Metoden præsenteres muliggør vellykket isolation af glomeruli at undersøge glomerulær RNA og protein. Derudover kan primære glomerulær cellekulturer udføres fra de isolerede glomeruli. Proteinuri er kendetegnende for glomerulær skade og normalt ledsager afbrydelse af glomerulær filter1. Filteret glomerulær er sammensat af fenestrated endothelium, glomerulær basalmembran og podocytes. Filteret glomerulær delikat molekylære struktur er meget dynamisk og underlagt celle overflade protein menneskehandel både raske og syge nyrer2,3,4,5,6 . Endocytose af celle overflade proteiner har vist sig at være afgørende for overlevelse af podocytes7. Nephrin og podocalyxin er transmembrane proteiner udtrykt på podocytes. Nephrin er rygraden i glomerulær slids mellemgulvet, mens podocalyxin er en sialoglycoprotein belægning sekundære mund processer i podocytes8,9,10. Endocytic handel har tidligere vist for nephrin og podocalyxin3,11,12,13,14.
Til bedste af vores viden, har endocytose af celle overflade proteiner ikke endnu blevet beskrevet i glomerulær endotelceller i litteraturen. Imidlertid udtrykke endotelceller generelt alle nødvendige proteiner for de forskellige typer af endocytose (dvs. clathrin-afhængige, tømmerflåde-afhængige endocytose)15,16. Derfor, endotel celle overflade handel kan studeres med denne metode bruger, for eksempel vaskulære endotel (VE)-cadherin og intracellulære vedhæftning molekyle (ICAM-2) som en celle overflade markør protein for glomerulær endotelceller17 .
Desværre, der er ingen nøjagtige in vitro- model for de sarte tre-lags glomerulær filter i hvilken celle overflade protein handel kan studeres. Målet med denne metode er således at studere glomerulær protein menneskehandel i vivo. Denne protokol indeholder desuden oplysninger om, hvordan at isolere glomeruli, muliggør yderligere analyse af glomerulær RNA, proteiner eller celler. Lignende glomerulær isolation teknikker har været beskrevet af forskellige grupper18,19.
Vi og andre har tidligere brugt ex vivo mærkning af glomerulær celle overflade proteiner af biotinylation2,3,4,20,21. Men i denne ex vivo metode, isolerede glomeruli blev udsat for mekanisk stress, som kan påvirke endocytic handel. Alternativt, immunofluorescens mærkning af glomerulær celle overflade proteiner har flittigt blevet brugt i litteratur2,20,22. Med denne metode, men kun et lille antal proteiner kan analyseres inden for ét dias, og kvantitering af immunofluorescens billeder er ofte vanskelig.
Denne roman i vivo metode giver et pålideligt redskab til at studere glomerulær celle overflade protein overflod og menneskehandel præcist i raske og syge nyrer, og det kan bruges som et supplement til immunofluorescens test.

isolation of glomeruli and in vivo labeling of glomerular cell surface proteins

Proteinuri skyldes forstyrrelse af det glomerulær filter, der er sammensat af fenestrated endothelium, glomerulær basalmembran og podocytes med deres slids membraner. Endotelprotein vaskulær endotel cadherin og intracellulær vedhæftning molekyle to er også immunoprecipiteret inden for biotin fraktion.I den tidlige fase af nefrotoksisk nefritis observeres et reduceret ekspression af celleoverfladen nefrorin hos nefrotoksiske nefritiser sammenlignet med kontrol. Densitometiske analyser viser faktisk en signifikant reduktion af biotin laded nephrin hos nefrotoksiske nefritiser sammenlignet med kontrollører. Kvantitativ analyse af det samlede nephrin til actin-forhold afslører ingen signifikante forskelle mellem kontrol- og nefrotoksiske nefritis mus.Desuden observeres der lige store mængder podocytter hos nefrotoksisk nefritis og kontroldyr. I slutningen af nefrotoksisk nefritis udviser nefrotoksiske nefrotiske dyr en genfinding af nefromanudtryk uden signifikante forskelle i celleoverfladens nephrin målt mellem kontrol og nefrotoksiske nefrotgiftmus og en samlet total nephrinreduktion i nefrotoxiske nefrotiske dyr.
