Oppervlakte markeringen worden vaak gebruikt om specifieke celtypen in vitro en in vivo17,18te labelen en te zuiveren. De ontdekking van oppervlakte markers levert een grote bijdrage aan regeneratieve geneeskunde en onderzoek door stamcel door moleculaire hulpmiddelen te bieden om een stamcel populatie selectief te verrijken van normale of pathologische weefsels en cultuur gerechten, met een gezuiverde cel bron voor klinisch gebruik of studie van biologische eige...

Neurale stamcellen worden gedefinieerd als een multipotente celpopulatie die zichzelf kan verlengen om een stamcel zwembad te behouden en zich te differentiëren in neuronen en gliacellen. Ze zijn de belangrijkste celtypen in het zenuwstelsel en kunnen een groot therapeutisch potentieel bieden in regeneratieve geneeskunde door middel van celtransplantatie in zieke en gewonde hersenen1,2. Naarmate de ontwikkeling vordert, wordt de neurale stamcel populatie heterogene3,4, en het aandeel van neurale stamcellen in de hersenen neemt geleidelijk af5. Over het algemeen bevinden embryonale stamcellen en andere neurale voorlopercellen, gezamenlijk neurale stam-en voorlopercellen (Nspc's), zich in de Germinal zones, de ventriculaire zone en de subventriculaire zone bij muizen6. In het embryonale brein genereren neurale stamcellen direct of indirect neuronen via tussenliggende voorlopercellen (ipc's), en in sommige soorten door de buitenste subventriculaire zone-voor ouders (orgs)7,8. De specifieke moleculaire handtekening, morfologie, locatie in de stamcel niche en differentiatie potentieel bepalen allemaal de rol van elk subtype in hersen organogenese en klinische toepassingen9. De momenteel beschikbare celoppervlak markeringen kunnen echter niet ondubbelzinnig verschillende subtypen van Nspc's discrimineren en zuiveren, waardoor het begrip en het gebruik van deze subtypen worden beperkt.
De identificatie van primaire NSPCs-oppervlak markeringen wordt beperkt door drie grote hindernissen. De eerste is het beperkte celnummer van Nspc's in het weefsel, waardoor het moeilijk is om celoppervlakte proteïne monsters voor te bereiden voor gemeenschappelijke massaspectrometrie analyse. De tweede beperking is de moeilijkheid bij het produceren van zuivere celsubtypen voor het genereren van subtype-specifieke membraan proteïne gegevens. Ten slotte is de derde uitdaging de lage ratio van celoppervlakte eiwitten in hele celeiwitten, wat hun detectie gevoeligheden belemmert door massaspectrometrie analyse.
Om deze problemen te overwinnen, ontwikkelden we een chemoproteomische benadering om celoppervlakte eiwitten in primaire Nspc's selectief te verrijken en te identificeren door de sialoglycoproteïnen10metabolisch te labelen. Om een voldoende aantal Nspc's te genereren, maakten we gebruik van een vastgesteld protocol om de primaire embryonale Nabc's in ongedifferentieerde toestanden uit te breiden en te handhaven in vitro, door het coculeren van Nspc's met muizen hersen endotheliale cellijnen met behulp van een permeabele ondersteuning matrix insert (bijv. trans well) systeem11. In tegenstelling, npscs gekweekt alleen zonder endotheel cellen genereren gedifferentieerde nageslacht11,12. Zo kunnen eiwit monsters van deze twee kweeksystemen relatief worden geanalyseerd om eiwitten te identificeren die differentieel uitgedrukt zijn in Nspc's en gedifferentieerde neuronen. Aangezien de meeste celoppervlakte eiwitten worden gemodificeerd door sialic acid13, onnatuurlijke sialic acid precursor analoge N-azidoacetylmannosamine-tetraacylated (AC4Mannaz) werd gebruikt om de intrinsieke metabole route te kapen, zodat endogene, nieuw gesynthetiseerde sialoglycans zijn gelabeld met meer groepen, het genereren van een chemische handvat14. Via azido-alkyne-gemedieerde bioorthogonale reacties, die biotine aan sialoglycans Conjugeren, kunnen celoppervlakte eiwitten worden gevisualiseerd en verrijkt voor proteomische identificatie door middel van een streptavidine-gekoppelde de of matrix14.
Hier voeren we kleuring van SDS-PAGE gel analyse van het oppervlak sialoglycoproteome uit Nspc's uitgebreid in een endotheliale co-cultuur en differentiërende cellen in een niet-co-cultuur systeem. We zuiveren ook selectief oppervlak sialoglycoproteome in de twee kweeksystemen voor proteomische vergelijking. Ons protocol, in vergelijking met de traditionele centrifugatie-based cel oppervlakte zuivering protocollen15, verhoogt de extractie werkzaamheid door het verminderen van de oppervlakte-EiwitExtractie procedures door middel van specifieke tag conjugatie en affiniteit Zuivering. Ondertussen, het verhoogt de extractie zuiverheid van celoppervlakte eiwitten op basis van de veronderstelling dat verminderde gebeurt meestal op het celoppervlak eiwitten. Hoewel endotheliale factoren de differentiatie van uitgebreide Nspc's niet volledig kunnen blokkeren, biedt de vergelijkende studie tussen een co-cultuur en gedifferentieerde cultuur een handige methode om stamcel verrijkte oppervlakte eiwitten te lokaliseren zonder de noodzaak om Analyseer eiwitten uit Npc's gezuiverd door FACS16. Wij zijn van mening dat deze aanpak kan worden toegepast op onderzoek van oppervlakte eiwitten in andere systemen met de juiste aanpassingen.

Neurale stam-en voorlopercellen (Nspc's) zijn de cellulaire basis voor de complexe structuren en functies van de hersenen. Ze bevinden zich in gespecialiseerde niches in vivo en kunnen worden geïsoleerd en uitgebreid in vitro, dienen als een belangrijke bron voor celtransplantatie om hersenbeschadiging te herstellen. Nspc's zijn echter heterogene en niet duidelijk gedefinieerd op moleculair niveau of gezuiverd als gevolg van een gebrek aan specifieke celoppervlak markeringen. Het gepresenteerde protocol, dat eerder is gerapporteerd, combineert een neurale-endothelial co-cultuur systeem met een metabole ontwikkelen-etiketterings methode om het oppervlak sialoglycoproteome van primaire nspc's te identificeren. Het NSPC-endotheliale co-cultuur systeem maakt zelf vernieuwing en uitbreiding van primaire Nspc's in vitromogelijk, waardoor een voldoende aantal nspc's worden gegenereerd. Sialoglycans in gekweekte nspc's zijn gelabeld met behulp van een onnatuurlijke sialic acid metabole reporter met bioorthogonale functionele groepen. Door het sialoglycoproteome te vergelijken met zelfverlengde Nspc's die in een endotheliale co-cultuur met differentiërende neurale cultuur zijn uitgebreid, identificeren we een lijst van membraan eiwitten die verrijkt zijn met Nspc's. In detail omvat het Protocol: 1) het opzetten van een NSPC-endotheliale co-cultuur en NSPC-differentiërende cultuur; 2) etikettering met azidosugar per-O-geacetoneerde N-azidoacetylmannosamine (AC4Mannaz); en 3) Biotine conjugatie aan gemodificeerde sialoglycan voorbeeld vorming na fixatie van neurale cultuur of EiwitExtractie uit neurale cultuur voor massaspectrometrie analyse. Vervolgens worden de NSPC-verrijkte oppervlak marker kandidaten geselecteerd door vergelijkende analyse van massaspectrometrie gegevens uit zowel de uitgebreide NSPC en gedifferentieerde neurale culturen. Dit protocol is zeer gevoelig voor het identificeren van membraan eiwitten van geringe overvloed in de uitgangsmaterialen, en het kan worden toegepast op marker ontdekking in andere systemen met passende modificaties

Neurale stam en voorloper cellen kunnen zichzelf vernieuwen en differentiëren in verschillende neurale celtypes, ondersteuning van het zenuwstelsel ontwikkeling en het aanbieden van een bron voor regeneratieve geneeskunde. Deze cellen zijn heterogeen, en we moeten hun specifieke celoppervlakmarkers kennen om een gezuiverde celpopulaties te krijgen en hun functies te begrijpen. Ons protocol biedt een eenvoudige, gevoelige en efficiënte techniek om membraaneiwitten van primaire neurale stam- en voorlopercellen te analyseren, waardoor nieuwe oppervlaktemarkers voor deze cellen worden geïdentificeerd.Het belangrijkste voordeel van ons protocol is het vermogen om het oppervlakproteoom van primaire neurale stam- en voorlopercellen direct te analyseren met de verbeterde gevoeligheid en specificiteit. Dit wordt bereikt door ze uit te breiden door co-cultuur binnen endotheelcellen in vitro en hoge tractine intrinsieke cellulatie MAPK-ERK traject naar oppervlakte porines label. Met porie pro-modificaties op het uitbreiden van stamcellen in vitro, kan ons protocol gemakkelijk worden toegepast om oppervlaktemarkers van andere stamceltypen te identificeren.Om de endotheelcellen voor te bereiden, schorst u een BEN3-celpellet opnieuw van een petrischaal van 10 centimeter bij 90%-samenvloeiing met negen milliliter BEN3-celmedium. Voeg een milliliter van de cel suspensie in een doorlaatbare steun insert. Voeg nog eens twee milliliter BEN3 medium per goed aan de onderkant kamer van de matrix.Blijf de cellen een dag kweken bij 37 graden Celsius met 5%kooldioxide. Een dag na het plating van de cellen in de inserts, zachtjes aspirate het medium, eerst uit de onderste kamer en vervolgens van de inserts. Was het gezicht van de wisselplaten drie keer met voorverwarmde DMEM.Was het buitenoppervlak van de wisselplaten door te spoelen met voorverwarmde DMEM. Voeg vervolgens een milliliter voorverwarmde AM toe in één insert. Breng de wisselplaten in de putten met primaire corticale cellen.Broed de co-cultuur op 37 graden Celsius met 5% kooldioxide gedurende 12 uur. Los Ac4ManAz op in DMSO om een voorraadconcentratie van 200 millimolar te bereiken. Haal de neurale-endotheel co-cultuur plaat uit de couveuse.Voeg een microliter van de Ac4ManAz-voorraad toe aan elke bodemkamer en 0,5 microliter per insert in de co-cultuur. Kweek de cellen op 37 graden Celsius met 5% kooldioxide gedurende vijf dagen. Terwijl de cellen zijn kweken voeg 100 microliter RM per insert en 200 microliter rm per onderste kamer om opnieuw voeden de endotheel en neurale cellen om de andere dag.Verwijder eerst de wisselplaten van de co-cultuurplaten en haal het kweekmedium uit de bodem van de putten. Volgende wassen de neurale cellen een keer met voorverwarmde PBS. Aspirate de PBS uit de putten en voeg een milliliter van een voorgekoelde 4%paraformaldehyde in PBS aan elke put.Bevestig de cellen op kamertemperatuur gedurende 10 minuten. Was vervolgens de cellen drie keer met voorgekoelde PBS. Aspirate de PBS en voeg een milliliter vers bereide biotine geconjugeerde buffer 1 in elke put.Incubeer bij kamertemperatuur gedurende 10 minuten. Daarna de reactiebuffer uit de putten aanzuigen en de cellen drie keer wassen met PBS. Voeg een milliliter kleuring buffer aan elke put van cellen en uitbroeden bij kamertemperatuur gedurende 30 minuten.Vervolgens, aspirate de kleuring buffer uit de putten en was de cellen drie keer met voorgekoelde PBS. Voeg een milliliter blokkeerbuffer toe aan elke put van cellen en incubeer 10 minuten bij kamertemperatuur. Verwijder de blokkeerbuffer uit de putten en voeg een milliliter primaire antilichaamoplossing toe aan elke put.Broed de cellen 's nachts op vier graden Celsius. De volgende dag, verwijder de primaire antilichaam oplossing uit de putten. Was de cellen drie keer met voorgekoelde PBS.Aspirate de PBS uit de putten en voeg een milliliter secundaire antilichaam aan elke put. Broed de cellen twee uur lang op kamertemperatuur. Spoel hierna de oplossing uit de putten aan en was de cellen drie keer met voorgekoelde PBS.Bereid eerst BTTAA cupric sulfaat complex 2, volledige eiwit re-suspensie buffer, eiwit wassen buffer 1, eiwit wassen buffer 2 en eiwit wassen buffer 3 zoals beschreven in het tekstprotocol. Verwijder vervolgens de wisselplaten van de co-cultuurplaten. Aspirate de cultuur medium uit de onderste putten en was de neurale cellen een keer met voorgekoelde PBS.Aspirate de PBS en voeg 200 microliters van voorgekoelde RIPA buffer aan elke put. Incubeer de platen op ijs gedurende vijf minuten en verzamel vervolgens de eiwitlyse in 1,5 milliliter buizen. Centrifuge op 12000 keer G en op vier graden Celsius gedurende 10 minuten om de cel puin pellet.Breng de supernatant in nieuwe 1,5 milliliter buizen en bepaal de eiwitconcentratie met een BCA kit volgens de instructies van de fabrikant. Pas vervolgens de eiwitconcentratie aan op één milligram per milliliter. Voeg 100 micromolar alkyde biotine, 2,5 millimolar natrium ascorbaat en 1XBTTAA cupric sulfaat complex 2 tot een milliliter eiwitlyse.Meng de oplossing goed en broed het een uur op kamertemperatuur. Breng hierna de reactieoplossing over in 20 milliliter voorgekoelde methanol in een conische buis van 50 milliliter. Meng goed en incubeer 's nachts bij min 30 graden Celsius om de eiwitten neer te halen.Centrifuge op 4500 keer G en bij vier graden Celsius gedurende 15 minuten om het eiwit neer te laten neerslaan. Was de pellet twee maal met 20 milliliter voorgekoelde methanol per wasbeurt. Vervolgens, aspirate de supernatant en opnieuw opschorten van de eiwitpellet in vier milliliter van re-suspensie buffer.Breng de eiwithersuspensie over in een nieuwe conische buis van 15 milliliter. Was 50 microliter streptavidinkralen drie keer met PBS. Voeg de gewassen kralen toe aan de eiwithersuspensie en broed de oplossing drie uur lang in vier graden Celsius op een verticale rotator bij een rotatiesnelheid van 20 tpm.Daarna was je de kralen achtereenvolgens met elke wasbuffer die hier wordt getoond. Stel de gewassen kralen opnieuw op met 20 microliter PBS en breng ze over in een nieuwe buis van 1,5 milliliter. Voeg 20 microliter 2x eiwitbeladingsbuffer toe en behandel 10 minuten bij 95 graden Celsius.Verwerk vervolgens de eiwitmonsters met SDS-PAGE en bevlek ze met Coomassie Brilliant Blue zoals beschreven in het tekstprotocol. In deze studie primaire embryonale NSPCs worden uitgebreid en metabolisch gelabeld in vitro. Succesvolle endotheelco-cultuur leidt NSPCs om grote blad-als klonen te vormen.Immunostaining met antilichamen blijkt dat in de kloon, de meeste van de cellen zijn Nestin positieve NSPCs, terwijl zeer weinig zijn beta tubuline 3 positieve neuronale cellen. In tegenstelling, het percentage van Nestin positieve cellen en beta tubuline 3 positieve neuronale cellen in klonen gevormd in niet-co-cultuur systeem zijn bijna hetzelfde. De chemische verslaggever Ac4ManAz is een metabolische analoog en kan worden opgenomen in de intrinsieke eiwit sialytatie traject.Een geoptimaliseerde etiketteringsconcentratie van 100 micromolar voor primaire NSPCs wordt gebruikt en een combinatorische evaluatie van celdood aangegeven door cellulaire en kernmorfologie suggereert dat deze etiketteringsconcentratie geen duidelijke cytotoxische effecten veroorzaakt en in staat is om NSPCs efficiënt te labelen. De kloonmorfologie, zelfvernieuwing en differentiatiepotentieel van NSPCs in zowel de endotheelco-cultuur als het niet-co-cultuursysteem worden niet beïnvloed. Eiwit monsters bereid uit de Ac4ManAz gelabeld cultuur door biotine conjugatie en streptavidin kralen zuivering tonen sterke Coomassie Brilliant Blue kleuring signaal in SDS-PAGE gels.Ondertussen waren er alleen vlekken achtergrond en niet-specifieke bindende signalen in de rijstroken geladen met eiwit monsters van de DMSO controlegroep. Dit duidt ook op de efficiënte etikettering van NSPCs door Ac4ManAz. Bij het proberen van dit protocol moeten alle oplossingen die nodig zijn voor biocellulaire reactie vers worden voorbereid en moet de reactietijd nauwkeurig worden gecontroleerd om overte komen van onvoldoende reactie te voorkomen.Volgens ons protocol kan de structuur van neurostamcel verrijkt oppervlak glycan worden geanalyseerd. Ons protocol van neurale stam en voorloper cel verrijkte oppervlakte-eiwitten niet alleen stijging markers, maar ook spelen belangrijke functies op de verhoogde patroon zuivering strategie en de illustratie van rigidititory signaal circuits voor deze cel populatie.
