Lspr 相关的光学行为高度依赖于纳米粒子的大小和形状, 可以用多种光学显微镜技术进行研究。然而, 为了破译具有各向异性 (即非球面) 形状的等离子体纳米粒子的取向信息, 有必要利用光场的极化。通过以较小的增量仔细旋转偏振场或样品基板, 可以监测单个纳米粒子的方向依赖性光谱特性。旋转和极化还可以帮助确定光谱特征是由于纳米粒子表面电子的双极振荡还是高阶振荡造成的。然而, 在各向同性 (即球形) 纳米粒子的情况下, 在偏振光下旋转样品时, 光谱剖面基本保持不变。

要执行单个粒子光谱, 重要的是使用正确的光学元件并正确对齐它们。必须使用能够使用 DIC 显微镜的客观特性。对于单粒子实验, 80x 或100x 油目标是理想的。Nomarski DIC 棱镜通常有三个品种: 标准、高对比度和高分辨率。理想类型在很大程度上取决于实验的目的和纳米粒子的大小。标准棱镜适用于许多实验;但当使用较小的纳米粒子 (< 50 纳米) 时, 高对比度棱镜可能是有益的, 因为颗粒对比度会随着颗粒尺寸的减小而减小。根据显微镜品牌或型号6的不同, 通过旋转偏光片或翻译其中一个 dic 棱镜来调整 dic 对比度。

DIC显微镜在复杂环境下的成像质质纳米粒子方面优于暗场。然而，DIC 也需要更多的知识和技能才能获得可重复的结果。DIC显微镜提供高横向分辨率和浅景深。在监测细胞内或复杂环境中的纳米粒子时，这两种特性都极为重要。首先，使用划线笔在每个玻璃盖玻片的中心放置一个浅和短的划痕，然后清洁玻璃，如随附的文本协议中所述。接下来，使用微移器从原始存储容器中去除 100 微升的 05 毫克每毫升金纳米颗粒溶液，然后将溶液喷射到 1.5 毫升离心管中。在6000倍的重力下将样品离心10分钟。完成后，用微管去除上表面液，以去除纳米颗粒溶液中多余的表面活性剂。接下来，在离心管中加入100微升的超纯水。短暂地旋转样品分解颗粒，然后立即对颗粒进行声波化20分钟，以完全重新释放纳米粒子的骨量。使用微管，将六微升纳米颗粒溶液滴到清洁和刮伤的玻璃盖玻片上。然后小心地翻转盖玻片，放在第二块较大的显微镜玻璃上。掉落应迅速均匀地分布在两块玻璃之间。小心避免气泡夹在两块玻璃之间。使用窄线指甲油密封盖玻片的边缘，以防止液体蒸发。将样品放在显微镜阶段，并对齐显微镜的目标和冷凝器。调整焦点以查找具有样本的焦点平面，并找到之前创建的划痕。关注它，微调焦点，直到纳米粒子进入视野。要确定冷凝器的准确位置，请利用科勒照明方法，从 20 倍目标开始，然后移动到高放大倍率，如 80 倍或 100 倍。接下来，在样本中选择一个感兴趣的区域进行成像。将区域居中摄像机的视场中，并根据需要调整对焦。如果显微镜具有 de Senarmont 设计，则从接近最大背景消光的偏振器设置开始，然后逐渐旋转偏振器以减少背景消光。背景强度将逐渐增加。当纳米粒子达到与局部背景平均值相比的最大强度差时，就实现了理想的设置。对于质质纳米粒子，最大对比度通常是在最大背景消亡时或接近最大背景的相对较暗的背景下实现的。关闭房间照明，以防止杂散照明与过程交互。在半最大带通滤波器上安装 10 纳米全宽全宽滤波器，其中央波长与主局部表面磁共振波长位于同一位置，以便查看感兴趣的区域。接下来，调整灯的强度或曝光时间，直到背景电平处于相机最大容量电平的 5% 到 40%。感兴趣区域内任何物体都不应表现出超过摄像机最大强度水平 90% 的信号强度。使用一系列带通滤波器对样品进行成像，每个滤波器的全宽最大为 10 纳米，并且作为一个整体，可以在整个波长范围内进行成像。通过调整曝光时间而不是灯功率，确保图像中的背景强度保持在 5% 以内。切换滤镜后，在捕获图像之前重新聚焦样本。将图像保存为未压缩的 TIFF 文件和/或软件的本机文件格式，以便保留所有相关信息。要开始分析，请使用 ImageJ 打开图像。使用矩形工具在主感兴趣区域周围绘制矩形。然后在工具栏上选择图像，转到缩放，然后选择选择。成像窗口将放大所选区域。接下来，选择图像，去调整，并选择亮度/对比度。要改善样本区域的视图，请调整图像的最小值、最大值、亮度和对比度。这些调整不会改变科学数据，它们只会使样本区域的可见性更好。现在，再次使用矩形工具，并在要测量的第一个纳米粒子周围绘制一个框。盒子应该只比纳米粒子的通风盘稍大一点。选择后，转到分析并选择度量值。将显示一个新窗口，报告位于选定框内的像素的最小、最大值和平均强度。保留盒子的原始大小，将框拖动到紧邻粒子的区域，背景对比度相对均匀，并且不存在颗粒或污染物。再次使用测量工具确定背景区域的均值强度。对系列中所有图像中的每个粒子重复此过程。然后将数据导出到电子表格中，以计算每个粒子在所有波长和角度中的对比度或强度。输入以下公式以计算每个粒子的对比度。最后，通过绘制沿 X 轴的波长和沿 Y 轴的对比度或强度的数据，在给定的纳米粒子位置绘制光谱轮廓。为了确定最佳成像设置，这些金纳米圈在多个偏振器设置下进行成像。在零度时，粒子看起来大部分是白色的，中间部分有一条暗条纹。这表明南圈样品的交叉极化。当偏振器旋转到不同角度时，粒子会向一个角投射暗阴影。但是，粒子对比度值会显著变化。对于此示例，最佳成像设置为偏振器移位 10 度。这里的波长是绘制的对金纳米圈的对比。每个数据点表示每个粒子直径的平均 20 个纳米圈。每个粒子的峰值对比度会根据其大小而略有变化。强度和旋转对各向异性形状（如金纳米棒）更为重要。在这里，波长在两个不同的偏振器设置下根据强度绘制。在每种情况下，光明面的强度都比黑暗面强得多。在舞台旋转过程中，单个金纳米棒在其局部表面的磁共振波长的强度剖面显示，当样品旋转 180 度时，暗边和亮边之间的强度差异很大。样品成像非常重要。它需要显微镜师的勤奋和耐心。如果您正在执行一个需要在一系列几天内重新成像示例的实验，那么地标（如划痕）对于重新定位您感兴趣的区域至关重要。研究纳米粒子的研究人员对DIC显微镜的兴趣日益增加，特别是在动态和细胞环境中。因为 DIC 提供最佳的空间分辨率，尤其是在复杂的采样环境中。
