この方法は、白血病細胞における代謝経路の設定に関する重要な質問に答えることができます。この技術の主な利点は、生細胞で一次白血病細胞の代謝をリアルタイムで測定できることです。まず、PBSで白血病患者から得られた骨髄サンプルを1対1の比率で希釈することから始めます。慎重に、15ミリリットル円錐形チューブで作りたての密度勾配培地の6ミリリットルにわたって希釈された骨髄サンプルの6ミリリットルを層し、密度勾配遠心分離によって細胞を分離する。パスツールピペットを使用して、単核細胞の界面層を5ミリリットルのPBSを含む新しい50ミリリットルの円錐管に慎重に移し、遠心洗浄を行います。次いで、単核細胞ペレットを、カウント用の滅菌PBSの2ミリリットルで再懸濁する。1ミリリットル当たり7個の細胞に3倍10倍に希釈します。そして、摂氏37度と5%CO2で16〜24時間インキュベーションのために20ミリリットルのRPMI培地を含む2つのT75フラスコのそれぞれに1ミリリットルの細胞を加えます。一方、細胞外フラックス分析用のプレートを準備するために、2つのウェル、8ウェル細胞外フラックスアナライザプレートの各ウェルに12.5マイクロリットルの細胞接着溶液を追加します。20分後、細胞接着剤を吸引し、洗浄ごとに200マイクロリットルの無菌水でそれぞれをよく2回洗浄します。2回目の洗浄後、プレートをフードに入れて、井戸が乾くまで残します。センサーカートリッジをラボベンチの上に逆さまに置きます。フラックスアナライザのユーティリティプレートとセンサーカートリッジを分離します。そして、ユーティリティプレートの各井戸に200マイクロリットルのキャリブラントを充填し、各堀を400マイクロリットルのキャリブラントで井戸の外側に埋めます。キャリブラントが入ったユーティリティプレートにセンサーカートリッジを戻し、CO2を一晩も入れずに加湿37°Cのインキュベーターにカートリッジアセンブリを入れます。その後、細胞外フラックス分析装置をオンにし、一晩で摂氏37度まで暖めます。翌朝、フラスコから50ミリリットルの円錐管に細胞を移して遠心分離する。そして、ペレットをカウントするための適切な実験培地の1ミリリットルで再懸濁する。実験培地濃度の400マイクロリットルで6細胞に4倍10で細胞を再懸濁する。そして、フラックス分析板のGを通してウェルBに細胞の50マイクロリットルを加える。そして、実験培地の180マイクロリットルを、バックグラウンド制御井戸としてウェルAおよびHに入る。遠心分離後、実験培地の130マイクロリットルをG.を通してウェルBにゆっくりと慎重に加え、顕微鏡下の各ウェルの底部に細胞の安定な付着を視覚的に確認する。その後、CO2なしで加湿した37°Cインキュベーターに30分間、フラックス分析プレートを戻します。インキュベーション終了の20分前に、表に示すように実験プロトコルに従ってカートリッジの適切なインジェクターポートに化合物をロードする。次に、適切な細胞外フラックス解析プログラムを設定します。プログラムを起動し、プロンプトが表示されたら、校正用プレートをアッセイプレートに交換します。解糖解析のストレステストでは、細胞が栄養素を奪われるように、基底培地のみが使用されます。得られた第1のパラメータは、細胞に貯蔵されたグルコース量を反映すべき基底酸性化である。最初の注射の後、細胞がグルコースを利用するにつれて細胞外酸性化速度が上昇し、グルコースを乳酸に発酵させることができる。第2の注射におけるオリゴマイシンAはATP合成酵素を阻害し、したがって細胞外酸性化率のさらなる上昇を引き起こす解糖によってATPを産生するように細胞を導く。2-デオキシDグルコースの注入は完全に解糖を阻害し、細胞外酸性化率が低下する。細胞の糸球菌ストレス試験では、グルタミンとグルコースを添加した培地を用い、細胞が全ての栄養素を奪われないようにする。基礎呼吸パラメータは、その基底代謝状態を反映しています。オリゴマイシンAによる最初の注射の後、細胞はミトコンドリア呼吸を阻害し、酸素消費率の低下として表される解糖に切り替える。しかし、FCCPの2回目および3回目の注射は、細胞が現在最大速度で酸素を消費するように、呼吸からATP産生を切り離す。そして、酸素消費率は最高値に上昇します。ロテノーンとアンチマイシンA混合物の最後の注入は、酸素消費率をほぼゼロに減少させるミトコンドリア呼吸を完全に阻害する。この手順を試みる間、白血病芽細胞の代謝パラメータのみを測定するために、サンプル中の爆風の割合を評価することが重要です。この方法を実装する際には、栽培条件やデータの正規化に慎重に最適化を適用する必要があります。
