Topdown proteomics (TDP) streeft naar de grootschalige karakterisatie van proteoforms binnen een Proteoom. TDP hangt af van de effectieve scheiding van vloeistof-fase van intacte eiwitten vóór electrospray ionisatie-tandem massaspectrometrie (ESI-MS/MS) analyse als gevolg van de hoge complexiteit en dynamisch bereik van de hoge concentratie van de Proteoom1,2 ,3,4,5. Capillaire zone elektroforese (CZE) is een krachtige techniek voor de scheiding van biomoleculen, op basis van hun grootte-naar-charge ratio's6. CZE is relatief eenvoudig, waarbij alleen een open tubular-gesmolten siliciumdioxide capillaire, een achtergrond elektrolyt (BGE), en een voeding. Een steekproef van intacte eiwitten in het capillair met behulp van druk of spanning kan worden geladen en scheiding wordt geïnitieerd door de beide uiteinden van het capillair in de BGE onder te dompelen en het toepassen van een hoogspanning. CZE-MS vereist een zeer robuuste en gevoelige interface aan paar CZE aan MS. large-scale top-down proteomics using capillary zone electrophoresis tandem mass spectrometry

Capillaire zone elektroforese-electrospray ionisatie-tandem massaspectrometrie (CZE-ESI-MS/MS) is erkend als een nuttig hulpmiddel voor proteomics van de top-down dat gericht is op het karakteriseren van proteoforms in complexe proteomes. Laat alle andere parameters op hun standaardwaarden staan en klik op de startknop rechtsonder in de grafische gebruikersinterface. Het representatieve elektroferogram voor het standaard eiwitmengsel wordt getoond.Het standaard eiwitmengsel wordt meestal ten minste in tweevoud uitgevoerd om de scheidingsefficiëntie en de reproduceerbaarheid van het systeem te evalueren. De reproduceerbaarheid kan worden geëvalueerd door de relatieve standaarddeviaties van eiwitintensiteit en migratietijd.Het CZE MS/MS platform kan worden gebruikt voor grootschalige karakterisering van proteoforms in verschillende complexe proteomen, waarbij meer dan 500 proteoforms en 190 eiwitten uit een E.coli proteoom in één run met veel vertrouwen worden geïdentificeerd. Hier kan een ingezoomd oog op het elektropherogram worden gebruikt om het scheidingsvenster van het systeem te beoordelen. De zeer lage E-waarde en spectrale FDR suggereert het hoge vertrouwen van de proteoform ID.The hoge aantal gematchte fragmentionen geeft verder het hoge vertrouwen van de ID.While een poging tot deze procedure, is het belangrijk om te onthouden om de geëtste scheiding capillaire draad in de elektrospray zender langzaam en voorzichtig.Stabiele isotopenetikettering of labelvrije methoden kunnen worden opgenomen in de CZE MS-procedure om te bepalen hoe proteoform overvloed verandert in cellen over verschillende omstandigheden. Na de ontwikkeling maakte deze techniek de weg vrij voor onderzoekers op het gebied van top-down proteomics om de rol van proteovormen te onderzoeken bij het reguleren van verschillende biologische processen in cellen. Vergeet niet dat het werken met fluorzuur zeer gevaarlijk kan zijn en voorzorgsmaatregelen zoals goede persoonlijke beschermingsmiddelen en het hebben van noodprocedures moeten altijd worden genomen bij het uitvoeren van deze procedure.
