Welches hat die mögliche Anwendung für die Bewertung der klinischen Wirksamkeit von CAR T Zellprodukten. Beginnen Sie mit der Dissoziierung von Glioblastom-Tumorsphären aus einer Glioblastom-Tumorsphärenkultur. Mit einem Milliliter Kaltaccutase und 30 bis 60 Sekunden Pipettierung.
Wenn die Tumorsphären gestört sind, stoppen Sie die Reaktion mit fünf Milliliter warmes Co-Kulturmedium. Und pellet die Tumorzellsuspensionen durch Zentrifugation. Setzen Sie die Glioblastomzellen auf 1,6 mal 10 bis die fünfte Tumorzelle pro Milliliter frischer Cokulturmittelkonzentration aus und verdünnt T-Zellen, die aus einer CAR-T-Zellkultur geerntet werden, auf den entsprechenden Prozentsatz der CAR-positiven T-Zellen pro Milliliter Co-Kultur-Mittlerer Konzentration.
Als nächstes fügen Sie 100 Mikroliter der verdünnten Tumorzellen zu jedem Brunnen einer 96 gut flachen Bodengewebekulturplatte hinzu. Und 100 Mikroliter CAR-T-Zellen in jeden Brunnen von Tumorzellen mit sanfter Mischung. Dann legen Sie die Platte in einem 37 Grad Celsius, 5% Kohlendioxid-Inkubator.
An den Tagen zwei, vier und sechs der Kultur, sorgfältig entfernen 50 Mikroliter Medium aus jedem Brunnen der Tumorzelle, T-Zelle Co-Kultur-Platte für rechallenge nach der Tabelle geplant. Wenn CD4-positive und CD8-positive CAR-T-Zellen im Verhältnis eins zu eins gemischt werden, führt diese Zellkombination CD8-positive, aber nicht CD4-positive CAR-T-Zellen in Bezug auf die langfristige Zytotoxizität aus. Darüber hinaus wird die Expansion von CD8-positiven CAR-T-Zellen durch die Zugabe von CD4-positiven T-Zellen induziert.
Während CD4 positive CAR T Zellexpansion in Gegenwart von CDa-positiven Zellen gehemmt wird. 24 Stunden nach der anfänglichen Co-Kultur werden sowohl CD4-positive als auch CD8-positive CAR-T-Zellen in ähnlicher Weise aktiviert. CD8-positive CAR-T-Zellen sind auch anfälliger für Erschöpfung im Vergleich zu CD4-positiven CAR-T-Zellen, die nach drei Tagen Co-Kultur nach ihrer PD1-, LAG3- und TIM3-Expression beurteilt werden. Weiterhin wurden keine Unterschiede bei der CDa-positiven Car-T-Zellerschöpfung in Gegenwart oder Abwesenheit von CD4-positiven T-Zellen beobachtet, was darauf hindeutet, dass CD4-induzierte CD8-positive CAR-T-Zellexpansion nicht mit einer besseren Effektorfunktion verbunden ist. Dieser Aufsatz kann auch mit der Sortierung der T-Zellen gekoppelt werden, die die Kokultur bilden, um weitere Analysen zu Transkriptomen, Proteomik und spezifischen Genen von Interesse durchzuführen.
Diese sich wiederholende Tumorherausforderung könnte auch als Plattform für die Untersuchung der biologischen Ereignisse nach der Erkennung von CAR T-Zelltumoren genutzt werden.
