この方法は、材料科学と生物医学に幅広い応用を有するナノダイヤモンドの効果的な表面機能化に関する重要な質問に答える助けとなる。この方法はナノダイヤモンドに使用できます。また、金属ナノ粒子、磁性ナノ粒子、または活性バイオポリマーコーティングを必要とする表面などの他の材料にも適用できます。この方法では、ナノダイヤモンドは、普遍的な接着剤であるポリドーパミンのコーティングでファンタニオン化されます。PDA層の厚さは、ドーパミンの濃度を変化させることによって十分に制御される。まず、30.29グラムのトリスHCl粉末を100マイクロリットルの脱イオン水に溶解します。溶液を250ミリリットルの体積フラスコに移します。フラスコを脱イオン水でラインに充填し、それを混ぜて1つのモルトリスHClバッファーを得る。このストック溶液から、シリアル希釈により0.1モルトリスHClバッファーの20ミリリットルを調製します。pHメーターでバッファーを監視しながら、1モル塩酸を使用してpHを8.5に調整します。次に、100ナノメートルの単結晶ナノダイヤモンドを1ミリリットルに1ミリリットルに1ミリリットルの0.02ミリリットルをpH 8.5トリスバッファーで1ミリリットルに希釈する。混合物を10分間攪拌し、1ミリリットルナノダイヤモンド懸濁液あたり0.02ミリグラムを得る。次いで、2ミリリットルのpH 8.5トリスバッファーに2ミリリットルのドーパミン塩酸塩を溶解し、30秒間ボルテックスすることにより、1ミリリットルのドーパミン塩酸塩溶液あたり10ミリグラムの緩衝液を得た。ナノダイヤモンド溶液に調製したドーパミン溶液の5、7.5、または10マイクロリットルを加え、最終的なドーパミン塩酸塩濃度が50、75、または1ミリリットル当たり100ミリグラムであるかに応じて。反応量を調整した後、暗闇の中で12時間摂氏25度で激しく攪拌する。次いで、ポリドーパミンコーティングナノダイヤモンドの懸濁液を1.5ミリリットル遠心分離チューブに移し、16,000gで遠心分離機を2時間行う。上清を取り除き、ナノダイヤモンドを16,000gで1ミリリットルの部分で3回洗浄します。次に、洗浄された固体に200マイクロリットルの脱イオン水を加え、混合物を30秒間超音波処理してポリドーパミンコーティングされたナノダイヤモンドを再分散させます。ポリドーパミンコーティングされたナノダイヤモンドの懸濁液を40マイクロリットルを脱イオン水で2回連続して希釈する。次いで、10ミリリットルの硝酸銀を渦水で10ミリリットルに溶解する。ヒュームフードでは、硝酸銀溶液に1つの大臼歯水性アンモニアを加え、滴下し、黄色の沈殿物が形成されるまで、定期的に溶液を振る。沈殿が消失するまでアンモニアを添加し続け、水酸化ジアミン銀の溶液を得る。希釈したナノダイヤモンド分散液の40マイクロリットルにダイアミン銀溶液の4.3または6.4マイクロリットルを直ちに加え、それぞれ0.4または0.6ミリグラムの最終濃度を1ミリリットル当たりにする。この後、脱イオン水で100マイクロリットルにボリュームを調整します。混合物を10分間超音波処理します。次いで、分散液を16,000gで15分間遠心し、遊離銀イオンを除去する。上清を捨て、銀ナノ粒子で装飾されたポリドーパミンコーティングされたナノダイヤモンドを、16,000gで100マイクロリットル分の100マイクロリットル部分に3回、毎回5分間洗浄します。銀ナノ粒子で装飾されたナノダイヤモンドに100マイクロリットルの脱イオン水を加え、30秒間超音波処理によって再分散させます。250~550ナノメートルのUV-Vis分光スキャンでナノダイヤモンドを特徴付けます。次に、銀ナノ粒子で装飾されたナノダイヤモンドの5マイクロリットルをプラズマ洗浄されたカーボンコーティングされた銅グリッドに堆積させ、3分間座らせます。その後、フィルターペーパーで余分な溶液を吸い取ります。各グリッドを1滴の脱イオン水を塗布し、15秒間座らせてから、ろ紙でくねくねします。透過型電子顕微鏡でサンプルを可視化する前に、グリッドを空気乾燥させます。コーティングされていないナノダイヤモンドはマイクロクラスターと凝集体を形成する傾向があり、ポリドーパミンコーティングされたナノダイヤモンドは良好な分散を形成した。より高いドーパミン濃度は、ナノダイヤモンド表面に厚いポリドーパミン層の形成をもたらした。コーティングされていないナノダイヤモンドの分散は、透明で無色であった。ナノダイヤモンドを5ナノメートルの厚いポリドーパミン層でコーティングすると、分散液は白濁して茶色に見えた。分散の外観は、より厚いポリドーパミンコーティングで徐々に暗くなりました。15ナノメートル厚のポリドーパミン層を被覆したナノダイヤモンドのジアミン銀の還元は、水酸化ジアミン濃度が1ミリリットル当たり0.4〜0.6ミリグラムであったときに最も成功した。より低い濃度で形成されたナノダイヤモンドは、より小さすぎて効果的に研究できませんでした。最大吸光度値は、1ミリリットル溶液当たり0.4~0.6ミリグラムから形成されたナノ粒子の直径はそれぞれ約20ナノメートルと30ナノメートルであることを示した。TEMは、1ミリリットルのダイアミン銀溶液当たり0.4ミリグラムから生成された銀ナノ粒子は幅約24ナノメートルであり、1ミリリットル溶液当たり0.6ミリグラムから生成されたナノ粒子は幅約28ナノメートルであった。ナノダイヤモンド表面上のナノ粒子の数も、より高いダイアミン銀濃度で大きかった。この手順を使用した後、制御可能なPDA厚さを有する十分に分散したナノダイヤモンドが形成された。この技術は、研究者が触媒、バイオセンサー、ナノキャリアのナノダイヤモンドアプリケーションを探求する道を開きます。PDA支援型の物質化プロセスは、追加の還元剤を使用することなく、金属前駆体の還元時に銀ナノ粒子の形成を誘導し、PDA被覆表面に固定化することができます。さらに、PDA層には、連続およびアミン修飾生体分子を共役させるためにさらに利用できるオープン官能基が含まれています。
