsius vannbad.Overfør en 50 mikroliter kontrollprøve til platen og bland ved å pipetter opp og ned. Eventuelt legger du til 10 mikroliter per milliliter anti-IgM til cellene for å starte simuleringstidskurset og bland ved å pipettere opp og ned. Etter dette vasker du de beisede cellene to ganger med strømningsvask. Sentrifuge ved 500 ganger G i fem minutter.Kast de overnaturante og suspender cellene i 150 mikroliter strømningsvask. Utfør deretter flytcytometrianalyse som beskrevet i tekstprotokollen. I den presenterte protokollen utføres tredimensjonal barkoding ved å kombinere tre barkodingfargestoffer.Individuelle prøver blir deretter de-convoluted av påfølgende gating på hver barcoding reagens versus side scatter område. Basal- og stimuleringsinduserte fosforyleringsnivåene på 20 signalmolekyler nedstrøms av B-cellereseptoren karakteriseres deretter i B-celler fra KLL-pasienter og normale kontroller under ulike forhold. Stat3 fosfo tyrosin 705 er sett på som betydelig oppregulert.Celler stimuleres med anti-IgM i opptil 30 minutter for å identifisere signalaberrasjoner indusert gjennom B-cellereseptorbanen. Mens KLL-celler fra pasienter med uforandert IGVH viser økt følsomhet overfor anti-IgM stimulering, er det bare statistisk signifikant for AKT fosfor serin 473. Celler blir deretter utsatt for PI 3-kinase delta inhibitor idelalisib for å teste om avvikende AKT pS473 signalet kan reverseres.Som vist her, reduseres avvikende nivåer betydelig ved behandling på en konsentrasjonsavhengig måte som viser at kinasehemmere kan brukes til å normalisere avvikende signalering i KLL-celler. Før du prøver denne prosedyren er det viktig å huske å titrere barcoding reagenser og antistoffer, og for å kontrollere at valgte overflatemarkører er kompatible med fiksering og permeabilisering reagenser. Cellene må være i suspensjon for fosforanalyse, fortsatt protokollen kan tilpasses avvikende celler eller vev følgende prosedyrer for å oppnå enkeltcellesuspensjon, for eksempel hele trypzination.
