这是第一篇解决从细胞外囊泡中产生的小RNA测序技术问题的论文。NGS 样品需要严格的制备和质量控制。由于EV的性质，EV样品的QC过程偏离了标准。该协议的优点是其中为首次使用的用户执行的 QC 步骤。演示程序将是苏俊义，一个研究助理，从我的实验室。首先，在5个T175烧瓶中，以每平方厘米2000个细胞的填充人类中质干细胞。然后在5%的二氧化碳环境中在37摄氏度下生长细胞，每两到三天更换一次介质，直到获得5个90%汇合细胞的烧瓶。接下来，用每瓶20毫升PBS清洗细胞三次。在每个烧瓶中加入 20 毫升 EV 收集介质。并在5%的二氧化碳环境中孵育37摄氏度48小时。收集介质，在300克和4摄氏度下离心10分钟。然后收集上一代，在2000克和4摄氏度下将介质离心20分钟。再次收集上一代，在 15，500 g 和 4 摄氏度下将介质离心 30 分钟。最后一次收集上一个。接下来，将介质传输到超离心管。此处使用 60Ti 固定角转子，颗粒将固定在管的一侧。标记需要颗粒的盖子侧面，并在需要颗粒的管侧画一个圆圈。在10万克和4摄氏度下，将exRNA分粒90分钟。最后离心后，取出上经。使用小块吸水纸干燥管内，而不接触管的底部。然后在吸水纸上反转管子。要重新暂停颗粒，请在每个管中加入200微升PBS，然后旋转管30秒。移液上下 20 次。然后用纳米粒子跟踪分析评估细胞外囊泡和其他生物分子。将颗粒储存在零下80摄氏度，直到进一步实验。解冻样品后，使用RNA隔离试剂盒根据制造商的协议提取RNA。在 100 微升无 RNAse 水中从 RNA 隔离试剂盒中提供的柱中分离出 RNA。要通过乙醇沉淀浓缩RNA，加入1微升糖原、10微升双摩尔pH-5.5醋酸钠和250微升预冷却99%乙醇放入100微升纯化RNA中。然后在零下20摄氏度下孵育样品过夜，沉淀RNA。为了对RNA进行颗粒，在16，000克和4摄氏度下离心20分钟。接下来，去除上一代，用一毫升75%乙醇清洗RNA颗粒。再次在16，000克和4摄氏度的温度下离心5分钟，以对RNA进行颗粒。取出乙醇，使RNA管盖打开5至10分钟，对RNA颗粒进行空气干燥。将RNA颗粒重新浓缩在7微升无RNA水中，并使用基于芯片的毛细管电泳机检测RNA质量和浓度。为了开始库建设，移液器将RNA的5微升放入预冷却的0.2毫升PCR管中。接下来，以1比10的体积比在无RNAAse水中稀释3'适配器。将稀释的适配器的0.5微升加入RNA管中，并上下移液8次。短暂离心，收集管底的所有液体。在70摄氏度下孵育RNA适配器混合物，在预热热循环器中孵育两分钟。然后将样品放回冷藏块上。接下来，将一微升的结扎缓冲液、0.5微升的RNAse抑制剂和0.5微升的T4RNA连接糖加入RNA适配器混合物。移液上下八次，并短暂离心。然后在28摄氏度下在预热热循环器中孵育管一小时。接下来，将 0.5 微升的止损溶液添加到样品管中，使管留在热循环器中。移液上下8次，并在28摄氏度下继续孵育15分钟。在无 RNAse 水中以 1 比 10 的体积比稀释 5'适配器。将稀释的适配器的 0.5 微升添加到单独的 0.2 毫升 PCR 管中。在 70 摄氏度下加热 5'适配器两分钟。然后将样品放在冰冷块上。将 0.5 微升 10 纳米摩尔 ATP 和 0.5 微升 T4 RNA 连接酶添加到 5' 适配器管中。移液上下八次。并短暂离心收集所有液体到底部。接下来，将 5'适配器混合物的 1.5 微升添加到 RNA 样品中。移液器非常轻柔八次，并在28摄氏度下孵育样品一小时。为了获得cDNA，以1比10的体积比稀释无RNAAse水中的逆转录酶底转。将稀释的底流板的0.5微升加入RNA样品中，轻轻移液器8倍，并短暂离心。然后在70摄氏度下孵育RNA和底土混合物，在预热热循环器中孵育两分钟。然后将样品放在冰冷块上。接下来，在样品管中加入一个5X第一链缓冲液、0.5微升12.5毫摩尔DSP混合物、0.5微升100毫摩尔DTT、0.5微升RNAse抑制剂和0.5微升逆转录酶。移液八次非常轻轻地。和离心机简要。在预热热循环器中，在 50 摄氏度下孵育样品一小时。接下来，在cDNA样品中加入4.25微升的超纯水、12.5微升的PCR混合物、1微升的指数底向器和1个万能的微升。移液上下八次，并短暂离心。最后，将样品放在热循环器中，然后根据手稿设置循环器。要净化RNA库，请使用自动DNA大小分馏器对140至160个碱基对之间的波段进行测序。在基于列的 PCR 净化套件上运行提取的库，并在 10 微升无 RNAse 水中对库进行解处理。要检查库的大小，请将净化库的一微升加载到 DNA 芯片上，然后按照制造商的协议进行。为了量化库浓度，首先在10毫摩尔pH-8.0 Tris-Hcl中稀释纯化库的一微升，以0.05%聚索酸酯20，体积比为1比1，000。然后使用商业库定量套件的 DNA 标准运行 QPCR。最后，根据测序机的要求，根据高通量测序系统进行序列，池中等量的库。细胞外RNA的电谱图包括广泛的RNA。相比之下，细胞RNA表现出非常明显的峰值，对应于5StRNA、5.8SrRNA、18SrRNA和28SrRNA。由此产生的两个图书馆的质量是高和可比的。两个库的长度分布显示，在22个核苷酸时，有一个峰值，与微RNA相关。相比之下，来自细胞外RNA的库更异构，并包含两个与tRNA、一半和片段或piRNA相关的峰值。65%的细胞RNA读取被映射到人类微RNA，其他每个小RNA在映射的读取中只占一小部分。相比之下，只有8%的细胞外RNA与人类微RNA相匹配。读取映射到的最丰富的小RNA是tRNA，不匹配的读取后来与细菌序列匹配。与牛基因组的比较表明，FBS不是污染源。因此，与正常细胞RNA相比，细胞外RNA表现出非典型特征。库准备是该协议的关键部分。必须非常轻柔地进行混合，以避免气泡形成。此方法还可用于研究来自其他类型的细胞的 exRNA 的配置文件，这有助于研究 exRNA 的功能。
