Utilizando una fuente adecuada de células madre neurales es muy importante para los neurólogos. Las células madre neurales (NSC) residen en diferentes regiones del cerebro adulto y el embrión y que tienen una tendencia a generar diferentes tipos de neuronas y células gliales1. Las zonas subventricular del cerebro adulto de mamíferos2 y eminencia ganglionar en el cerebro del embrión son de las regiones ricas de NSC3. En el cerebro en desarrollo, la eminencia gangliónica ofrece la mayoría de las interneuronas corticales y particularmente de interneuronas de GABAérgico3. También hay métodos menos invasivos para la generación de células madre neuronales de las células madre embrionarias (ESCs) o uso de células madre pluripotentes inducidas (iPSCs) que reducen la demanda para el animal. A pesar de que la generación de NSCs de CES o iPSCs es posible4,5, tiene algunas ventajas y desventajas en comparación con el aislamiento de las células madre neurales del adulto o embrionario cerebro6,7, 8. Los protocolos para la inducción de la diferenciación del CES y iPSCs hacia fenotipos neuronales son siempre tiempo y coste de consumo y la tasa de éxito (70-80% Nestin las células positivas)5 es inferior comparada con aislamiento directo de NSCs del (cerebro animal más de 99% Nestin las células positivas)9. Por otra parte, las células madre pierden su tendencia genética de la diferenciación y estabilidad después de varios pasajes10,11. Aunque hay otros nuevos informes acerca de la conversión directa de células somáticas en NSCs, estas células son genéticamente modificadas y no son fácilmente accesibles en cada laboratorio12. Por lo tanto, todavía hay una gran demanda para el aislamiento de las células madre neurales del cerebro animal; es posible reducir la cantidad de uso animal mediante la mejora de las técnicas quirúrgicas. Las células madre neurales (NSC) son multipotentes y puede dar lugar a los tres tipos de células importantes del sistema nervioso central (SNC). In vitro cultura y expansión de NSCs proporcionan una fuente adecuada de células de neurocientíficos a estudiar la función de las neuronas y células gliales junto con sus interacciones. Limpiar y desinfectar la piel del área del abdomen rociando con 70%etanol. Controla la piel hacia arriba del abdomen y luego corta la piel y subraya el facial con tijeras para descubrir la cavidad abdominal y los cuernos uterinos.Retire los cuernos uterinos que contienen los embriones mediante tijeras y transfiéralos a un tubo cónico de 50 ml que contenga 25 ml de tampón HEPES MEM frío. Coloque el tubo cónico debajo de la capucha de flujo laminal. Abra la tapa debajo del capó.Saque los cuernos uterinos del tubo y enjuague tres veces con suficiente volumen del tampón HEPES MEM frío para eliminar los desechos y la sangre. Sostuvo el cerebro y colocaba la eminencia gangliónica diseccionada en el tubo centrífugo de 15 ml que contiene medios de células madre neurales.Repita este procedimiento hasta que todo el cerebro haya sido micro-diseccionado. Corte la eminencia gangliónica diseccionada colocando la punta de la pipeta contra la parte inferior del tubo y pipetee la suspensión hacia arriba y hacia abajo durante 2 5 veces para romper el tejido para obtener una suspensión de una sola célula. Centrifugar la suspensión a 110 g durante 5 minutos.Retire el sobrenadante y resusppend las células en 1 ml de tubo 0.05% en EDTA. Incubar la suspensión celular en un grado de 37 grados Centígrados durante 10 minutos. Y luego añadir un volumen equivalente de inhibidor de la trippsina de soja para detener la actividad de la trippsina.Pipetear la suspensión celular hacia arriba y hacia abajo para asegurarse de que la trippsina ha sido totalmente inactivada. Centrifugar la suspensión celular a 110 g durante 5 minutos. Retire el sobrenadante y resusppend las células en 1 ml de medio de células madre neurales completas y cuente el número de células.Placar las células en el medio de células madre neurales en el tamaño adecuado matraz de cultivo de tejido. Transfiera 5 ml de 2T25, 20 ml a T75 y 40 ml a matraces T175. Las neuroesferas aparecerán después de 3 días de cultivo a 37 grados Celsius en una incubadora humidificada con 5%CO2.Para aspirar el cultivo de las neuroesferas, transfiera el medio con neuroesferas suspendidas a la centrífuga a centrifugar a 110 g durante 5 minutos, y luego deseche el sobrenadante. A 1 ml de 0.05%trypsin EDTA e incubar a 37 grados Celsius durante 10 minutos. Luego inactivar la tripina usando inhibidor de la tyrpsina de soyben y pipetear las neuroesferas suavemente hacia arriba y hacia abajo para hacerlas células individuales.Centrifugar la suspensión celular a 110 g durante 5 minutos. Deseche el sobrenadante y resusppend las células en 1 ml de medio de células madre neurales. Estas células están listas para el cultivo o cultivo en el siguiente paso en laminado y superficie completamente recubierta para experimentos avanzados.Al cultivar un millón de células madre neuronales primarias después de siete días, el número de células aumentará a tres a cuatro millones. Después de seis a siete días las esferas deben medir entre 150 a 200 micrómetros de diámetro y estarán listas para el subcultivo en platos laminados recubiertos de poli ornitina en ausencia de bFGF y EGF para la diferenciación neuronal. Los resultados del ensayo de citometría de flujo muestran que cerca del 95% de las células que constituyen las neuroesferas eran Nestin positivas, que es un marcador conocido para las células madre neurales.Los ensayos que utilizan anticuerpos de proteínas fibrosas beta tubulina III y ganglio revelan que al cultivar células madre neurales en superficie recubierta alrededor del 94% mostraron fenotipo neuronal mostrado y alrededor del 5% muestran fenotipo glial.
