בסרטון זה, אנו מציגים כיצד להשתמש צילינדר ומבחן שדה פתוח כדי לצמצם את תפקוד המנוע במודל MPTP של מחלת פרקינסון. היתרון העיקרי הוא שהעכברים לא צריכים להיות מאומנים לפני הבדיקה ההתנהגותית. בנוסף, שיטה זו יעילה ועלות הציוד נמוכה.אנשים חדשים במבחנים התנהגותיים עשויים להמעיט בערך ההשפעה של התערבות סביבתית. שמירה על שקט והימנעות משינויים בטנה יעזרו. כאן, בדיקות התנהגותיות מתבצעות 24 שעות לאחר הזריקה הסופית של משטר של שבעה ימים של MPTP או זריקות מלוחים ו -40 דקות לאחר ניהול תוך-גזי של בסדר גמור לתת קבוצה של בעלי החיים.אם בדיקות התנהגותיות אינן מבוצעות באותו חדר שבו שוכנים בעלי החיים, התאקלם בעכברים לחדר החדש במשך 30 עד 60 דקות לפני הבדיקה. התחל בהנחת גליל זכוכית שקוף במרכז השולחן. הקיפו את הצילינדר משלושה צדדים עם קרטון שחור, והניחו ארבעה עד שמונה ס"מ הרחק מהגליל כדי להפחית את השפעת התאורה הסביבתית.השאירו צד אחד של הצילינדר הפונה למצלמה להקלטת וידאו. חבר מצלמה המסוגלת להיות ברזולוציית פיקסלים של לפחות מיליון 40 עד 60 ס"מ מהגליל כדי להבטיח שכל הצילינדר יהיה כלול בשדה הצפייה. מניחים עכבר אחד לתוך הצילינדר בכל פעם ולהתחיל לצלם בבת אחת.במהלך תהליך זה נסו להימנע מרעש או משינויי אור כדי למנוע השפעה על התנהגות העכבר. מעליות כפות רגליים מתרחשות כאשר העכבר מתרומם על גפיו האחוריות, מרים הן את הרגליים הקדמיות מעל פני הכתף והן את האדמות. הפסיקו לצלם אחרי שלוש דקות והחזרו את העכבר לכלוב הביתי.נקה את הצילינדר במים ולאחר מכן רסס 70% אתנול על הקיר הפנימי כדי לחטא אותו ולהסיר ריחות עכבר. יש לנגב את הצילינדר יבש לפני בדיקת העכבר הבא. לאחר השלמת הבדיקה, הפעל את הווידאו בקצב של פי 0.5 מהמהירות הרגילה וספיר את מספר הרמות הכפות על הקיר של כל עכבר.אם העכבר מרים את צלעותיו מעל רמת הכתף מספר פעמים ברציפות ללא נחיתה, יש לספור אותו רק פעם אחת. בצע בדיקות שדה פתוח במהלך אותה הפעלה כמו בדיקת הצילינדר. הכן תיבת תגובת שדה פתוחה שקופה עם צלחת עץ, מכוסה במטלית שחורה או לבנה מנוגדת בהתאם לצבע העכבר.תקן מצלמה מעל השדה בגובה של מטר אחד. כוונן את המצלמה כדי לוודא שתיבת התגובה של השדה הפתוח נמצאת ממש במרכז הווידאו. שים עכבר בדיקה אחד לתוך התיבה ולתת לעכבר להכיר את הסביבה במשך כדקה אחת.הקלט וידאו של חמש דקות באמצעות המצלמה המחוברת למחשב. לבסוף, לנקות את כל הצואה בתיבת תגובת השדה הפתוח, לרסס 70% אתנול על הקופסה, ולנגב אותו. במבחן הצילינדר נצפתה ירידה במספר העורפים האחוריים כנגד הקיר בעכברים שטופלו ב-MPTP בהשוואה לעכברים שטופלו בתמיסת מלח.אחוריים מוגברת נצפו בעכברים שטופלו MTPT נתון L-דופמין ביום הבדיקה לעומת עכברים שטופלו MPTP מקבל מלוחים. תמונה זו מציגה את מעקב התנועה הייצוגי ואת התפלגות הזמן הסטטי, זמן ההליכה ותזמן הריצה בעכברי בקרה שלא טופלו באמצעות MPTP. עכברים שטופלו ב- MTPT הדגימו מהירות תנועה נמוכה יותר, מרחק תנועה קצר יותר, זמן סטטי ארוך יותר וזמן ריצה קצר יותר מאשר עכברים מטופלים מלוחים, המדגימים גירעון מוטורי המושרה על ידי MTPT.עכברים שטופלו ב- MPTP שקיבלו L-דופמין הראו מהירות תנועה גבוהה יותר, מרחק תנועה ארוך יותר, זמן סטטי קצר יותר וזמן ריצה ארוך יותר מהעכברים שטופלו ב- MPTP בלבד, מה שמצביע על כך ש- L-דופמין מפחית את הגירעון המוטורי המושרה על-ידי MPTP. בעת שינוי העכברים נבדקים לנקות את האזור לחלוטין לרסס אתנול כדי לחסל את ההשפעה של הריח של העכברים לשעבר. בעקבות הליך זה, אמצעים אחרים כגון מבחן הליכה קרן, מבחן מוט יכול להתבצע.נתונים הקשורים ליכולת איזון ויכולת מוטורית ניתן להשיג באמצעות שיטות אלה.
