preparation of acute spinal cord slices for whole-cell patch-clamp recording in substantia gelatinosa neurons

Recente studies van de gehele-cel patch-clamp van neuronen van de substantia gelatinosa (SG) hebben een grote hoeveelheid informatie over de spinale mechanismen die ten grondslag liggen aan zintuiglijke transmissie, Nociceptieve regulering en chronische pijn of jeuk ontwikkeling gegeven. Implementaties van elektrofysiologische opnamen samen met morfologische studies op basis van het nut van acute ruggenmerg segmenten hebben verder verbeterd ons begrip van neuronale eigenschappen en de samenstelling van lokale circuits in SG. Hier presenteren we een gedetailleerde en praktische gids voor de bereiding van segmenten van het ruggenmerg en Toon representatieve geheel-cel opnemen en morfologische resultaten. Dit protocol staat ideale neuronale behoud en in vivo voorwaarden tot op zekere hoogte kan nabootsen. Kortom, de mogelijkheid om het verkrijgen van een in vitro -voorbereiding van de segmenten van het ruggenmerg stabiele stroom en spanning-kogelklem opnames maakt en dus grondig onderzoek naar de eigenschappen van het intrinsieke membraan, lokale circuits kunnen vergemakkelijken en neuronale structuur met behulp van uiteenlopende experimentele benaderingen.

Acute ruggenmerg segmenten opgesteld volgens het schema in Figuur 1afgebeeld. Na snijden en herstel, werd een segment van het ruggenmerg overgeplaatst naar de kamer van de opname. Preparation of Acute Spinal Cord Slices for Whole-cell Patch-clamp Recording in Substantia Gelatinosa Neurons

Voorbereiding van Acute ruggenmerg segmenten geheel-cel Patch-clamp opname in neuronen van de Substantia Gelatinosa

Hier beschrijven we de essentiële stappen voor geheel-cel patch-clamp opnamen gemaakt van substantia gelatinosa (SG) neuronen in het ruggenmerg in vitro segment. Met deze methode kunt de eigenschappen van het intrinsieke membraan, synaptische transmissie en morfologische karakterisering van SG neuronen worden bestudeerd.

Deze methode kan helpen verduidelijken van de spinale mechanismen die ten grondslag liggen aan sensorische transmissie, nociceptieve regelgeving en chronische pijn of pijn ontwikkeling Het belangrijkste voordeel van deze techniek is dat het kan toestaan dat ideale neuronale bewaring en het kan nabootsen in vivo voorwaarden tot op zekere hoogte. Begin met het opnemen van het opnemen van elektroden van borosilicaat glazen microcapillaries met behulp van een haarvatentrekker. Vul vervolgens een mal met gesmolten agar om een 1,2 centimeter bij 1,5 centimeter bij 2,0 centimeter agar blok voor te bereiden.Knip het blok bij om een centraal kanaal te hebben als u dwarse secties plant. Snijd vervolgens de omliggende weefsels weg en isoleer snel het lumbosacrale segment van de wervelkolom.Breng het lumbosacrale segment over op een glazen schaal met ijskoude sucrosegebaseerde ACSF. Met de ventrale kant naar boven, gebruik fijne schaar te snijden door de wervelpeddicle bilateraal en bloot het ruggenmerg zorgvuldig. Isoleer een twee centimeter lang deel van het ruggenmerg met de lumbosacrale vergroting en breng de spinale sectie in een andere glazen schotel gevuld met koude sacharose ACSF.Werk onder een dissectiemicroscoop om de hersenvliezen en het pia-arachnoïdale membraan te verwijderen. Snijd alle ventrale en rugwortels zo snel mogelijk weg. Zorg ervoor dat het letsel aan het ruggenmerg, vooral de rughoorn bij het verwijderen van de hersenvliezen en de ruggenwortels, vermijdt.Plaats het ruggenmerg op het eerder bijgesneden agarblok. Om dwarse plakjes voor te bereiden, bevestigt u de ventrale kant van het ruggenmerg aan de agar met de rugzijde naar het blad. Om parasagittale plakjes te bereiden, bevestigt u de ventrale kant met superlijm in een verticale richting aan de agar.Monteer vervolgens het agarblok op een platform van een vibratome met superlijm. En bereid 300 tot 500 micron dwars- of parasagittale plakjes voor met een geavanceerde snelheid van 0,025 millimeter per seconde en een trillingsfrequentie van 80 Hertz. Het ruggenmerg moet dorso-ventrally worden gesneden.Voor de beste resultaten moet het segment binnen 15 tot 20 minuten worden bereid. De dikte van het ruggenmergsegment mag niet meer dan 600 micron zijn om de zichtbaarheid van de cel te bevredigen. Gebruik een plastic bijgesneden pipet om de plakjes over te brengen op nylon gaas in een opslagkamer met continu zuurstofrijk ACSF op 32 graden Celsius.Voor het uitvoeren van hele-cel patch klem opnames van substantia gelatinosa of SG neuronen, gebruik kaliumion gebaseerde intracellulaire oplossingen voor de opname, terwijl de toepassing van cesium gebaseerde oplossing alleen voor de registratie van remmende post-synaptische stromen. Verplaats het dwarslaesiedeel voorzichtig naar de opnamekamer. En stabiliseer het dan stevig met een U-vormige platina draad met nylon draden voor optimale slice stabiliteit.Maak het segment gestaag over te maken met bubbled ACSF bij kamertemperatuur en stel de overfusiesnelheid in op twee tot vier milliliter per minuut om voldoende oxygenatie te bereiken. Dan, met behulp van een lage resolutie microscoop lens, identificeren van het gebied van SG neuronen als een doorschijnende band. Kies een gezond neuron, gekenmerkt door een driedimensionale vorm met een helder en glad membraan als doelcel met behulp van de doelstelling met hoge resolutie en pas het aan het midden van het scherm van de videomonitor aan.Vul een micropipet met een passend volume van kaliumionen op basis van of cesium ion gebaseerde intracellulaire oplossing. Steek vervolgens de micropipet in de elektrodehouder en zorg ervoor dat de intracellulaire oplossing contact maakt met de zilverdraad in de houder. Breng de micropipet scherp en gebruik de micromanipulator om deze onder te dompelen in de ACSF.Breng een milde positieve druk aan om vuil en vuil weg te dringen. Beweeg de micropipet geleidelijk naar het beoogde neuron. Zodra de pipet nadert het neuron en een zeer klein kuiltje vormen op het neuronale membraan, laat de positieve druk om een gigaseal te vormen.Verander het bedrijf potentieel tot min 70 millivolt, die dicht bij de fysiologische rust membraan potentieel van een cel. Breng vervolgens een tijdelijke en zachte zuigkracht aan op de micropipet om het membraan te scheuren en een goede hele celconfiguratie te creëren. Record bakken eigenschappen door het testen van het afvuren patroon van elk neuron in de huidige klem met een reeks van een seconde depolariseren huidige pulsen van 25 tot 150 picoamps met 25 picoamp stappen bij rust membraan potentieel.Om somatische subthreshold stromen te registreren, houd het membraanpotentieel op min 50 millivolt in spanningsklemwijze en breng u vervolgens een reeks hyperpolaire spanningspulsen van een seconde van min 60 millivolt tot min 120 millivolt aan met een afname van 10 millivolt. Neem excitatory post-synaptische stromen op met de op kaliumion gebaseerde intracellulaire oplossing in spanningsklemmodus bij een houdpotentieel van min 70 millivolts. Leg IPSC's vast met behulp van caesium ionen gebaseerde intracellulaire oplossing voor het opnemen van IPSC's.Na het houden van het membraanpotentieel op min 70 millivolt gedurende ongeveer vijf minuten, geleidelijk veranderen van het bedrijf potentieel tot nul millivolt. Wacht een paar minuten op stabilisatie en begin vervolgens met het opnemen van de IPSC-gebeurtenissen. Het afvuren van patronen bepaald door het injecteren van een reeks van een seconde depolariseren de huidige pulsen in een SG neuron bij rust membraan potentieel kan worden geclassificeerd als tonic-vuren, vertraagd-vuren, gap-firing, initial-burst, phasic-bursting, single-spike en terughoudend-vuren.Dit beeld toont het roepende protocol voor subthreshold stromen in voltageklem. Deze representatieve sporen tonen de reactie op hyperpolariserende huidige injectie geclassificeerd als IH, IA en IT. Dit is een representatief spoor van SEPSC's geregistreerd van SG neuronen op min 70 millivolt in de afwezigheid en aanwezigheid van 50 micromolar APV en 20 micromolar CNQX. Dit spoor toont de geregistreerde SEPC's vóór de toevoeging van APV en CNQX op een uitgebreide tijdschaal.Na deze procedure zijn de methoden zoals gepaarde patch klem opnames of optogenetica kan worden uitgevoerd om aanvullende vragen te beantwoorden, zoals het karakteriseren van specifieke neuronale microcircuits in substantia gelatinosa.
