リンパ腫は組織学的に複雑な腫瘍である。したがって、多重化蛍光免疫組織化学は、従来の発癌性免疫組織化学よりも、腫瘍細胞および微小環境における関心のマーカーを研究する利点を提供する。この技術は、腫瘍微小環境の複雑さのために歴史的に困難であった組織学的リンパ腫サンプルにおける抗体ベース染色のスコアリングを標準化する可能性を有する。具体的には、多重化蛍光免疫体化学およびスペクトルイメージングにより、1枚のスライド内で複数の染色の定量的測定が可能となり、臨床物質内における抗原の共発現およびスペシャス関係の評価が可能となる。まず、脱ワックスおよび水分補給スライドをマイクロ波保存ガラス瓶の標準的な抗原検索バッファに浸漬し、適切なマイクロ波で熱誘導エピトープ検索を行います。最終マルチプレックス配列における抗体の位置に基づいてマイクロ波ストリッピングの追加ラウンドを行います。例えば、第4抗体は、染色前にマイクロ波ストリッピングの3ラウンドを追加する必要があります。ストリッピングプロセスが完了した後、鉗子と耐熱手袋を使用して、スライドを室温で蒸留水に移し、少なくとも10分間冷却します。抗原回収液が冷却されたら、市販のペルオキシダーゼブロックで組織ペルオキシダーゼ活性を10分間遮断し、続いてトリス緩衝生理食物、またはTBS、および非イオン性洗剤で5分間洗浄する。ブロッキングバッファーでの10分間のインキュベーションを行い、その後室温で目的とする一次抗体を30分間インキュベーションし、TBS-Dバッファーで5分間3回の洗剤を行い、非特異的染色をブロックします。次に、サンプルを適切な西洋ワサビペルオキシダーゼ共役二次抗体を室温で15分間インキュベートし、続いてTBS-Dバッファーに3回の洗剤を加えます。最後の洗浄後、適切なチラミドベースの蛍光試薬をスライドに適用し、室温で5分間インキュベーションし、その後に3回の5分間のTBS-D洗浄を行います。新しい抗原検索溶液中の一次抗体および二次抗体を除去するために、マイクロ波ベースの追加ストリッピングを実行します。取り出しの最後に、蒸留水でスライドを冷却し、ラボワイプでティッシュなしでスライド上の領域を乾燥させます。室温で10分間ダッピーでスライドをインキュベートし、その後3回のTBS-D洗い物をします。次に、組織に接触することなくラボワイプでスライドを慎重に乾燥させ、イメージング用の適切な取り付け媒体でサンプルを取り付けます。モノプレックススライドスキャンの場合、サンプルの標識に使用するフルオロフォアから適切なフィルタを選択し、各マーカーとそれに対応する蛍光チャネルを調べて、クリーンなシグナルを得るための適切な露出時間を特定し、マーカーに対して最も強いシグナルを持つべき組織成分に焦点を当てます。ピクセルの強度をクロスサンプルで比較できるようにするには、ライブカメラの設定を使用して、ライブカメラの画像に露出超過の領域がなくなるまで、個々のチャンネルの露出時間を調整します。すべてのスライドをスキャンしたら、シミュレートされた明視野画像を取得して、通常の免疫組織化学的パターンと視覚的に比較し、染色パターンが正しいかどうかを判断します。組織マイクロアレイスライドをスキャンするには、画像システムで組織マイクロアレイスキャンモードと適切なオートフォーカスアルゴリズムを使用します。組織セクション全体のスライドについては、最も代表的な腫瘍領域から最適な画像を選択するために、修飾された病理学者によってレビューされたスライドを持っています。分析を開始する前に、腫瘍マーカーを選択して、分析対象の細胞を特定します。病理学者に画像を確認してもらい、組織のセグメンテーションが必要かどうかを判断します。必要に応じて、セグメンテーション用の適切な制御領域を選択し、画像解析ソフトウェアでそのような領域を正しく識別できるかどうかを確認します。細胞セグメンテーションの後、病理学者にセグメンテーションマップをレビューし、意図したセグメンテーションアプローチの忠実性を確認し、個々の画像を確認して、細胞セグメンテーションが適切であるかどうか、または腫瘍細胞、ストロマ、および壊死のために富む領域を選択するために追加の組織セグメンテーションが必要かどうかを判断します。次に、各マーカーの光学強度正カットオフ値を病理学者と組み合わせて決定し、適切な統計ソフトウェアプログラムでヒストグラムを生成し、細胞あたりのマーカー強度の周波数分布を解析する。定義済みのセル内の特定のマーカーのパーセンテージデータを生成するには、データシートにピボット テーブルを挿入し、[合計] を選択して、対象の 1 つのマーカーの陽性率を計算します。マーカーの正のセルの総数を合計数で割った値を計算します。結果は、1 つのコアまたは 1 つのスタディ番号を持つマーカーの正のセルの割合です。数値データを生成するには、サンプル内のすべての細胞研究における対象の各マーカーの平均強度を取得し、各コアまたは研究番号の各マーカーの中央値の正規化されたカウントを抽出します。ここで、c-MYCおよびBCL2遺伝子再構成を伴う拡散性大細胞B細胞リンパ腫試料に対する代表的な多重蛍光免疫体細胞化学的画像が示されている。シミュレートされた明視野免疫組織化学画像は、同様の腫瘍マーカー染色パターンを示す。血管免疫芽細胞性T細胞リンパ腫におけるT細胞パネルへの多重蛍光免疫組織化学画像の適用により、腫瘍試料の細胞不均一性が明らかになる。ここでは、扁桃腺制御試料の最適化と結果データ分析を示す。腫瘍細胞および腫瘍微小環境におけるマーカー発現および局在化を定量化するために、アンミックスステップの後に様々な画像解析ソフトウェアプログラムを使用することができる。この技術は、研究者がリンパ腫の単一組織セクションで特定の細胞型内の複数のマーカーの共同発現を研究する道を開きます。マイクロウェーブステップ中に熱による損傷を防ぎ、デッキで行われるスキャンステップ中の落下リスクに注意してください。
