この標準化プロトコルのセットは、優れた眼のサルカス、またはSOS、発達中の目の最近発見された構造に新しい洞察を提供するために使用することができます。これらのプロトコルは、世界中の実験者がゼブラフィッシュの目の開発中にSOSを明確に視覚化することを可能にします。ヘテロ接合性成長分化因子6 Aオスとメスのゼブラフィッシュを、ペアリングの前の晩にデフラリン水のタンクの仕切りの別々の側面に置くことから始めます。翌朝、仕切りを取り除き、魚が30分以下繁殖できるようにします。繁殖期間の終わりに、28.5度のインキュベーターでインキュベートするためにE3培地を含むペトリ皿に胚を集める。受精後20時間で上清を0.004%1-フェニル-2-チオ尿を補充したE3培地に置き換え、色素産生を防ぎ、光顕微鏡で胚を調べ、それらがすべて適切な発達段階にあることを確認する。サンプルを解剖顕微鏡の下に置きます。発達期の未熟な胚を捨て、細かい鉗子を使って穏やかに結び目を引き離します。目の裏の余白にインデントまたは線として表示される SOS を視覚化します。次に、SOS閉鎖遅延を示す胚をそうでない胚から並べ替えます。解剖顕微鏡を用いてこれらの胚を撮影するには、E3培地に1%アガロースを含むペトリ皿を準備し、アガロースの中心を軽く刺して、胚の黄身を置くことができる浅い穴を作ります。次いで、麻酔した胚をアガロースの上に横に置く。化合物または共焦点顕微鏡を使用して胚を画像化するには、ゲル化されていない1%低融点アガロースの小さなボーラスを含む35ミリメートルのペトリ皿に胚をE3培地に移し、細かい釣り糸を使用して胚を横位置に素早く配置する。アガロースが冷却されたら、アガロースをカバーするのに十分なE3培地を皿に注ぎ、SOSを視覚化するために20倍の水浸し対物レンズを使用します。イメージングの後、胚からアガロースをそっと引っ張り、4%パラホルムアルデヒドで組織を固定するか、ゼブラフィッシュがその発達を続けることを可能にする。蛍光mRNA発現によりSOSを可視化するために、マイクロインジェクション装置を用いて、1細胞段階で300個の強化されたGFP-CAX mRNAを胚に注入する。次に、蛍光ステレオスコープを使用して、明るい強化されたGFP発現を有する胚をスクリーニングし、実証された画像化方法の1つによって胚の画像を得る。胚性ゼブラフィッシュラミニンの全実装免疫蛍光染色の場合、デモンストレーションされたように胚をデコールニオンさせ、室温シェーカーで2時間調製したばかりの4%パラホルムアルデヒドでマイクロ遠心分離チューブに胚を固定する。固定の終わりに、PBSプラストゥイーン(PBST)で胚を洗浄ごとに5分間洗浄し、1回で1ミリリットル当たり10マイクログラムのパーメアビレーション、室温でプロテイナーゼKを5分間洗浄します。ペルメアナイズされた胚をPBSTで4回洗浄し、室温シェーカーでPBSTの5%ヤギ血清および1ミリリットル当たり2ミリグラムのウシ血清アルブミンをブロックする。ブロッキングインキュベーションの終わりに、一晩の震える上で摂氏4度で目的の一次抗ラミニン抗体の胚をインキュベートする。翌朝、PBSTで胚を1回15分間5回洗浄し、光から保護されたシェーカーの摂氏4度で一晩適切な二次抗体で組織にラベルを付けます。翌朝、PBSTで1回15分間胚を4回洗い、新鮮なPBSTを含む小さなペトリ皿に胚を入れます。細かい鉗子を使用して、卵黄を穏やかに破壊し、黄身嚢の軽度の擦り傷を通して黄身細胞を取り除く。脱ヨルク胚をPBSで新たに調製した30%グリセロールに移し、胚を溶液の上に置き、以前の溶液をできるだけ少なく移し、胚がチューブの底に沈むのを待ちます。胚が容器の底に達したら、ちょうど示したように、PBS溶液中の逐次50%および70%のグリセロールに移します。胚が70%グリセロール容器の底に沈んだ後、サンプルを小さなプラスチック皿に移して分離し、後部の胚を後脳に切断する。細かい解剖ツールを使用して、できるだけ少ないグリセロールでガラススライドに1つの頭を移動し、内側から前脳心室に細かいミヌティエンピンをそっと挿入し、頭の右半分と左半分を互いに分離するために下に押します。ヘッドが完全に正中線に充填されたら、ヘッドの各側を正中線下に、目側を上に取り付けます。SOSは、受精後20〜23時間の解剖顕微鏡を通して、そして発達中の残り線の鼻と側頭の半分を分離する背側眼の細い線として差動干渉コントラストイメージングを介して観察可能である。さらに、目の裏の境界に微妙なインデントが見える場合があります。ラミニンの免疫蛍光染色に続いて、細線を原膜として確認することができる。SOS閉鎖が遅れると、その長期の存在は、解剖顕微鏡の下で眼の側側の顕著な裂け目として観察することができる。この特徴は、微分干渉コントラストイメージングを用いて複合顕微鏡で観察した場合、SOSによって分離された眼の鼻側と側頭側と、後眼の線としてはっきりと見える場合に、さらに顕著である。野生型ゼブラフィッシュの受精後28時間までに、ラミニン染色はSOSが完全に閉鎖されていることを明確に示し、裂け目閉鎖の遅延を監視するための理想的な段階です。強化されたGFP-CAX mRNAの注入はSOS閉鎖につながる細胞の形態の変化を追跡するために生きているかまたは固定胚の細胞膜の視覚化を可能にする。SOS の視覚化は狭く一時的であるため、難しい場合があります。ただし、後で終了の遅延を評価する場合は、一貫性のあるスコアリング方法を維持することが不可欠です。これらの技術は、適切なSOSの形成および閉鎖に影響を与える遺伝的要因または薬理学的薬剤を同定するための実験的操作と組み合わせることができる。
