Generellt individer som är nya för denna metod kommer att kämpa eftersom hälla lutningarna och lägga till cellerna till lutningarna kan vara ganska svårt. Visuell demonstration av denna metod är kritisk, eftersom vi visar de tekniker som är svåra att bemästra, vilket ger förslag och tips för att hjälpa användaren. Till att börja, välj en enda koloni från en lagerplatta med en steril slinga, och inokulera 10 milliliter av en lämplig buljong.
Inkubera kulturen över en natt. Överför sedan kulturen till ett 15 milliliterrör och centrifug. Därefter kasta supernatant, och åter upphäva pelleten i två milliliter PBS.
Upprepa centrifugeringen, och kassera den resulterande supernatanten. Sedan åter avbryta pelleten i två milliliter PBS. För att skapa densiteten gradient utspädningar, kombinera densitet gradient medium med PBS.
Sedan alikvot 600 mikroliter av varje gradient utspädning i enskilda två milliliter rör. Ta upp 100 mikroliter celler med en 200 mikroliterpipett, och placera pipettspetsen på sidan av röret strax under menisken i en av de många lutningarna. Aspirera bakteriecellerna på lutningen mycket långsamt, var noga med att inte blanda gränssnittet.
Centrifugera de preparerade rören i 10 minuter vid 8, 000 g.a. Efter detta överför rören till ett rack för att visualisera den minsta gradientsutspädning som krävs för att behålla cellerna strax ovanför övertoningsskiktet. Överför först en milliliter av den mest utspädda densitetsgradientens utspädning till ett rundbottenrör på fem milliliter.
Med hjälp av en spruta med en 1,5 tums nål, ta upp en milliliter av den näst mest koncentrerade densitetsgradienten. Placera nålen längst ner på röret och injicera innehållet i sprutan mycket långsamt. Det måste finnas ett tydligt gränssnitt vid varje steg i densitetsgradienten.
Om lagren blanda, kommer du inte uppnå ren separation av dina bakterier. Ta ut nålen ur övertoningen försiktigt för att inte störa gränssnittet mellan gradientens utspädningar. Placera sedan röret i ett rack.
Tillsätt försiktigt 600 mikroliter av de preparerade cellerna till överst på gradienten i röret. Placera sedan röret i en röradapter, och väg röret med adaptern för att säkerställa balans. Efter detta, placera röret adaptern i en fast vinkel rotor inom en bänk-top centrifug, och centrifug röret i 30 minuter vid 3, 000 g's.
Slutligen, efter centrifugeringen, ta bort röret försiktigt, placera den i ett rack, och fotografera resultaten. I detta protokoll användes densitetsgradientcentrifugeringen för att separera bakteriestammar baserat på hur mycket kapsel de producerar. Även om de exakta resultaten beror på bakteriearter, de flesta stammar kommer att migrera till en enda plats inom lutningen.
Tillämpning av denna metod på en bakteriell mutant bibliotek bör producera ett större band ovanför gradienten, ett mindre tätt band genom det översta lagret av övertoning, och en mindre acapsular fraktion i botten. Medan du försöker med den här proceduren är det viktigt att komma ihåg att gradientgränssnitten inte ska blandas, och övertoningen bör inte innehålla luftbubblor. Efter ingreppet bör valideringsmetoder som uronic acid assay eller mikroskopi utföras för att undersöka kapselmängder.
Detta är särskilt viktigt när du använder denna teknik för nya stammar. Glöm inte att det kan vara farligt att arbeta med Farogrupp 2-patogener.
