We beschrijven een eenvoudige methode om te onderzoeken hoe de aanvulling component C1q, interactie met hyaluronzuur, in staat is om het gedrag van de primaire cellen geïsoleerd van menselijke tumor weefsels moduleren. Zowel HA als C1q zijn overvloedig aanwezig in het weefsel micromilieu, zowel onder fysiologische en pathologische omstandigheden, die deelnemen aan verschillende cellen biologische processen. Bijvoorbeeld, C1q is aangetoond aanwezig te zijn in de microomgeving van de placenta waar het gunsten extravillous...

De tumor microenvironment (TME) beïnvloedt de ontwikkeling van kanker en progressie, omdat het kan een tolerante niche voor cel overleving, groei en invasie te bieden. De identificatie van nieuwe belangrijke spelers in TME kan nuttig zijn voor de ontdekking van nieuwe moleculaire hulpmiddelen voor doel therapie. TME omvat een complex en dynamisch netwerk van niet-kwaadaardige cellen, zoals endothelial cellen, fibroblasten en cellen van het immuunsysteem, ingebed in de omliggende extracellulaire matrix (ECM) componenten, waaronder collageen, laminins, fibronectins, proteoglycanen en hyaluronans. Zowel de tumor en niet-tumorcellen synthetiseren en scheiden ECM componenten samen met cytokines, chemokines, groeifactoren en inflammatoire en matrix remodeling enzymen die de algehele verandering van de fysieke, chemische en signalering eigenschappen van TME. Onder deze bestanddelen, hyaluronzuur (HA) is ontstaan om een cruciale rol in de Tumorbiologie uit te oefenen. Ondanks de eenvoudige chemische samenstelling, HA, samen met zijn HA-bindende moleculen (hyaladherins), kan moduleren bloedvat, immuunsysteem responsiviteit en ECM remodeling in een grootte en concentratieafhankelijke wijze1.
Het complement (C) systeem maakt ook deel uit van de lokale TME, die recentelijk steeds meer aandacht heeft gekregen. Het C-systeem omvat een set van oplosbare en membraan-gebonden eiwitten die betrokken zijn bij de eerste verdedigingslinie tegen niet-zelf-cellen, ongewenste host-elementen en pathogenen. Functioneel, verbindt C de twee-effect wapens van ingeboren en aanpassingssystemen om of directe cel het doden of montage van een ontstekingsreactie2te bevorderen. C de activering kan de tumorgroei onderdrukken, door kankercellen te vernietigen of hun uitgroei te remmen, maar het is steeds duidelijker geworden dat het een tumor-bevorderende activiteit kan bezitten door chronische ontsteking te ondersteunen, bevorderend de totstandbrenging van een immunosuppressieve milieu, inducerende bloedvat, en het activeren van kanker-gerelateerde signalering trajecten3. In deze context, C1q, de eerste erkenning molecuul van de klassieke route van het C-systeem is ontstaan om belangrijke functies uit te oefenen in de tumor microomgeving onafhankelijk van C activering4. C1q is aangetoond dat lokaal worden uitgedrukt door een reeks van menselijke kwaadaardige tumoren, waar het kan voorstander van kanker cel adhesie, migratie en proliferatie in aanvulling op bloedvat en metastasen5. Interessant C1q wisselwerking met een belangrijk bestanddeel van ECM zoals HA.
We ontwikkelden een techniek om de primaire kankercellen te isoleren van de tumormassa. Verder creëerden we de matrix, die kan stimuleren tumor microenvironment, met name de interactie tussen C1q en hoge moleculair gewicht hyaluronzuur. C1q gebonden aan HA was in staat om de adhesie van de tumorcellen te induceren.

Het is meer en meer aangetoond dat de tumor microenvironment een actieve rol speelt in neoplasie groei en metastase. Door verschillende wegen, kunnen de tumorcellen stromale, immune en endothelial cellen efficiënt aanwerven door stimulerend factoren, chemokines en cytokines af te scheiden. Op zijn beurt kunnen deze cellen veranderen de signalering eigenschappen van de micromilieu door het vrijgeven van groei-bevorderende signalen, metabolieten en extracellulaire matrix componenten om hoge proliferatie en gemetastaseerde competentie te ondersteunen. In deze context, identificeren we dat de aanvulling component C1q, sterk uitgedrukt lokaal door een reeks van menselijke kwaadaardige tumoren, op de interactie met de extracellulaire matrix hyaluronzuur, sterk van invloed op het gedrag van primaire cellen geïsoleerd van de menselijke tumor Specimens. De aanvulling component C1q is een pro-inflammatoire molecuul sterk uitgedrukt in het weefsel microomgeving die kunnen interageren met de extracellulaire matrix. Hier beschrijven we een methode om te testen hoe C1q gebonden aan hyaluronzuur effecten cel adhesie.

Een uitgebreide procedure om het samenspel tussen het complementeiwit C1q en hyaluronzuur te evalueren bij het bevorderen van celhechting. We hebben onlangs gedefinieerd dat de complementcomponent C1q een molecuul van het aangeboren immuunsysteem sterk tot uiting komt in de tumor en placentamicro-omgeving en in staat is om te interageren met de component van de extracellulaire matrix. In dit geval hyaluronzuur.Wij stellen een methode voor voor het bestuderen van de biologische functies van eiwitten in de hele ecronomische omgeving. Om het vermogen van deze moleculen te onderzoeken om de celdelingseigenschappen van extracellulaire matrixfactoren vorm te geven. Om te bestuderen hoe deze interactie kan worden waargenomen door de cellen die aanwezig zijn in het weefsel van de omgeving, hebben we deze eenvoudige methode opgezet waarbij binaire cellen afkomstig van menselijke weefsels c1q mochten naleven die gebonden zijn aan extracellulaire matrix, in dit geval hyaluronzuur.Voor coating procedure de eerste stap is de binding van een hoog moleculair gewicht hyaluronzuur tot 96 goed weefsel gecoate microplaat. Voor coating wordt hyaluronzuur verdund in 0,1 mol koolstofbuffer PH 9.6 bij een concentratie van 15 microgram per milliliter. En voorzichtig pipeting wordt gerequited om een homogene oplossing te bereiken.Een 100 microliter van de oplossing worden gebruikt om de putten te coaten. De plaat wordt vervolgens 's nachts bij vier graden Celsius overgebracht. De volgende dag vacuüm aspirate de behandelde putten.Was één keer met dPBS. Voeg C1q of BSA toe in de bijbehorende putten. C1q is tegenwoordig commercieel verkrijgbaar bij een concentratie van één milligram per milliliter en wordt verdund met een concentratie van 25 microgram per milliliter.En dPBS met 0,5% BSA en 0,7 millimol calcium en magnesium en divalent cations. BSA LPS gratis wordt gebruikt bij de concentratie van 0,5% en datum PBF. Plaat wordt vervolgens 's nachts bij vier graden Celsius overgebracht.C1q dosis afhankelijke interacties met geïmmobiliseerd hyaluronzuur wordt bepaald door enzym gekoppelde immuno SA. Cellen labelen met snelle DiI. De lipofiele tracer snel DiI wordt gebruikt om de cellen van belang label. Cellen die opnieuw worden opgetrokken in dPBS worden gebruikt bij de concentratie van een miljoen cellen per milliliter.Snelle DiI wordt verdund een tot 100 direct in de cel suspensie. De celsuspensie wordt handmatig gemengd en gedurende 15 minuten bij 37 graden Celsius overgebracht. Na vijf minuten incubatie, meng de cel suspensie en herhaal deze operatie twee keer meer.Om het overschot aan snelle DiI te verwijderen, voeg 10 milliliter dPBS toe. Draai zachtjes op en neer en centrifugeer zeven minuten lang op 250 g. Resuspend de celpellet in een milliliter menselijk endofiele serumvrij medium in aanwezigheid van 0,1%BSA.Cellen hechting op hyaluronzuur C1q matrices. Vacuüm zuigt de dPBS uit de gecoate putten. Verdeel 100 microliter van gelabelde celsuspensie naar de gecoate putten.200 microliters van gelabelde cellen worden uit elkaar gehaald in een ependure buis te gebruiken voor standaard curve generatie. Broed de plaat 35 minuten bij 37 graden Celsius. Haal de plaat uit de couveuse.De onevenwichtige cellen voorzichtig aanzuigen. Was één keer met dPBS met 0,5% BSA en 0,7 millimol calcium en magnesium. Lys de aanhangende cellen door het toevoegen van 200 microliters van lyse buffer.Bereid tegelijkertijd de standaardcurve voor op één tot twee seriële verdunning van de gelabelde cellen in lysebuffer. Voer de lezing van de florescence signaal op oneindige F 200 T-can instrument met behulp van 544 nanometer als excitatie golflengte en 590 nanometer als emissie golflengte. Representatieve resultaten.Zoals blijkt uit de representatieve resultaten, is het duidelijk dat celhechting op hyaluronzuur gevonden C1q als sterk verbeterd in vergelijking met HA alleen of HA gebonden BSA. Het percentage aanhangende cellen op elke matrix wordt berekend op basis van de standaardcurve. Conclusies. In onze experimenten hebben we allereerst het vermogen van de complementcomponent C12 geëvalueerd om de extracellulaire matrix in dit geval hyaluronzuur te binden met behulp van een arisaal metaal.Ten tweede analyseerden we het vermogen van menselijke primaire cellen bevlekt met fluorescerende kleurstof om gebonden hyaluronzuur na te streven. Het model dat we in deze studie hebben voorgesteld, is een eenvoudigere snellere en goedkopere methode in alternatief voor het 3D-model van extracellulaire matrixsteigers voor de evaluatie van deze cellulaire reactie op een combinatie van stimuli.
