Thee is een van de meest geconsumeerde non-alcoholische dranken in de wereld1,2. Thee wordt geproduceerd uit de bladeren en knoppen van de Woody meerjarige Camellia sinensis L. thee planten worden geteeld in grote plantages en zijn vatbaar voor tal van insectenplagen3,4. Organophosphorus en neonicotinoid insecticiden worden vaak gebruikt als systemische insecticiden5 om thee planten te beschermen tegen plagen zoals whiteflies, Leaf hoppers, en sommige Lepidoptera soorten6,7. Na toepassing, worden deze insecticiden geabsorbeerd of in de installatie verplaatst. Binnen de plant kunnen deze systemische insecticiden worden getransformeerd door middel van hydrolyse, oxidatie of reductiereacties door plantaardige enzymen. Deze transformatie producten kunnen meer polaire en minder giftig dan de bovenliggende verbindingen. Echter, voor sommige organofosfaten, de bioactiviteiten van sommige producten zijn hoger. Bijvoorbeeld, wordt acefaat omgezet in giftigere methamidofos8,9, en dimethoaat in omethoaat,10,11. Plant aardige weefselculturen zijn bewezen een nuttig platform voor het onderzoek van het metabolisme van pesticiden, met de geïdentificeerde metabolieten vergelijkbaar met die gevonden in intact planten13,14,15. Het gebruik van weefselculturen, in het bijzonder de cellen schorsing culturen, heeft een aantal voordelen. Ten eerste, experimenten kunnen worden uitgevoerd vrij van micro-organismen, waardoor de interferentie van pesticide transformatie of degradatie door microben te voorkomen. Ten tweede, weefselcultuur biedt consistente materialen voor gebruik op elk gewenst moment. Ten derde, de metabolieten zijn gemakkelijker te halen uit weefselculturen dan van intacte planten, en weefselculturen hebben vaak minder interring verbindingen en lagere complexiteit van verbindingen. Een platform voor het bestuderen van insecticide metabolisme met behulp van in vitro weefsels van theeplant werd ontwikkeld. De bladeren van steriele thee plantjes werden veroorzaakt om losse aanroepus op Murashige en Skoog (Mej.) basismedia met de installatie hormonen te vormen 2, 4-dichlorophenoxyacetic zuur (2, 4-D, 1,0 mg L-1) en KINETIN (KT, 0,1 mg l-1). Aanroepus vormde zich na 3 of 4 rondes van subculturing, elk die 28 dagen duurt. Losse Callus (ongeveer 3 g) werd vervolgens geënt in B5 vloeibare media met dezelfde plantenhormonen en werd gekweekt in een schudden incubator (120 rpm) in het donker bij 25 ± 1 ° c. In behandelde Tea Cell culturen, zeven metabolieten van thiamethoxan en twee metabolieten van dimethoaat werden gevonden, terwijl in behandelde intacte planten, slechts twee metabolieten van thiamethoxam en een van dimethoaat werden gevonden. Het gebruik van een cel schorsing vereenvoudigde de metabole analyse in vergelijking met het gebruik van intacte thee planten, vooral voor een moeilijke matrix zoals thee.

Thee is een van de meest populaire drankjes over de hele wereld. Pesticiden worden soms gebruikt om theeplanten te beschermen tegen ongedierte. Sommige systemische pesticiden kunnen worden gemaakt voor dit door enzymen in theeboom.En vervolgens geïntegreerd door oxidatie, hydrolyse, of vermindering reacties. Plant cel suspensie culturen kunnen worden gebruikt als een, gemakkelijk om plant metabolisme gedrag van pesticiden te bestuderen. Deze methode heeft verschillende voordelen.Ten eerste kan het worden bediend op omstandigheden van vrije micro-organismen, waardoor de interferentie van de afbraak van pesticiden veroorzaakt door microben wordt vermeden. Ten tweede kan het op elk moment constant materiaal voor ons leveren. Ten slotte kan het ook gemakkelijk worden gebruikt om theorieën over pesticidengedrag te vergelijken bij een enkel experiment.In deze studie, stellen we theecel suspensie culturen van variabele kleuren van theebladeren. De optimale culturen status van celsuspensies werd vervolgens gebruikt om de dissipatie gedrag van zes pesticiden te vergelijken. Dit experiment zal worden uitgevoerd door Jiao Weiting en Ge Guoqin.Dit zijn onze theesteriele planten die werkt als de bron van explant. Snijd langs de middelste vleugel van een steriel blad met een schaar. En dan, verdeel elke helft in kleine stukjes van ongeveer 0,3 keer 0,3 centimeter in een petrischaaltje.Plaats de steriele explants op ms visuele medium met 2, 4-D en KT. Zes explants kunnen in een kolf van 300 milliliter worden geplaatst. Kweek het blad explants bij een constante temperatuur van 25 graden Celsius in het donker. Selecteer na 28 dagen de eerste generatie geïnduceerde callus en breng vervolgens over op een verse kolf.Verwerf de losse en variabele eelt na drie tot vier subculturen. Snijd de grote, variabele en losse eelt van het vaste medium in kleine stukjes met behulp van een steriel chirurgisch mes onder steriele omstandigheden. We zijn ongeveer drie gram van de kleine stukjes van een eelt.Plaats de eelt in B5 met 2, 4-D en de KT. Kweek de vloeibare celsuspensie bij een constante temperatuur in de schuddende couveuse in het donker. Na zeven tot tien dagen van het kweken, verwijder de cultuurfles, en laat ze staan voor een paar minuten. Neem de supernatant als sediment hier zal de cultuur dwingen tot vers medium.Verwijder de neergeslagen grote eelt. Verkrijg de laatste goed verzorgde celsuspensiecultuur na vier tot drie subcultuurcycli van elk 28 dagen. Houd een monster van levende cellen op honderd graden Celsius gedurende tien minuten als de controlecel voor levensvatbaarheid vlekken.Centrifuge alle cel suspensie cultuur gedurende acht minuten op 6,000 G.Verwijder de supernatant voordat de cellen op te schorten in 5 milliliter PBS buffer en schud het voor een minuut met de hand. Voeg 2,5 milliliter van de TTC-oplossing toe en schud opnieuw met de hand. Broed het mengsel een uur lang in een staande couveuse bij 13 graden Celsius.Voeg een aliquot van vierhonderd microliter toe om de voorraadoplossing van alle neonicotinoïden Thiamethoxam, Acetimoprid, Imidacloprid en Imideproxase te steriliseren. Allemaal naar organische eerste fase. Dimethoate en omethoaat in de cel suspensie culturen respectievelijk.Cultuur monsters van celsuspensies met insecticiden bij constante temperatuur, en schudden incubator snelheid. Neem de monsters op verschillende dagen. Verwijder een 1 milliliter aliquot van de homogene celkweek om het monster met een neonicotinoïde te testen.Plaats het in een 1,5 milliliter plastic centrifuge buis, en centrifugeren op 4, 000 G gedurende twee minuten. Geef de supernatant door een filtermembraan van 0,22 micrometer voor analyse door HPLC-UV en UPLC QTOF. Om het monster te testen, gaat u verder in de organische eerste fase.Verwijder een 500 microliter aliquot van de celcultuur en plaats in een 35 milliliter centrifugebuis of 1,5 milliliter plastic centrifugebuis. Voeg 0,1 gram natriumchloride en vijf milliliter extract oplosmiddel in de 35 milliliter centrifuge buis van de 500 microliter monsters. Vortex het mengsel voor een minuut.En laat ze dan 10 minuten rusten. Neem 2,5 milliliter supernatant in een 10 milliliter glazen buis en verdampen tot bijna-droogte met behulp van een stikstof verdamper op 14 graden Celsius. Los het residu op met één milliliter aceton.Vortex voor een minuut, geef het door een 0,22 micrometer filtermembraan voor analyse door GC-FPD. Zet vijf planten in vier liter plastic potten voor vijftien dagen. Voeg nul microgram per milliliter of 100 microgram per milliliter thiamethoxam of dimethoaxe toe in plastic potten, respectievelijk.Bereid het intacte plantenmonster volgens de vorige methode voor, behalve voor het voorweken en analyseer vervolgens door Orbitrap massaspectrometrie. Gebruik een HPLCUV om de inhoud en metabole producten van de Thiamethoxam en Acetimoprid te detecteren op golflengten van 254 nanometer, en van Imidacloprid en Imideproxase op 270 nanometer. Detecteer de inhoud van dimethoaat en omethoaat door GC-FPD met behulp van een buisvormige kolom.Detecteer de metabolieten van insecticiden in de celkweek met behulp van de UPRC QTOF met een Carbon-18 kolom. Detecteer de metabolieten van insecticiden in het extract van de testplant met behulp van UPLC Orbitrap massaspectrometrie. De eelt van een steriele plantlet heeft een lagere verontreiniging bruining, maar een hogere inductiviteit dat van eelt van geplukte theebladeren.De eelt afgeleid van steriele bladeren was altijd helder geelachtig, terwijl de eelt afgeleid van geplukte bladeren was wit met bruine vlekken. Toen de KT-concentratie 0,1 milligram per liter was, was de eelt geelachtig van kleur en los van textuur. De groei snelheid van eelt was de hoogste, tot 61,5%, toen de concentratie van 2, 4-D was een milligram per liter met de concentratie van KT was 0,5 milligram per liter.De groeigegevens van callus was de beste en de groei snelheid van callus was de hoogste bereikt 46,9%Daarom, de beste combinatie van plant homent was een milligram per liter van 2, 4-D en 0,1 milligram per liter KT voor de thee callus groei. De eelt bedekte de bladeren volledig door de vierde subcultuur, en toen de subcultuurcyclus 28 dagen was. De eelt was krachtig gegroeid met gelige kleur en losse textuur en geen bruining.Na het vergelijken van de bruto gegevens van callus en de kleur van celsuspensie, was het B5 vloeibare medium meer geschikt voor celsuspensiecultuur. De OD-waarde van de celsuspensiecultuur werd gebruikt om de hoeveelheid celgroei weer te geven. Tijdens de 75 dagen van de teelt, de verhouding van 15 gram per 40 milliliter had een OD waarde aanzienlijk hoger dan die van de andere twee ratio's.Cel levensvatbaarheid binnen de theecel suspensie cultuur werd getest door TTC kleuring. De kleurloze TTC-verbinding kan worden omgezet in rode formadin door dehydrogenase in mitochondriën van levende cellen, maar de kleur kan niet worden veranderd in dode cellen. Dit is het hele proces van het vestigen van een theecelsuspensiecultuur.Dit is de eerste vergelijkende studie van het metabolisme van verschillende pesticiden in thee met behulp van een cel suspensie cultuur techniek. Verschillende metabolieten werden geïdentificeerd, en sommige van hen werden verder onderzocht in bewaarde theeplanten. Deze nieuwe methode zou kunnen dienen als het model voor pesticide metabole studies in thee of andere planten.
