De eerste doelstelling van deze studie was om te beoordelen van de haalbaarheid van hypoxische hoge intensiteit opleiding in muizen en de adequate karakteristieken van het protocol dat goed zou worden verdragen door muizen te kunnen bepalen. Met opzet, want er geen informatie met behulp van supramaximal (dat wil zeggen, meer dan Vmax) intensiteit training in muizen is, moesten we proeven op basis van eerdere protocollen ontwikkeld met atleten, die bestond uit vier tot vijf sets van vijf all-out sprints (ongeve...

In hypoxie leidt het verminderde geïnspireerd Fractie van zuurstof (O2) tot hypoxemia (verminderde arteriële druk in hypoxie) en een gewijzigde capaciteit O2 vervoer-1. Acute hypoxie induceert een verhoogde sympathische vasoconstrictor activiteit gericht op skeletspieren2 en een tegen 'compenserende' vasodilatatie.
Deze 'compenserende' vasodilatatie, ten opzichte van hetzelfde niveau van oefening onder normoxic omstandigheden,, is submaximal intensiteit in hypoxie, gevestigde3. Deze Vasodilatatie is essentieel om te zorgen voor een verhoogde doorbloeding en onderhoud (of beperken van de wijziging) van de zuurstoftoevoer naar de actieve spieren. Adenosine werd aangetoond dat niet een zelfstandige rol in dit antwoord, terwijl stikstofmonoxide (NO) de primaire endothelial bron lijkt aangezien belangrijke afstomping van de vergrote vasodilatatie werd gemeld met stikstofoxide synthase (NOS) remming tijdens hypoxische oefening4. Verscheidene andere vasoactieve stoffen spelen waarschijnlijk een rol in de compenserende vasodilatatie tijdens een hypoxische oefening.
Deze verbeterde hypoxische oefening hyperemia is evenredig aan de val van hypoxie-geïnduceerde in arteriële O2 inhoud en is groter naarmate de uitoefening intensiteit toeneemt, bijvoorbeeld tijdens intense incrementele oefening in hypoxie.
De NO-gemedieerde component van de compenserende vasodilatatie wordt geregeld via verschillende wegen met toenemende intensiteit van de oefening3: als β-adrenergic receptor-gestimuleerd geen component verschijnt voorop tijdens de lage-intensiteit hypoxische oefening , de bron geen bijdrage te leveren aan compenserende maagdilatatie lijkt minder afhankelijk is van β-adrenergic mechanismen zoals de trainingsintensiteit toeneemt. Er zijn andere kandidaten voor het stimuleren van geen vrijgave tijdens hogere-hypoxische intensiteitsoefening, zoals ATP verlost van erytrocyten en/of endotheel afkomstige prostaglandines.
Supramaximal oefening in hypoxie (genaamd herhaalde sprint opleiding in hypoxie [RSH] in de literatuur van de Fysiologie oefening) is een recente opleiding methode5 prestatieverbetering in team - of -racketsport spelers verstrekken. Deze methode verschilt van interval training in hypoxie uitgevoerd op of in de buurt van maximale snelheid6 (Vmax) Aangezien RSH uitgevoerd bij maximale intensiteit leidt tot een grotere spier perfusie en oxygenatie7 en specifieke spier transcriptionele Reacties8. Verschillende mechanismen zijn voorgesteld om uit te leggen van de effectiviteit van RSH: tijdens sprints in hypoxie, de compenserende vasodilatatie en de bijbehorende hogere doorbloeding baat de fast-twitch vezels, meer dan de slow-twitch vezels. RSH-efficiëntie is bijgevolg waarschijnlijk-type vezel selectieve en intensiteit afhankelijk. Wij speculeren dat het verbeterde reactievermogen van het vaatstelsel in RSH primordiaal is.
Oefening opleiding is uitgebreid bestudeerd in muizen, zowel in gezonde individuen pathologische muis modellen9,10. De meest voorkomende manier om te trainen van muizen een knaagdier loopband gebruikt, en de traditioneel gebruikte regime is lage-intensiteit opleiding, op 40%-60% van Vmax (bepaald met behulp van een incrementele loopband test11), voor 30-60 min12,13 ,14,15. Maximale intensiteit intervaltraining en het effect ervan op pathologieën zijn veel bestudeerd in muizen16,17; interval training van de lopende protocollen voor muizen zijn zo ontwikkeld. Die protocollen bestaan meestal uit ongeveer 10 aanvallen van op 80-100% van V-max waarop een knaagdier gemotoriseerde loopband, voor 1 – 4 min, afgewisseld met actieve of passieve rust16,18.
De belangstelling voor muizen met supramaximal intensiteit (bijvoorbeeld boven de Vmax) in hypoxie komt uit de resultaten van de vorige die de microvasculaire vasodilatory vergoeding en de intermitterende oefening prestaties zowel op meer toegenomen supramaximal dan bij maximale of matige intensiteiten. Om onze kennis is er echter geen eerdere verslag van een supramaximal opleiding protocol in muizen, in normoxia of in hypoxie.
Het eerste doel van de huidige studie was om te testen de haalbaarheid van supramaximal intensiteit training in muizen en de bepaling van een aanvaardbare en adequate protocol (intensiteit, duur van de sprint, herstel, enz.). Het tweede doel was om te beoordelen van de effecten van verschillende opleiding regime in normoxia en hypoxie op de vasculaire functie. Daarom testen we de hypothesen dat (1) muizen tolereren goed supramaximal oefening in hypoxie, en (2) dat dit protocol induceert een grotere verbetering in vasculaire functie dan oefening in normoxia, maar ook dan oefening in hypoxie bij lagere intensiteiten.

supramaximal intensity hypoxic exercise and vascular function assessment in mice

Oefening opleiding is een belangrijke strategie voor het behoud van de gezondheid en het voorkomen van veel chronische ziekten. Het is de eerste lijn van de behandeling aanbevolen door internationale richtlijnen voor patiënten met cardiovasculaire aandoeningen, meer in het bijzonder, extremiteit slagader ziekten onderste, waar de patiënten lopen capaciteit aanzienlijk is veranderd, waardoor hun kwaliteit van het leven.
Van oudsher, zowel lage continu oefening en intervaltraining hebben gebruikt. Onlangs, heeft ook supramaximal opleiding aangetoond dat atleten voorstellingen via vasculaire aanpassingen, onder andere mechanismen te verbeteren. De combinatie van dit soort opleiding met hypoxie kon brengen een aanvullende en/of synergetische effect, die zou kunnen van belang voor bepaalde pathologieën zijn. Hier beschrijven we hoe u supramaximal intensiteit trainingen in hypoxie op gezonde muizen op 150% van hun maximale snelheid, met behulp van een gemotoriseerde loopband en een hypoxische doos. Ook laten we zien hoe de muis ontleden om op te halen van de organen van belang, met name de longslagader en de abdominale aorta de iliacale slagader. Tot slot laten we zien hoe uitvoeren ex vivo vasculaire functie beoordeling op de opgehaalde vaartuigen, met behulp van isometrische spanning studies.

Om onze kennis is de huidige studie de eerste om te beschrijven van een programma van supramaximal intensiteit training in normoxia en hypoxie voor muizen. In dit protocol liep muizen vier sets van vijf 10 s sprints met een 20 s herstel tussen elke sprint. De sets werden afgewisseld met 5 min van herstel periodes. Ons protocol is belangrijk omdat het voor het eerst beschrijft hoe de Supramaximale Intensiteitstraining bij muizen uit te voeren, vooral bij hypoxie, en als het specifieke vasculaire endotheelaanpassingen veroorzaakt. Het belangrijkste voordeel van het gebruik van deze techniek met muizen is dat het trekt verder onderzoek van vasculaire mechanisme. Oefening training is een belangrijke strategie voor het behoud van de gezondheid en het voorkomen van veel chronische ziekten.Het combineren van lichaamsbeweging met hypoxie leidt tot specifieke cellulaire en moleculaire aanpassingen, met potentiële therapeutische toepassing.
