بالإضافة إلى الجينات التي تشارك مباشرة في إنتاج ميكوتوكسين كأهداف محتملة من استراتيجيات مكافحة المحورة وراثيا، أظهرت amylases الفطرية تلعب دوراً حاسما في الحفاظ على النجاح في إنتاج المرضية وميكوتوكسين خلال المراحل المبكرة الإصابة بالنبات المضيف. عدد قليل من الأمثلة على بيثيوم بليروتيكوم (عامل السببية تعفن رهيزومي الزنجبيل)، سولاني الفوزاريوم (وكيل المسببة الذبول قرنبيط)، حيث لوحظ الارتباط الإيجابي بين الإمراضية و α-الأميليز التعبير والنشاط 7،8. تثبيط النشاط α-الأميليز أما عن طريق خروج المغلوب الجينات أو النهج ضربة قاضية يؤثر تأثيراً سلبيا على إنتاج النمو والسمية الفطرية. وكان المسخ خروج المغلوب α-الأميليز من الخنزير (أ) غير قادر على إنتاج الأفلاتوكسين عندما تزرع في النشا الركيزة أو ديجيرميد حبات الذرة9. وبالمثل، في فيرتيسيليويديس مغزلاويه فشل عبئا خروج المغلوب α-الأميليز لإنتاج الفيومونيزين ب1 (ميكوتوكسين) خلال العدوى من حبات الذرة10. في دراسة حديثة، أظهر جيلبير et al. (2018) أن تدق على أساس [رني] أسفل الخنزير ألف- α-الأميليز التعبير من خلال هيجس انخفاضا ملحوظا الخنزير (أ) النمو والافلاتوكسين الإنتاج خلال العدوى نواة الذرة11 .
مثبطات محددة من النشاط α-الأميليز قد أسفرت أيضا عن نتائج مماثلة كما تم الحصول عليها من أسفل-تنظيم التعبير α-الأميليز. وجاء التقرير الأول بشأن دور المانع α-الأميليز في مقاومة الفطريات من عزل وتوصيف مثبط التربسين-α-الأميليز كاتشين 14 من خطوط الذرة مقاومة الخنزير (أ)12. كذلك فحص عدة مئات من الأنواع النباتية فاخوري ووولوشوك أدت إلى تحديد بروتين مثبط مثل α-الأميليز كاتشين 36 (أيلب) من بذور الفول صفير، لابلاب purpureus ل13. وأفادت تسلسل الببتيد ليكتينس أيلب تشبه لعائلة المانع يكتين – أرسلان – α-الأميليز المشتركة فول14،15. أيلب المنقي لا يحمل أي نشاط المثبطة نحو التربسين الثدييات وكذلك توصيف في المختبر أظهرت تثبيط كبيرة الخنزير (أ) النمو والإنبات كونديل13. التقارير المقدمة هنا وضوح يظهر α-الأميليز يمكن أن تخدم كهدف في نهج الرقابة للحد من مسببات الأمراض أو الآفات التي تعتمد على تعبئة النشا (من خلال نشاط α-الأميليز) واقتناء السكريات القابلة للذوبان كمصدر للطاقة خلال هذه الممرضة التفاعل مع النباتات المضيفة.
الأميليز ألفا ومن المعروف أن تكون حاسمة في الخنزير (أ) القدرة الإمراضية9،10،11، ونظرا لأهمية أيلب بوصفها ' مكافحة قوية-الخنزير ألف عامل (α-الأميليز تثبيط/لﻹنصاف)13، نحن تولد النباتات الذرة المعدلة وراثيا التي تعرب عن لابلاب أيلب الجينات تحت المروج كامب 35S التأسيسي. وكان الهدف التحقيق إذا كان تعبير مغايرة لهذا المانع α-الأميليز في الذرة فعالة ضد الخنزير (أ) إنتاج المرضية والافلاتوكسين أثناء الإصابة بنواة الذرة. نتائجنا تثبت أن حبات الذرة المعدلة وراثيا معربا عن أيلب إلى حد كبير انخفاض إنتاج الخنزير (أ) النمو والافلاتوكسين أثناء الإصابة بالنواة.

(أيلب) في الذرة ضد الخنزير (أ). أيلب هو بروتين 36-كاتشين، الذي هو مثبط تنافسي لأنزيم الأميليز α الخنزير (أ) ، وينتمي إلى أسرة بروتين مثبط يكتين – أرسلان – α-الأميليز المشتركة فول. وقد أثبتت الدراسات في المختبر قبل العمل الحالي دور أيلب في تثبيط النشاط α-الأميليز الخنزير (أ) ونمو الفطريات. وتم رصد نمو الفطريات وإنتاج الأفلاتوكسين في حبات ناضجة في الوقت الحقيقي باستخدام سلالة الإعراب عن بروتينات فلورية خضراء الخنزير (أ) . بسيط جداً لإعداد نواة هذا الفحص المقايسة (المملكة العربية السعودية) ويوفر بيانات موثوقة واستنساخه في الإصابة ومدى انتشار التي يمكن قياسها كمياً لتقييم الأصول الوراثية وخطوط محوره وراثيا. الأسفار من إجهاد بروتينات فلورية خضراء النمو ارتباطاً وثيقا للفطريات، واستطراداً، أنها مرتبطة جيدا لقيم الأفلاتوكسين.  وكان هدف العمل الحالي لتطبيق هذه المعرفة السابقة في محصول تجارياً هاما مثل الذرة لزيادة مقاومة الأفلاتوكسين. نتائجنا تشير إلى انخفاض 35%-72% في نمو الخنزير (أ) في حبات الذرة المعدلة وراثيا معربا عن أيلب التي، بدورها، ترجمت إلى 62%-88% انخفاضا في مستويات الأفلاتوكسين.

هنا نقدم فحص نواة بسيطة فحص تحليل Aspergillus flavus النمو وإنتاج الأفلاتوشين في حبات الذرة التي تعبر عن البروتين المضاد للفطريات. باستخدام بروتين الفلورسنت الأخضر التعبير عن سلالة الفلافات Aspergillus، ونحن رصد العدوى وانتشار الفطريات في حبات ناضجة في الوقت الحقيقي. هذا الفحص البسيط يوفر وسيلة موثوقة لتقييم قدرة نواة الذرة على مقاومة عدوى الزلاجة و إنتاج الأفلاتوشين.نتائج هذا المختبر فحصاً التي أجريت في ظل ظروف خاضعة للرقابة على حبات ناضجة يرتبط جيدا مع نتائج العدوى في ظل ظروف الميدان. ومن خلال هذا الإجراء، سيكون الدكتور راجتيلاك ماجومدار، وهو من مختبرنا، جنبا إلى جنب مع كريستين سيكلر وديفيد أمبروسيو، فنيي العلوم البيولوجية أيضا من مختبرنا. للبدء، الغراء 20 اثنين من قبعات المفاجئة مم في طبق بيتري لإنشاء قبعات المملكة العربية السعودية.دع الغراء يجف لمدة 48 ساعة قبل استخدام القبعات. في مجلس الوزراء السلامة البيولوجية رش علبة bioassay مربع مع 70٪ الإيثانول في الماء. دع صينية الهواء تجف.أضف ورق كروماتوغرافيا معقمة إلى صينية جافة. رش تسعة من القبعات السعودية مع 70٪ الإيثانول والسماح لهم الهواء الجاف. ثم ضع القبعات على علبة اِيُوائي.لكل خط ذرة معدلة وراثيا يجري اختبارها، حدد 20 حبة غير التالفة ووضعها في أنبوب الطرد المركزي 50 مل. باستخدام المجهر ستيريو الطاولة حدد فقط حبات غير التالفة للمجايسة. حبات التالفة هي أكثر عرضة لهجوم Aspergillus وإنتاج المزيد من الأفلاتوشين، وبالتالي انحراف النتائج وليس يمثل الصفات المضادة للفطريات أو مضادات الميكروبات الحقيقية من مجموعة متنوعة.إضافة 70٪ الإيثانول إلى كل أنبوب. دع الحبوب تعقيم لمدة أربع دقائق بينما تهزها بلطف في بعض الأحيان. بعد هذا بلطف صب قبالة الإيثانول وشطف بلطف حبات في الماء غير المؤين العقيمة ثلاث مرات.الحصول على ثقافة ستة أيام من وصمة عار 70 اسبرجيلوس flavus التعبير عن GFP نمت على V8 المتوسطة. إضافة 20 مل من العقيمة 0.02٪ تريتون X-100 في المياه ديوند. واستخدم حلقة معقمة لتكشط من الجراثيم.الماصات قبالة inoculum ونقلها إلى 300 مل منقار معقمة. إعداد تخفيف خمسة أضعاف مع 0.02٪ تريتون X-100 واستخدام مقياس الهيموسيت لإجراء العد بوغ. تخفيف مرة أخرى إذا لزم الأمر للحصول على 100 مل مع تركيز أربعة ملايين جراثيم لكل مل.صب قبالة inoculum في القارص فارغة. ضع حبات في منقار معقمة 300 مل مع شريط ضجة ويحرك لمدة ثلاث دقائق. باستخدام ملقط نقل حبات إلى طبق bioassay، مع التأكد من وضع نواة واحدة فقط في كل غطاء.أضف 30 مل من الماء غير المؤين إلى الجزء السفلي من كل صينية من علب البيوراسا. واحتضان في 31 درجة مئوية لمدة سبعة أيام. بعد سبعة أيام من التلقيح مع A.flavus، التقاط صور من أربع حبات التي تم تعيينها للتحليل المجهري والتصوير الفوتوغرافي.ثم استخدام الأنسجة الرخوة والمياه ديوند لتنظيف الخارج من حبات. قم بأداء أقسام طولية من حبات التقط الصور مباشرة تحت المجهر الفلوري. لتحضير حبات للتحليل، قم بتنظيف السطح الخارجي من حبات المتبقية بنسيج ناعم وماء غير مأون.مكان أربع حبات، والتي تشكل مندوب واحد، في 15 مل المسمار قبعة البولي قارورة تحتوي على اثنين من كرات الفولاذ المقاوم للصدأ. تجميد على الفور قارورة في النيتروجين السائل وتخزينها في 80 درجة مئوية حتى تصبح جاهزة للمضي قدما في مزيد من المعالجة والتحليل. عندما تكون جاهزة، وإزالة حبات من الفريزر واستخدام التجانس لطحن لهم في 1500 دورة في الدقيقة لمدة ثلاث دقائق.استخدم نفس المواد الأرضية لتحليل GFP وتحديد الأفلاتوشين والتحليل الجزيئي، بحيث تكون جميع القيم ممثلة للعينات. في حين ينصح على الأقل من ثلاثة تكرارات، أكثر هو دائما أفضل اعتمادا على توافر حبات ذات نوعية جيدة. في هذه الدراسة يتم تحويل الأجنة غير ناضجة من خطوط الذرة HI-II باستخدام Agrobacterium tumefaciens EHA101 سلالة تحتوي على ناقلات المقصد النباتية النهائية التي تعبر عن الجين Lablab purpureus AILP تحت سيطرة المروج 35S من فيروس فسيفساء القرنبيط.أظهر تضخيم PCR لجين AILP المستهدف وجود 548 bp amplicon لوحظ فقط في خطوط الذرة المعدلة وراثيا. أظهر الجين AILP التعبير في خطوط المعدلة وراثيا في حين لم يلاحظ أي تعبير AILP في محطات التحكم. تتعرض الجراثيم في مرحلة مبكرة من الزرع إلى مستخلصات أوراق خام من نباتات الذرة المعدلة وراثياً الصغيرة لمدة 20 ساعة.ارتفاع طول كامل من الجراثيم المعرضة لاستخراج أوراق معدلة وراثيا تظهر 58 إلى 80٪ انخفاض مقارنة مع السيطرة السلبية. سلالة A.flavus المستخدمة تحتوي على مراسل GFP الذي يتيح رصد وكمية النمو الفطري في الوقت الحقيقي. ويلاحظ انخفاض كبير في الفلورية GFP في حبات AILP المعدلة مقارنة بالتحكم السلبي الأيزوجيني.ويُنظر إلى انخفاض كبير في النمو الفطري لخطوط AILP أربعة وخمسة و69٪ و72٪ على التوالي. خطوط أخرى تظهر انخفاضا من 35 إلى 60٪ في مقارنة مع حبات التحكم. ولوحظ انخفاض بنسبة 62 إلى 88٪ في محتوى الأفلاتوشين في حبات AILP مقارنة بالتحكم.ويُنظر إلى الخط الرابع على أنه أدنى محتوى من أفلاتوشين عند 486 نانوغرام لكل غرام، يليه خطوط أخرى تتراوح بين 640 و1498 نانوغرام لكل غرام في حبات. بعد هذا الإجراء، يمكن تنفيذ طرق إضافية مثل استخدام مقياس الفلور لتحديد كمية GFP ونموه الفطري. يمكن استخدام مجموعات تجارية تعتمد على المناعة أو HPLC أو EPLC لإجراء كمية الأفلاتوشين.يوفر توافر المملكة العربية السعودية وسيلة سريعة وبسيطة لتحديد مقاومة التلوث أفلاتوشين. ويمكن تقييم خطوط تربية الذرة المختلفة بسرعة باستخدام هذه التقنية. في حين أن فحص النواة ليس خطرا، يجب على المشغل حماية نفسه من التعرض لأسبيرجيلوس الجراثيم flavus، واستخدام معدات الحماية المناسبة.
