De relatief korte hoeveelheid tijd voor isolatie, infectie en primaire acinaire cellen plating is een voordeel van deze methode. Daarentegen weinig hands-on tijd kweken acinaire cellen van een uitvloeisel explant vereist, maar het duurt zeven dagen voor de uitgroei van acinaire cellen13. Acinaire-naar-ductaal metaplasie (ADM) is een beschermend mechanisme tijdens pancreatitis en een belangrijke proces rijden van pancreatic kanker ontwikkeling1 waarvoor verdere mechanistische inzicht. Ontsteking-geïnduceerde ADM weliswaar omkeerbare2, oncogene KRAS mutaties, die in 90% van pancreatic kanker gevallen3aanwezig zijn, te voorkomen dat differentiatie terug naar een acinaire fenotype4,5, 6. Culturing en differentiatie van primaire acinaire cellen in ductaal cellen in 3D cultuur zorgt voor de studie van de moleculaire mechanismen tot regeling van het ADM-proces, dat is moeilijk om te studeren in vivo. Dergelijke ex vivo studies inschakelen visualisatie in real time van ADM, de bestuurders en de regelgevers. Diverse stuurprogramma's van de ADM-proces, evenals de mechanistische inzicht op hun downstream signaalroutes, zijn geïdentificeerd of geverifieerd met behulp van de hier beschreven methode. Deze omvatten ADM inductie door TGF-α (alpha-transformatie groeifactor)-gemedieerde uitdrukking van MMP-7 (matrix metalloproteinase-7) en activering van Inkeping7, evenals RANTES (geregeld op activering, normale T cel uitgedrukt en uitgescheiden; ook bekend Als chemokine ligand 5 of CCL5) en TNFα (tumor necrose factor Alfa)-geïnduceerde ADM via activering van NF-recombination (nucleaire factor-recombination)8. Een extra bemiddelaar van ADM is oncogene KRas9,10, waardoor een stijging van de oxidatieve stress die vergroot het ADM-proces door middel van de verhoogde expressie van EGFR (epidermale groeifactor receptor) en de liganden, EGF) epidermale groeifactor) en TGF-α11.
Terwijl het gebruik van pancreatic kanker cellijnen is gebruikelijk voor in vitro studies, bieden primaire celculturen vele voordelen. Bijvoorbeeld, primaire acinaire cellen van niet-transgene muizen of muizen herbergen initiatiefnemende mutaties, zoals KrasG12D, zijn het meest geschikte model voor het bestuderen van de vroege gebeurtenis van ADM, omdat de cellen enkele en gecontroleerde mutaties hebben, die bekend is dat ze aanwezig zijn in het begin van pancreatic kankerontwikkeling. Dit is in tegenstelling tot vele alvleesklierkanker cellijnen die meerdere mutaties hebben en gevarieerd expressie gebaseerd op passage nummer12. Bovendien zijn de signaalroutes geïdentificeerd door daartoe geverifieerd door dierproeven. Een waarschuwing van het gebruik van primaire acinaire cellen is dat ze niet kunnen zijn transfected als typische kanker cellijnen kunnen.
De methode hierin een overzicht van de technieken van acinaire cel isolatie, 3D cultuur die ADM bootst door het toevoegen van stimulerende middelen of modulerende eiwit expressie via adenovirale of lentivirale infectie, evenals opnieuw isolatie van cellen op het eindpunt voor verdere analyses.

mimicking and manipulating pancreatic acinar-to-ductal metaplasia in 3-dimensional cell culture

De differentiatie van acinaire cellen naar ductaal cellen tijdens pancreatitis en in de vroege ontwikkeling van pancreatic kanker is een belangrijke proces dat verdere studie vereist. Om te begrijpen van de mechanismen biedt reguleren acinaire-naar-ductaal metaplasie (ADM), ex vivo 3D cultuur en differentiatie van primaire acinaire cellen naar ductaal cellen vele voordelen ten opzichte van andere systemen. Met de techniek hierin is modulatie van eiwit expressie eenvoudig en snel, waarbij slechts één dag te isoleren, stimuleren of viraal besmetten, en beginnen met het kweken van primaire acinaire cellen om te onderzoeken het ADM-proces. In tegenstelling tot via kelder membraan matrix, het zaaien van acinaire cellen clusters in collageen ik extracellulaire matrix, kunt acinaire cellen te behouden hun acinaire identiteit voor manipulatie. Dit is cruciaal bij het testen van de bijdrage van de verschillende onderdelen aan de inductie van ADM. Niet alleen zijn de gevolgen van cytokines of andere ectopically door de overheid gereguleerde factoren testbare via deze techniek, maar de bijdrage van de gemeenschappelijke mutaties, verhoogde eiwit expressie of knockdown van eiwit expressie is testbare via virale infectie van primaire acinaire cellen, met behulp van adenovirale of lentivirale vectoren. Bovendien cellen kunnen worden opnieuw afgezonderd van collageen of kelder membraan matrix op het eindpunt en geanalyseerd voor eiwit expressie.

Voltooiing van het protocol hierin treedt op binnen één dag en na stimulatie, ADM wordt gezien in 3-5 dagen. Figuur 1 toont de volgorde van de methode, waarbij stappen 1 t/m 4 zijn voltooid op de eerste dag. Dit omvat de voorbereiding, de acinaire isolatie, de virale infectie en de verankering in collageen of kelder membraan matrix. Ons protocol biedt directe visualisatie van Acinar-to-Ductal Metaplasie en maakt het mogelijk de bijdrage van ectopisch toegediende factoren of eiwitmodulatie aan dit proces te bestuderen. Het belangrijkste voordeel van deze techniek is dat Acinar-to-Ductal Metaplasie in real time kan worden bekeken en celsignaleringsstudies kunnen worden uitgevoerd. Om de alvleesklier te ontleden van een geëuthanaseerde muis, eerst de poten van de muis vastpinnen aan het polystyreendeksel, het zo te oriënteren zodat de staart de onderzoeker onder ogen ziet en de buik met 70% ethanol spuit.Gebruik autoclaved tangen om de vacht en de huid op te heffen op de middellijn, en met autoclaved schaar, maak een incisie door de vacht en de huid van de urethrale opening naar het middenrif ribbenkast gebied. Maak extra insnijdingen naar links en rechts om de vacht en huid weg te snijden, waardoor een duidelijk zicht op de buikholte ontstaat. Snijd vervolgens in de buikvlies, in het midden en naar rechts en links en trek het weg van de organen.Gebruik de tangen om de darmen op te tillen en naar de linkerkant van de muis te verplaatsen, waardoor er ruimte ontstaat om de lichtroze alvleesklier aan de donkere, rode, ovale milt te zien. Het alvleesklierweefsel zal een zachte, sponsachtige textuur hebben. Knip de alvleesklier, die loopt langs de maag, verweven met de darmen.Scheid de milt van de alvleesklier en plaats de alvleesklier in een buis van 50 milliliter met 10 milliliter HBSS, met één keer penicilline streptomycine en breng deze over naar de laminaire stroomkap. Giet de inhoud van de buis in een lege weegboot en gebruik tangen om de alvleesklier te wassen door het te wervelen. Gebruik vervolgens de tangen om de alvleesklier naar een tweede weegboot te verplaatsen met HBSS met penicilline streptomycine en opnieuw te wassen door te wervelen.Verplaats de alvleesklier naar de derde weegboot met HBSS met penicilline streptomycine en begin met het snijden van de alvleesklier in vijf millimeter of kleinere stukken. Om de inhoud van de weegboot in een verse buis van 50 milliliter te gieten, gebruik eerst de tangen om de alvleesklierstukken in de vloeistof te bewegen aangezien de boot wordt getipt. Zodra de stukken niet meer aan de boot zijn bevestigd, giet de inhoud ervan in de buis.Pak de resterende stukken met de tangen en was de tang in de buis. Na het centrifugeren van de buis op 931 keer de zwaartekracht op vier graden Celsius gedurende twee minuten, gebruik maken van een vijf milliliter pipet om de HBSS en alle drijvende vet te verwijderen. Voeg vijf milliliter collagenase verdund in HBSS.Sluit de buis en zorg voor een gesloten deksel door de buis in plastic paraffinefilm te wikkelen. Plaats het in een couveuse, schudden op 220 RPM op 37 graden Celsius gedurende 20 minuten. Stel de pellet opnieuw op in 10 milliliter HBSS met 5%FBS, en centrifuge opnieuw bij 931 keer zwaartekracht gedurende twee minuten bij vier graden Celsius.Verwijder de supernatant met behulp van een vijf milliliter pipet en herhaal deze stap met nog eens 10 milliliter HBSS met 5%FBS. Dan opnieuw opschorten de pellet in vijf milliliter hbss met 5%FBS. Gebruik een P1000 pipet om de celsuspensie één milliliter per keer te filteren door een gaas van 500 micrometer in een buis van 50 milliliter.Om de resterende alvleeskliercellen door het gaas te wassen, voegt u nog eens vijf milliliter HBSS toe met 5%FBS. Gebruik dan een P1000 pipet om dit filtraat een milliliter per keer door de 105 micrometer mesh in een 50 milliliter buis. Voorzichtig pipette deze cel suspensie in een 50 milliliter buis met HBSS met 30%FBS, die een laag van cel suspensie zal vormen aan de bovenkant.Centrifuge deze cel suspensie bij 233 keer zwaartekracht bij vier graden Celsius gedurende twee minuten en verwijder de supernatant. Schort de pellet met de Acinar-cellen in één keer het complete medium van Waymouth op. Aangezien een alvleesklier voldoende is voor twee virale infecties, split de cel suspensie tussen de deksels van twee 35 millimeter platen, die het mogelijk maakt voor infectie in suspensie.Voor adenovirale infectie, voeg het virus bij een 1/1000 verdunning aan het deksel van elke plaat en werveling. Plaats de platen in een celkweek incubator op 37 graden Celsius en 5%CO2. Draai het eerste uur elke 15 minuten en zet de incubatie nog twee uur voort.Om een collageen gel te maken, combineer zeven milliliter van type een rat staart collageen, 700 microliter van 10 keer Waymouth's medium en 466,6 microliter van 0,34 molaire natriumhydroxide in een 50 milliliter buis op ijs om collageen gel te maken. Combineer in een buis van 50 milliliter na drie uur incubatie gelijke volumes celsuspensie en collageengel en meng voorzichtig. Plaat 2000 microliters, een goed per keer in een zes put plaat terwijl het houden van de plaat op ijs en wervelen om gelijkmatig te verdelen de collageencel laag.Om het medium te stollen, de plaat in een celkweek incubator in 37 graden Celsius en 5%Kooldioxide gedurende 30 minuten uitbroeden. Voeg vervolgens 1,5 milliliter per put van een keer waymouth's complete medium met de gewenste stimulus of remmers. Verander het medium de volgende dag en vervolgens om de andere dag.Afhankelijk van de gebruikte stimulus kan Acinar-to-Ductal Metaplasie of ADM worden waargenomen tussen dag drie en dag vijf. 24 uur na infectie met GFP adenovirus, beelden van cellen ingebed in collageen of kelder membraan matrix, bleek dat infectie efficiënt was. Op vijf dagen na infectie, cellen ingebed in collageen gevormd kanaal-achtige structuren wanneer gestimuleerd met 50 nanogram per milliliter van TGF alpha.Cellen ingebed in kelder membraan matrix gevormd kanaal-achtige structuren in de afwezigheid van een stimulus op vijf dagen na infectie. Terwijl grotere kanalen werden gevormd bij stimulatie met 50 nanogram per milliliter TGF alfa. Een van de meest kritieke punten in het protocol is de lengte van collagenase spijsvertering.Een goede spijsvertering wordt aangegeven door een troebele oplossing en enkele resterende weefselstukken.
