Kloof kruispunten (GJs) fungeren als intercellulaire kanalen om de verspreiding van kleine moleculen van < 1 kDa zoals voedingsstoffen en metabolieten signalering moleculen tussen aangrenzende cellen. De regulates elementen bevatten een hemichannel of connexon in elke cel en elke connexon vormt zes connexins (Cxs)1. GJs en Cxs zijn gebruikt in de toxicologie testen van kankerverwekkende stoffen zoals polycyclische aromatische koolwaterstoffen (PAK's), die GJ remmers,2,,3,4 zijn. Verstoorde GJIC is geassocieerd met nongenotoxic carcinogenese5,6. Als een potentiële therapeutisch doel, heeft in bijzonder subtypen van vangsten7,8, bescherming tegen hart- en hersenen ischemie/reperfusie letsel9, migraine met aura10GJ betrokkenheid gemeld drug-geïnduceerde lever schade6,11, en wond genezing van12. High-throughput screening (HTS) testen zijn vereist om GJ-modulerende chemicaliën of antilichamen voor drugontdekking, voor Toxicologie testen, te identificeren en te identificeren roman cellulaire regulatoren van GJ activiteit. HTS testen kunnen ook worden gebruikt om te onderzoeken van structuur-activiteit verhoudingen van GJ modulatoren2,13,14,15.
Sommige GJIC tests omvatten dye overdracht of dual patch-clamp technieken. Lucifer gele CH (LY) en calceïne acetoxymethyl ester (calceïne-AM) zijn gebruikt in dye-overdracht testen. Cellen zijn niet luchtdoorlatend LY, die is ingevoerd door microinjection, schraap laden of electroporation. Eenmaal in de cel, LY verspreidt in naburige cellen via GJs en GJ activiteit wordt bepaald door de omvang van de LY migratie16. Calceïne-AM testen is meestal sprake van herstel van de kloof-fluorescentie na photobleaching17,18. Calceïne-AM is een cel-permeant kleurstof die door een intrinsieke nte esterase intracellulair wordt omgezet in ondoordringbare calceïne. De bepaling vereist een confocal microscoop te observeren van de overdracht van calceïne-AM in een cel van die eromheen na laser photobleaching. Als functionele GJs aanwezig zijn, calceïne-AM in aangrenzende cellen binnenkomt de photobleached cellen en de fluorescentie wordt teruggewonnen. GJ activiteit wordt bepaald door de mate van herstel van de fluorescentie van de photobleached cellen. Dye-overdracht testen zijn moeizaam en tijdrovend of lage gevoeligheden. Dual patch klemmen is een elektrofysiologische methode die regulates geleidbaarheid maatregelen. Het is relatief gevoelig, met een directe afhankelijkheid van geleidbaarheid van het aantal open GJs19; echter, het is technisch veeleisende, tijdrovend en duur20. De ik-YFP-GJIC-assay werd ontwikkeld voor gebruik in de HTS.
Figuur 1 illustreert de onderdelen en de stappen van de ik-YFP-GJIC-test, die gebruik maakt van de acceptor cellen uiten een jodide-gevoelige YFP variant, rekening houdend met H148Q en I152L (YFPQL) en donor cellen uiten van een transporter jodide (SLC26A4)21 . De twee mutaties gedragen door YFPQL toestaan blussen van fluorescentie jodide22. Jodiden worden toegevoegd aan mede gekweekte acceptor en donor cellen; ze doen niet de acceptor cellen invoeren, maar worden overgenomen door de SLC26A4 vervoerders aanwezig op de donor cellen. Jodiden in de donor cellen verspreiden via functionerende GJs in aangrenzende acceptor cellen waar ze de YFPQL -fluorescentie quench. Als GJs gesloten of geblokkeerd door remmers, kunt geen jodide de acceptor cellen doven de fluorescentie invoeren. De YFPQL blussen tarief weerspiegelt GJ activiteit. De ik-YFP-procedure voor de bepaling van de GJIC is niet ingewikkeld of tijdrovend. Het is compatibel met HTS en kan worden gebruikt voor het testen van de effecten van een groot aantal verbindingen op GJ activiteit in een relatief korte periode. Het vereist alleen acceptor en donor cellen, en de twee evenwichtige zoutoplossingen. Het hieronder beschreven protocol is gebaseerd op de LN215 cellen waarvan grote Cx Cx4321 is. De LN215-YFPQL -receptor en de LN215-ik− donor cellen werden gegenereerd door transductie met lentivirussen YFPQL of SLC26A421,23te uiten.

an iodide-yellow fluorescent protein-gap junction-intercellular communication assay

Kloof kruispunten (GJs) zijn celmembraan kanalen waarmee verspreiding van moleculen kleiner dan 1 kDa tussen aangrenzende cellen. Als er fysiologische en pathologische rollen, moet er van high-throughput screening (HTS) tests om te identificeren GJ modulatoren in drug discovery en toxicologie testen. Een roman jodide-geel fluorescerende eiwit-gap junction-intercellulaire communicatie (I-YFP-GJIC) assay vervult deze behoefte. Het is een cel-gebaseerde test met inbegrip van de donor en acceptor cellen die zijn ontworpen om te stabiel express een gele fluorescerende eiwit (YFP)-variant, waarvan fluorescentie gevoelig door jodide, of SLC26A4, een transporter jodide, respectievelijk uitgeblust is. Wanneer jodide wordt toegevoegd aan een gemengde cultuur van de twee celtypes, zij voert de donor cellen via de SLC26A4 transporter en diffuus aan de cellen van de aangrenzende acceptor via GJs waar ze de YFP fluorescentie doven. YFP fluorescentie is goed door goed gemeten in een kinetische modus. De YFP blussen tarief weerspiegelt GJ activiteit. De test is betrouwbaar en snel genoeg om te worden gebruikt voor de HTS. Het protocol voor de ik-YFP-GJIC-assay met behulp van de LN215 cellen, menselijke glioma cellen, wordt beschreven.

Negenentwintig cultuur van de 96-wells-platen werden gebruikt om het scherm 2.320 chemische stoffen ter identificatie van roman GJIC modulatoren door ik-YFP-GJIC bepaling met behulp van de LN215-YFPQL en LN215-ik− cellen. Gap knooppunten zijn voorgesteld als drugs doelwitten. Iodide-YFP-GJIC-test is compatibel met high-throughput screening om gap junction modulators te ontdekken. Het is gebruikt om de effecten van een groot aantal verbindingen op gap junction activiteiten te testen in een relatief korte periode.Iodide-YFP-GJIC test is eenvoudig, robuust, herhaalbaar en goedkoop. Deze test vereist alleen acceptor- en donorcellen en twee evenwichtige zoutoplossingen. Er zijn geen extra reagentia nodig, zoals Lucifer geel en Calcein AM.Ook meet het totale gap junction activiteiten van de cellen in een enkele put die resulteert in een lage tussen-goed variabiliteit. Om te beginnen met deze procedure groeien LN215 cellen tot 80%samenvloeiing in de aangevulde DMEM zoals beschreven in het tekstprotocol. Een dag voor transductie was de cellen twee keer met 10 milliliter PBS.Voeg twee milliliter van 0,25% trypsine EDTA toe aan de cellen en broed gedurende drie minuten uit bij 37 graden Celsius. Met behulp van een 10-milliliter serologische pipet resuspend de cellen in vijf milliliter van cultuurmedium en pas de celdichtheid aan 50.000 cellen per milliliter. Voeg 400 microliter media die 20.000 cellen bevat toe aan elke put van een 24-put kweekplaat.Incubeer op 37 graden Celsius gedurende 24 uur. De volgende dag transduceren twee putten door het vervangen van de cultuur medium met 400 microliter van een een-op-een mengsel van een lentivirus en verse cultuur medium aangevuld met polybrene bij een uiteindelijke concentratie van vier microgram per milliliter. Voor de geen viruscontroles vervang het cultuurmiddel met vers cultuurmiddel dat met polybrene bij een definitieve concentratie van vier microgram per milliliter wordt aangevuld.Incubeer op 37 graden Celsius gedurende 15 uur. Dan aspirate het medium met lentivirussen, en verse cultuur medium. Incubeer de cellen op 37 graden Celsius voor een extra 72 uur.Daarna was je de cellen in elke put twee keer met 0,5 milliliter PBS. Voeg 300 microliter van 0,25% trypsine EDTA toe aan de cellen en broed gedurende drie minuten uit bij 37 graden Celsius. Resuspend de cellen in twee milliliter van cultuurmedium, en dan plaat ze in zes-goed cultuur platen met twee microgram per milliliter puromycine.Kweek de cellen tot alle cellen in de controleputten dood zijn, wat meestal ongeveer een week duurt. Ten eerste, afzonderlijk cultuur LN215-YFP en LN215-jodide cellen in 100-millimeter platen in cultuur medium om de populaties die nodig zijn voor de test te bereiken. Een dag voor de test, was elke plaat met 10 milliliter PBS.Behandel elke plaat met twee milliliter van 0,25% trypsine EDTA-oplossing en broed gedurende vijf minuten uit op 37 graden Celsius. Resuspend de cellen in vier milliliter van cultuurmedium en breng ze over naar een 15-milliliter conische buizen. Centrifuge op 1.000 g gedurende drie minuten om de cellen te pelleteren.Gooi de supernatant en resuspend elke cel pellet in vijf milliliter van cultuur medium. Pipette op en neer ongeveer 20 keer te breken een cel klonten. Gebruik een hemocytometer om de cellen te tellen.En verdun dan de cellen in kweekmedium om celsuspensie te maken bij de dichtheden die hier worden getoond. Meng vervolgens zeven milliliter van elke celsuspensie samen in een reservoir. Gebruik een multichannel pipet om 100 microliters van dit mengsel toe te voegen aan elke put van een 96-well cultuurplaat.Incubeer bij 37 graden Celsius met 5%kooldioxide gedurende 24 uur. Gebruik eerst een fluorescentiemicroscoop met 20x vergroting en een GFP-filter om de 96-put platen te controleren om er zeker van te zijn dat er geen klonten cellen zijn, en dat de culturen volledig samenvoezig en goed verdeeld zijn. Ten minste 30 minuten voordat de test zet een microplate lezer en zet het op 37 graden Celsius.Was de slang van een geautomatiseerde injector met drie milliliter van 70%ethanol, drie milliliter gedestilleerd water en drie milliliter I-oplossing. Verwarm vervolgens de C- en I-oplossingen tot 37 graden Celsius in een waterbad. Ofwel aspirate het groeimedium of omkeren van de plaat om het te legen.Tik op elk restmedium. Voeg vervolgens de C-oplossing toe aan een reservoir met behulp van een multichannel pipet om 200 microliters C-oplossing toe te voegen aan elke put van de plaat. Ofwel aspirate de oplossing of omkeren van de plaat om te legen, waardoor elke resterende oplossing tappen.Voeg 50 microliter C-oplossing toe aan elke put. Voeg vervolgens een microliter van een 2,5-millimolar chemische voorraad of DMSO als een voertuig, en voeg nog eens 50 microliter van C-oplossing aan elke put. Incubeer de cellen bij 37 graden Celsius in de lucht gedurende ongeveer 10 minuten.Begin tijdens de incubatie de parameters in het microplate-lezersprogramma in te stellen door op de knop Protocollen beheren te klikken. Klik op de knop Nieuw. Selecteer de fluorescentieintensiteit in de sectie Metingsmethode en Put-modus in de sectie Leesmodus.Klik vervolgens op de knop OK. Er verschijnt een nieuw tabblad. Stel in het menu Basisparameters de excitatiegolflengte in op 485 nanometer en de emissie op 520 nanometer.Selecteer de onderste optische om fluorescentie van onderen te lezen. Stel vervolgens de begintijd van de meting in op nul seconden, het aantal intervallen op 25, het aantal flitsen per put en interval op 20 en de intervaltijd op 0,4 seconden. Teken in het menu Indeling een gebied van de te lezen plaat.In het menu Concentraties/Volumes/Shacking stelt u de microplatelezer in om 100 microliter I-oplossing te injecteren voor elke put met een injectiesnelheid van 135 microliters per seconde. In het menu Injectietijd stelt u de begintijd van de injectie in op één seconde. Klik vervolgens op de knop Meting starten.Wanneer de incubatie is voltooid, breng je de 96-put platen over naar de microplate-lezer en start de meting door opnieuw op de metingknop Start te klikken. In deze studie wordt de jodiumgel fluorescerende eiwit-gap junction-intercellulaire communicatietest gebruikt met LN215-YFP en LN215-jodiumcellen om 2, 320 chemische stoffen te screenen om nieuwe gap junction intracellulaire communicatiemodulatoren te identificeren. Homosalaat wordt gezien als 50% van gap junction intracellulaire communicatie remmen.Terwijl terbinafine het volledig remt. Dit bevestigt terbinafine als een gap junction remmer. Deze test meet GJIC tussen naburige donor en acceptor cellen, zodat de donor en acceptor cellen volledig moeten worden gescheiden voordat plating.En de samenvloeiing van de gemengde cultuur moet 100% zijn om de vorming van kloofknooppunten tussen cellen te maximaliseren. Tijd cursus gegevens, dosis respons relaties, en de beoordeling van de omkeerbaarheid van gap junction modulators kunnen worden verkregen met Iodide-YFP-GJIC test. Ook Iodide-YFP specifieke connexin GJIC testen kunnen worden vastgesteld door het induceren van de expressie van een connexin van belang in de cellen verstoken van functionele kloof knooppunten.Dit maakt het mogelijk om de IC50-waarde van een chemische stof van een specifiek type connexin te bepalen. Iodide-YFP-GJIC-test kan worden gebruikt voor screening met hoge doorvoer, omdat het robuust, herhaalbaar en goedkoop is. Deze tests helpt bij het vinden van gap junction modulerende chemische stoffen voor drugs ontdekking en toxicologische beoordeling.
