De toename van diabetes, immunosuppressieve therapie toepassingen, HIV, AIDS-epidemie, invasieve klinische procedures en breedspectrum antibiotica verbruik in de afgelopen jaren gestegen de incidentie van Candida albicans gerelateerde ziekten1,2. C. albicans infecties zijn meestal gerelateerd aan biofilm ontwikkeling en Klinische manifestaties, zoals candidiasis of systemische weerslag, zoals candidemia1,2kunnen veroorzaken. Een van de opmerkelijkste virulentiefactoren van biofilm groei is de polysacharide extracellulaire matrix vestiging. Biofilm vorming samenwerkt om te verhogen van de weerstand tegen bestaande antifungale geneesmiddelen, milieudruk en host immuun mechanismen3.
De groei van de biofilm van C. albicans begint met de vroege hechting van planktonische cellen op een substraat, gevolgd door de verspreiding van gistcellen door middel van het oppervlak van het substraat, en hyphal groei. De laatste fase van de groei van de biofilm is de maturatie fase, waarin gist-achtige ontwikkeling wordt onderdrukt, de hyphal ontwikkeling breidt en de extracellulaire matrix omsluit de biofilm4. C. albicans exopolysaccharides (EPS) in de matrix samen vormen de mannan-glucaan complexe5,6. De interactie van exopolysaccharides is van cruciaal belang voor de verdediging van de biofilms tegen drugs7. Vandaar, de vermindering van EPS uit de extracellulaire matrix van C. albicans kan ondersteunen de ontwikkeling van nieuwe antibiofilm protocollen voor orale candidiasis controle
Licht regelt de groei, de ontwikkeling en het gedrag van verschillende organismen8 en deze is vereffend als een antimicrobiële in Fotodynamische antimicrobiële chemotherapie (PACT). PACT is een zichtbaar licht van een specifieke golflengte en een licht absorberende fotosensitizer9van toepassing. De photosensitizers hebben echter problemen in de biofilm, doordringende veroorzaakt lagere werkzaamheid10. Het falen van therapeutische agenten om te infiltreren volledig biofilms is een reden dat biofilms, af en toe van zich verzetten tegen traditionele antimicrobiële therapie3,5. Deactiveren van de bijgevoegde Microbiële cellen, moeten antimicrobiële stoffen doordringen via de extracellulaire matrix; Toch is het EPS kenmerkt een diffusional obstakel voor dergelijke moleculen door te vragen hun niveau van vervoer in de biofilm of door het beïnvloeden van de reactie van de antimicrobiële met de matrix zelf11.
Gezien de nadelen van het PACT, het gebruik van licht vanzelf naar voren komt als een waardevolle verbetering. Voorlopige gegevens bleek dat de behandeling met blauw licht twee keer per dag de productie van EPS-onoplosbaar in Streptococcus mutans biofilm aanzienlijk geremd. Door de daling van de EPS-onoplosbaar, blauw licht verminderd biofilm groei. De resultaten van fototherapie rood licht met C. albicans biofilms zijn echter schaars. Vandaar, is het doel van dit onderzoek was om te evalueren in welke wijze fototherapie met behulp van rood licht de groei en de rangschikking van C. albicans biofilm beïnvloedt. Voor de behandeling tweemaal daags, aangepast we onze laboratory's eerdere protocollen9,12 te bieden een gemakkelijk en reproduceerbaar biofilm model dat antwoorden met betrekking tot de levensvatbaarheid levert, droog gewicht en extracellulaire polysacchariden bedragen na rood licht behandeling. Hetzelfde protocol kan worden gebruikt voor het testen van andere therapieën.

Hier presenteren we een protocol voor de beoordeling van de resultaten van per dag rood licht behandeling op de groei van Candida albicans biofilm. Het vergroten van de planktonische groei van C. albicans SN425, groeide de inoculums op gist stikstof Base media. Voor de vorming van de biofilm, waren RPMI 1640 media, die hoge concentraties van aminozuren hebben toegepast om te helpen biofilm groei. Biofilms 48 uur twee keer per dag gedurende 1 min werden behandeld met een niet-coherente lichtapparatuur (rood licht; golflengte = 635 nm; energiedichtheid = 87.6 J·cm-2). Candida is de meest voorkomende oorzaak van verspreide schimmelinfectie in immuungecompromitteerde gastheren. Het falen van therapeutische middelen om biofilm volledig te infiltreren vanwege de matrix is een van de redenen waarom biofilms zich verzetten tegen traditionele therapieën. Evenzo moet een stam worden gebruikt voorafgaand aan 18 uur van de nachtelijke groei. Bij het gebruik van een spectrofotometer voor de eerste keer is het belangrijk om eerst kolonie boerderij te verkrijgen in eenheden telt, om te correleren met optische dichtheid waarden.Daarom zullen experimenten beginnen met het gebruik van het midden van de groeifase in de initiële celconcentratie van 10 tot zeven cellen per milliliter. Incubaal aërobe bij 37 graden Celsius gedurende 90 minuten om celhechting aan de onderkant van de putten mogelijk te maken.Behandel de biofilms gedurende één minuut met één milliliter van 0,12%chlorhexidine voor de positieve controlebiofilms. Voor de negatieve controle, behandel de biofilms gedurende een minuut met een milliliter steriel, 0,89% natriumchloride. Vervolgens was de putten twee keer, elk één minuut, met één milliliter steriele 0,89% natriumchloride om niet-aanhangende cellen te verwijderen.Zet vervolgens een niet-coherent rood licht aan en gebruik een vermogensmeter om de vermogensdichtheid van het licht te meten. Krachtig vortex de biofilm suspensie voor een minuut. Voor CFU-analyse moet u een aliquot van 0,1 milliliter van de biofilmsuspensie overbrengen naar een microcentrifugebuis met 900 microliter van 0,89% natriumchlorideoplossing voor elke verdunning.Verdun de biofilmoplossing van 10 tot de negatieve, tot 10 tot de negatieve vier, in zoutoplossing. Zaad 50 microliter van elke verdunning tot SDA-platen en broed de platen gedurende 48 uur uit op 37 graden Celsius. Tel de kolonies en pas de formule toe.Voor droge gewichtsanalyse, eerst wegen en label micro centrifuge buizen. Voeg 0,1 milliliter van de biofilm suspensie en een milliliter absolute ethanol toe aan de buizen en bewaar ze gedurende 18 uur op negatieve 20 graden Celsius. Dan, centrifuge het 10.000 keer G gedurende 10 minuten.Gooi de supernatant en plaats de buizen in een desiccator om de monsters te drogen voor een week. Weeg de microcentrifugebuizen opnieuw af en doe de aftrekking om het biomassagewicht te verkrijgen. In dit experiment, na per diem behandelingen met rood licht voor een minuut naar Candida albicans biofilms, de CFU werd verminderd, in vergelijking met de negatieve controle behandeld met natriumchloride.De resultaten van de biomassa van Candida albicans biofilms na dagelijkse behandelingen, tonen de rood licht behandelde groep gepresenteerd soortgelijke vermindering van biomassa aan die waargenomen in de positieve controlegroep, behandeld met chloorhexidine. De twee groepen vertoonden ook statistische gelijkenis met elkaar. Terwijl ze beide een vermindering van de biomassa vertoonden, vergeleken met de negatieve controle behandeld met natriumchloride.Inferieure hoeveelheden Candida albicans oplosbare en onoplosbare EPS, werd waargenomen in positieve controle, in vergelijking met negatieve controle. Hoewel niet statistisch significant, per diem toepassing van rood licht numeriek verminderde de hoeveelheden EPS oplosbaar en EPS onoplosbaar. De negatieve controle en de rode licht behandelde groep, toonde statistische gelijkenis.Het meten van de vermogensdichtheid van het licht, met behulp van een vermogensmeter voordat u met de behandeling begint om de exacte toedieningsperioden te bepalen. De stappen zorgen ervoor dat de energiedichtheid altijd 87,6 joules per vierkante centimeter zal zijn. Gebruik de eerder aangehaalde formule.De meting wordt gemaakt met behulp van hetzelfde van het licht naar de fles van de platen, vijf millimeter. Een verband tussen fototherapie en schimmeldodende geneesmiddelen kan worden bestudeerd om te controleren of de voorbehandeling met rood licht, gevolgd door toepassing van geneesmiddelen, de penetratie van geneesmiddelen in de biofilm verbetert. Voor de tweemaal dagelijkse behandeling hebben we de protocollen van ons laboratorium aangepast, met een eenvoudig en reproduceerbaar biofilmmodel, dat antwoorden geeft met betrekking tot levensvatbaarheid, droog gewicht en extracellulaire polysaccharidehoeveelheden na een roodlichtbehandeling.Het protocol is generiek en kan worden gebruikt voor het testen van andere therapieën, naast rood licht, met inbegrip van drugs, andere lichten, of de combinatie van licht en drugs. Werken met micro-organismen vereist speciale apparatuur, zoals veiligheidsbril, knoop lab jas, en nitro handschoenen.
