Infectie-gebaseerde experimentele modellen zijn inherent uitdagend vanwege hun afhankelijkheid van de startende staat voorwaarden van de gastheer en ziekteverwekker, de verschillende pathogen infectie strategieën en de moeilijkheid van de toerekening van de pathogeen - en gastinstellingen gestuurde processen gebaseerd op de resultaten. De bacterie Listeria monocytogenes (Lm) is een ideale pathogen sonde host defense reacties vanwege haar genetische en microbiologische werkwillig, haar snelle en processive cellulaire infectie-strategie en de relatief duidelijk geworden relatie tussen de mobiele - en organismic-level infectie fenotypen. De cellulaire infectie van Lm verloopt via vier fasen1: (i) cellulaire invasie die wordt afgesloten met Lm worden omsloten binnen een vacuole; (ii) Lm-gestuurde ontbinding van de vacuole membraan en introductie van Lm in het cytosol; (iii) intracytosolic replicatie; en (iv) fysieke penetratie van het plasma-membraan dat in ofwel de infectie van de direct aangrenzende cellen (zoals in een epitheliale blad) of, in de eenzame cellen resulteert, introductie van Lm in de extracellulaire milieu. Elk van deze fasen worden gepromoot door specifieke Lm-gecodeerd factoren (hierna aangeduid als 'virulentiefactoren') die, wanneer verwijderd, infectie defecten veroorzaken in zowel cellulaire en dierlijke modellen. Deze algemene infectie strategie is onafhankelijk geëvolueerd door een aantal van de zogenaamde 'cytosolische' ziekteverwekkers2.
Kolonie-vormende eenheid (CFU) testen te evalueren zowel in vitro algemeen werkzaam zijn (dat wil zeggen, cellulaire) zo goed als in vivo (d.w.z. organismic) infectie resultaten. Naast hun hoge gevoeligheid, met name voor in vivo infecties, voorzien CFU assays in een duidelijke uitlezing pathogen invasie en intracellulair overleven/proliferatie. CFU testen zijn uitgebreid gebruikt voor het analyseren van zowel Lm en gastheer cel bepalende factoren die van invloed zijn infectie. Zo informatief als deze voorafgaande studies zijn geweest voor het analyseren van cellulaire invasie en intracytosolic replicatie, CFU testen hebben, tot de beste van onze kennis, niet gebruikt voor het bijhouden van de vierde fase van het proces van de infectie Lm : cellulaire ontsnappen. Hier beschrijven we relatief eenvoudig hoe cellulaire ontsnappingsvoorzieningen (hierna te noemen 'opkomst') kan worden gecontroleerd door CFU assay (en stroom cytometry) en Toon voorbeelden van hoe beide factoren pathogen - en host-gecodeerde regelen deze fase van de Lm infectie cyclus. De analyse van de terminale fase van de cellulaire Lm infectie cyclus kan maken het mogelijk om extra pathogenen en host cel infectie-specifieke factoren en activiteiten te identificeren.

Er zijn een verscheidenheid van strategieën bacteriële pathogenen in dienst om te overleven en eenmaal binnen de eukaryote cel vermenigvuldigen. De zogenaamde 'cytosolische' pathogenen (Listeria monocytogenes, Shigella flexneri Burkholderia pseudomallei, Francisella tularensisen Rickettsia spp) toegang tot de geïnfecteerde cel cytosol door fysiek en enzymatisch verlaagt het primaire vacuolar membraan. Eenmaal in het cytosol, deze ziekteverwekkers zowel vermenigvuldigen als het genereren van voldoende mechanische krachten te doordringen van het plasma-membraan van de gastheercel om nieuwe cellen infecteren. Gebruik geconditioneerde media van niet-geïnfecteerde cellen als verdunningsmiddel om een 10 microgram per milliliter gentamicine oplossing voor te bereiden.Voeg na 30 minuten na infectie voorzichtig 30 microliter van de gentamicine-oplossing toe aan de schotel om een uiteindelijke concentratie van twee-punt-vijf microgram per milliliter te bereiken. Voorzichtig pipette de inhoud van de put te mengen en terug te keren van de cel cultuur schotel naar de couveuse. Om te oogsten, lichtjes kantelen van de schotel en voorzichtig gebruik maken van een pipet om de hele media te verwijderen uit de put.Voeg punt-vijf milliliter gedistilleerd, steriel water aan de put. Breng na 30 seconden het resulterende waterlysaat gedurende 10 seconden krachtig over op een microcentrifugebuis van één punt en vortex. Voeg vier-punt-vijf microliter en 50 microliter van het water lysaat aan 450 microliter van gedestilleerd, steriel water om de tienvoudige en honderdvoudige verdunningen voor te bereiden.Kort vortex om een grondige mix te garanderen. Voeg 50 microliter van de originele tienvoudig verdunde en honderdvoudig verdunde monsters toe aan lysogeny bouillonplaten en verdeel met een plaatspreader. Om te testen voor opkomende LM, extract 10 microliter van de overlaying media uit de schotel en verdeel het in twee vijf-microliter aliquots en micro centrifuge buizen.Voeg vijf microliter van DMEM/ FCS toe aan een aliquot en voeg vijf microliter DMEM/FCS met vijf microliter per milliliter gentamicine toe aan de tweede aliquot als controle. Wacht vijf minuten op kamertemperatuur. Voeg 90 microliter gedestilleerd steriel water toe aan elk aliquot en vortex de twee krachtig gedurende 10 seconden.Voeg vervolgens 50 microliter van het monster op LB agar platen en verspreid met behulp van een plaatspreader. Bewaar de platen in 37 graden celsius incubator voor twee dagen voor het opsommen van Lm kolonies. Met behulp van een geautomatiseerde cel teller, pas de voorbereide, ruwe twee-zes-vier-punt-zeven cellen aan de concentratie van een keer 10 tot de zesde cellen-per-milliliter.Voeg een milliliter van het monster toe aan putten van een zes-goed weefselkweekplaat. Infecteer de cellen met een voorbereid Lm-entmateriaal, wat overeenkomt met een veelheid aan infectie van 50. Op een-punt-vijf uur na infectie, in een one-point-five milliliter microcentrifuge buis gevuld met 500 microliters van vier procent PFA, verzamelen media uit de eerste set van putten en plaats de buis op ijs.Om intracellulaire Lm te verzamelen, voeg een milliliter gedistilleerd, steriel water aan elke put. Breng na 60 seconden het resulterende waterlysaat over naar een microcentrifugebuis van één punt met 500 microliter van vier procent PFA. Vortex de buis krachtig gedurende 10 seconden en plaats het op ijs.Voor de resterende putten, verwijder de media en vervang met een milliliter DMEM / FCS met vijf microgram per milliliter gentamicine te doden extracellulaire Lm.Prompt terug te keren naar de couveuse. Na 30 minuten, verwijder de media en vervang met DMEM / FCS zonder gentamicin. Na nog eens twee en vier uur, verzamelen media in lysates voor de tweede en derde keer punten in de buizen en leg ze op ijs om te koelen.Centrifuge buizen op 10.000 keer G gedurende zeven minuten. Verwijder met behulp van een pipet supernatant zonder de pellet te storen. Voeg kleuringcocktail met 50 microliters van vier procent PFA en 15 microliter phalloidin in de buis en meng om elke pellet op te schorten.Incubeerbuizen in het donker op vier graden Celsius gedurende 20 minuten. Na incubatie, voeg een milliliter FACS buffer aan elke buis en pipet op en neer te mengen. Spinbuizen in de centrifuge op 10.000 keer G gedurende zeven minuten.Verwijder supernatant zonder de pellet te storen. Voor onmiddellijk gebruik, hang elke pellet in 400 microliter van FAX buffer. Als opslag voor latere analyse, voeg 200 microliter fax buffer en 200 microliters van vier procent PFA in de buis en meng op te schorten.Met behulp van de infectie voorwaarden in de korte behandeling met het antibioticum gentamicine te elimineren niet-geïnternaliseerde Lm, nul-punt-een-vijf procent van de wilde-type Lm wordt hersteld na een-punt-vijf uur co-incubatie met gekweekte macrofagen. In de daaropvolgende co-incubatie was er een verviervoudiging en zevenmaal vervijfvoudigde toename van het herstel van levensvatbare Lm, die uitsluitend intracellulaire proliferatie vertegenwoordigt. Vergelijkbaar profiel werd waargenomen voor de Lm stam met een hypervirulente prfA tijdens de eerste infectie, maar niet tussen de drie en zes HPI.Wanneer de prfA uit de Lm-stam is verwijderd, is er een daaropvolgende vermindering van het herstel van de stam na langdurige co-incubatie. Wanneer gentamicine wordt verwijderd uit de putten op zes HPI, extracellulaire Lm kan worden gedetecteerd een uur later voor zowel wilde en hypervirulente prfA stammen, maar niet voor de stam met prfA verwijderd. Media die zowel niet-geïnfecteerde als geïnfecteerde macrofagen overlays bevatten lichtverstrooiing, bacterie-sized fijnstof.Echter, alleen de media van geïnfecteerde macrofagen bezat GFP positieve signalen binnen de grootte gating. Een tijdafhankelijke toename van het uiterlijk van phalloïden-positieve Lm werd waargenomen met geïnfecteerde macrofagen. Bij het plating van de macrofagen in de 48-well weefsel cultuur schotel, laat een aantal putten zonder cellen.Deze no-cell controleputten worden dan precies hetzelfde behandeld als de celhoudende putten, in termen van de toevoeging van de ziekteverwekker, antibiotica, enz. Vergelijkbaar met de experimenten in figuren een en drie, kunnen pathogene of gastheerfactoren en of kleine moleculen worden getest op hun impact op pathogene cellulaire efflux.
