该协议提供了一个快速和有效的策略，同时使用两个测定筛选数百种化合物，以了解其对细胞生长的影响。这些测定可以为我们提供有关特定化合物是否具有细胞毒性、抑制细胞生长或增强细胞生长的信息。通过利用这两种测定方法，我们能够提高准确性并识别细胞毒性化合物。在两种测定结果有分歧的情况下，此策略可能使我们能够识别对细胞功能具有独特的作用的化合物，可以通过其他方法进一步检查。这种策略的主要优点是快速、廉价和非劳动密集型。一旦掌握，它允许研究人员在一周内轻松执行多达200种化合物的初级筛选，然后在一周内对10至15种化合物执行详尽的剂量反应曲线。演示这个程序的将是我自己和罗希尼·马尼卡姆，一个工作人员科学家和实验室的博士后研究员。首先，在生物安全柜中，用NSC培养的T75烧瓶培养细胞，这样在开始检测时，它们至少会汇合80%。将烧瓶放在37摄氏度和5%的二氧化碳细胞培养箱中。一旦细胞达到80%的汇合，从培养箱中取出烧瓶，吸走NSC培养。加入三毫升细胞分离试剂，在培养箱中孵育五分钟。孵育后，在烧瓶中加入7毫升的NSC培养，并大力移液器，以确保所有细胞分离。将分离的细胞溶液转移到15毫升管中，以200倍G离心5分钟。从管中去除上一液，并在 10 毫升的 NSC 介质中重新悬浮细胞。使用自动细胞计数器或血细胞计来计算细胞，并将NSC培养素加入管中，将细胞浓度重新调整为每毫升20万个细胞。然后，在用细胞外基质涂层的3个96孔板的60个内部井中，使用8通道多通道移液器的8个槽中的6个，将100微升的细胞混合物柱一列地板入。将100个基基介质或NSC介质的微升添加到所有没有细胞的孔中，以最大限度地减少从含有细胞的最外层井的潜在蒸发。在细胞培养显微镜下，目视检查三个96孔板中每个孔至少10口，以确认细胞以预期密度位。在37摄氏度和5%的二氧化碳下孵育。分离后 16 至 24 小时从培养箱中取出细胞培养板，并使用 8 个通道多孔移液器，使用 8 个多孔槽中的 6 个，一列一列地吸走 NSC 介质。在三个复制板的每个孔中加入 95 微升的新鲜 NSC 介质，并将板放回培养箱中。使用八通道多孔移液器，在空U底、V底或圆底96孔板的内部60孔中加入49个NSC介质微升。解封准备好的主复合板。将一微升的化合物从主板移入每个内部60孔含有NSC介质，并混合稀释的化合物三次与一个板凳顶部移液器。接下来，从培养箱中取出三个96孔板的NSC，将稀释化合物的五微升分配到三个含有细胞的板块的每个孔中，对最终浓度为10微摩尔的最终化合物产生1至1000稀释，其中DMSO浓度为0.1%，化合物为72小时，然后进行细胞毒性测定。要进行剂量反应测定，请设置96井。使用第二列进行六次 DMSO 控制复制，并在六次剂量的连续稀释下测试多达三种不同化合物的三倍，从高剂量的 10 微摩尔开始。在0.5或1.5毫升微离心管中将4个100%DMSO或测试化合物稀释为196微升的NSC介质。加入25微升稀释的DMSO控制井，50微升稀释试验化合物到相应的井中。96 井板内部部分剩余的空井中，有 25 微升的 NSC 介质移液器。使用多通道移液器，将25微升化合物从10微摩尔井转移到5个微摩尔井，并至少混合5次。更换移液器尖端，然后通过混合剩余行来重复过程，以双重稀释产生三倍稀释剂，每个化合物总共六剂。将每次稀释的五微升转移到一个新的盘子里，培养细胞正在测试中。将板放在37摄氏度和5%的二氧化碳的培养箱中。细胞孵育72小时后，从培养箱中取出盘子，放在板读卡器内。打开成像器软件，为研究设置协议和实验文件。转到任务管理器上的成像器手动模式，然后单击"立即捕获"。选择 96 井板作为容器类型。选择 10 倍的放大倍率，红色荧光蛋白 531 和 593 用于成像 tdTomato。点击一个井。然后单击"自动对焦"以对焦图像，然后单击"自动曝光"以获得适当的曝光时间。如果需要，手动调整焦点和曝光。单击相机图标以捕获图片。然后单击图像上方的"过程分析"以继续构建协议。然后选择"分析"选项卡。在图像右侧，单击"添加分析步骤中的蜂窝分析"，然后单击"开始"。图像显示突出显示的单元格，以指示每个单独的单元格。单击"选项"以更改参数，以根据荧光阈值或细胞大小更好地选择单元格。如果成像器正确计算单元格，请单击屏幕底部的"添加步骤"。单击屏幕顶部的图标，从图像集创建实验。在打开的窗口中，单击"协议"选项卡下的过程。然后，在打开的新窗口中，选择"读取"。单击"全板"仅选择包含细胞的 60 个孔。单击"确定"以保存更改。然后在"过程"窗口中单击"确定"。单击"播放"图标以运行板。在提示中，保存文件，然后在"加载板"提示中，单击"确定"。完成映像后，将图成像器软件程序的单元格计数数据下载到电子表格中进行分析。在完成 tdTomato 成像的两个小时内，开始 MTT 测定。称出25毫克的MTT，并在5毫升的NSC介质中重新悬浮，使每毫升MTT库存溶液5毫克。涡流溶液几分钟，直到没有明显的MTT沉淀。从培养箱中取出细胞培养板，吸走细胞培养培养。在新鲜的 NSC 介质中稀释 MTT 1 至 10，并在每个细胞井中加入 100 微升稀释 MTT。在37摄氏度下孵育细胞两小时。紫色沉淀物与井中的细胞数量大致成比例可见。将 MTT 溶液从板材中吸走，并在每个孔中加入 50 微升 100%DMSO。在室温下将板放在摇床上 10 分钟，转速为 400 RPM。之后，将板转移到板读卡器。读取每一个井的吸收度在预设波长 595 纳米，并导出数据到电子表格进行分析，完全一样为细胞计数数据。对tdTomato荧光图像的检查显示，MTT和细胞计数测定之间的两口井对毒性不和谐。对于一个井，细胞计数数据表明没有毒性。但 Mtt 数据做到了。另一口井被MTT高估了相对于tdTomato计数的细胞计数，并且似乎有较大的细胞，而在MTT低估了相对于tdTomato的细胞计数的井中，细胞似乎更小。综上关于tDTomato测定，将11种化合物分类为有毒化合物，11种为生长抑制化合物，1种化合物为生长增强类，34种化合物对细胞生长无明显影响。MTT测定将13种化合物分类为有毒化合物，2种为生长抑制，1种为生长增强，其中41种对细胞生长无明显影响。化合物WP1066的可行细胞图显示了一条相对平坦的曲线，四个最低浓度无毒性。曲线在5微摩尔剂量下迅速下降，在10微摩尔时下降到几乎全毒性，这导致致命剂量50为4.4和6.0微摩尔用于tdTomato测定和MTT测定。在剂量响应屏幕中正确稀释化合物至关重要，以便生成正确的剂量反应曲线。通过这个程序确定影响细胞生长的化合物可以通过功能测定来进一步确定控制细胞生长的潜在新信号通路。对神经干细胞有毒的化合物已被测试为脑癌的潜在治疗剂，如胶质母细胞瘤和神经母细胞瘤。处理哺乳动物细胞、DMSO 和 MTT 时，始终戴上手套和实验室外套。根据您机构的化学废物指南处置 DMSO 和 MTT。
