1. 真菌分离物和孢子悬浮液

</ol>5. 从蓝莓花 (BB rw-FE)中收集雨水16
注: 蓝莓花雨水收集装置包括带连接适配器 (杯: 女线、适配器: 男性至男性螺纹) 的空气喷枪一次性喷漆杯、50毫升离心管 (聚丙烯)、副管和塑料涂层电线 (标准电话线, 单独的内部线绞线内容)。
<ol><li>在创建收集设备之前, 在蓝莓灌木中选择多个位置, 以捕获从鲜花中流出的雨水。这些包括直接下花序 (花) 到灌木 (冠) 的底部。记录选定位置的茎直径 (范围为1-5 厘米), 因为这将决定用于设备附件的孔的大小, 如下所述。</li>
	<li>对于每个选定位置, 创建一个收集设备。首先在喷雾杯底部钻一个孔 (杯向螺纹开口弯曲) 到相应的阀杆直径, 并在钻床上连接一个步位。然后, 从孔顶切割一条直线到喷杯的口。钻4等距孔 (足够大, 可以螺纹塑料涂层线), 在喷杯的口, 并连接一端的塑料涂层线, 使一端无自由。</li>
	<li>钻一个足够大的孔, 将喷漆杯适配器固定在50毫升的离心机盖上。密封适配器螺纹与副体, 以防止泄漏。将其连接到喷雾器杯的离心机盖和螺纹部分。连接配合50毫升的离心管。</li>
	<li>重复步骤 5.3-5.4, 根据选定位置创建多个设备, 每个采样位置至少有4个收集设备。</li>

</ol>6. 从越橘花中收集雨水 (CB rw-FE)
<ol><li>红莓中的花雨水收集装置由 7 X 7 厘米聚丙烯漏斗、50毫升离心管 (聚丙烯)、准体和4个标准的塑料涂层扭曲带 (每个设备) 组成。</li>
	<li>首先, 热穿刺8等距孔 (直径的扭曲领带) 周围的漏斗口使用金属探头。将扭曲带插入4个孔。将另一端连接到该孔的相对位置, 形成一个整齐的交叉图案。用护栏包漏斗下的阀杆, 放在一边。</li>
	<li>钻一个足够大的孔, 将漏斗下杆插入一个50毫升的离心机盖, 带有一个阶梯式。将准备好的漏斗插入离心机盖。重复步骤 6.2-6.3 创建多个设备, 每个采样位置至少4个收集设备。</li>
	<li>将标记 (datetime) 设备部署到选定的红莓沼泽中。在交叉的塑料扭曲领带下, 整齐地塞住了两座带花的花序 (被称为立柱)。然后垂直定向设备通过刺穿离心管进入越橘天篷 (补充图 7-8,补充电影 3)。</li>

</ol>7. 使用水基花卉提取物15,16 (活性-fe,通过-fe, rw-fe) 进行生物测定

	<li>将预先清洁的玻璃盖板放在细胞培养盘中浸泡过的纸巾上。将40μl 水处理混合物液滴放在覆盖液的中心。重复所需的治疗, 包括控制, 关闭细胞培养盘。在单独的细胞培养皿中重复复制 (至少 3个) 和时间点 (每个培养皿为1个时间点)。一旦所有的治疗和复制被分发, 把所有复制的细胞培养盘放入一个密封的塑料容器 (30 x 13 x 7 毫米), 并在25°c 在黑暗中孵育。</li>
	<li>在预定的时间点上, 在液滴中加入10μl 的固定剂, 如乳突棉蓝 (20.0 g 苯酚晶体, 20.0 ml 2.5% 乳酸, 40.0 毫升甘油, 0.05 克棉蓝), 停止生长, 半保持安装。<ol>

	</ol>
</li>

	<li>在盖板上 (初级圆锥花序、萌发的圆锥花序和新形成的次生圆锥花序) 以及400X 放大倍率 (计数16个字段) 或 200X (计算4个字段) 上的所有圆锥花序 (总圆锥花序) 进行计数, 总面积为3.08 毫米2. 复制整个生物测定进行统计分析。对于使用水性 FE 进行检测, 请按照 Waller 等人的描述分析数据.2017年16日, 通常显示为平均计数 2 (总圆锥花序或闭应菌)。</li>
</ol>8. 使用氯形式的花卉提取物 (ch-FE) 进行生物测定17
