人道的緊急事態の最初の段階では、被災者の生存のためには、清潔な水の供給、衛生、衛生へのアクセスが不可欠です。したがって、水質の監視は、水媒介性の発生を防ぐための優先事項です。汚染水が病気の原因になることはよく知られていますが、従来の実験法が利用できる場合でも、E型肝炎ウイルス(HEV)などのウイルス発生源を特定することは困難です。水質の制御は、FIB1、2、3、4の定量に基づいています。しかしながら、FIBの不在とロタウイルス(RoV)、ノロウイルス(NoV)、またはHEV5、6などのウイルス水媒介病原体の存在との間に相関関係がないことが広範囲に文書化されている。したがって、FIBに基づく水質基準を使用すると、水媒介性ウイルス病原体の存在に関連するリスクを過小評価する可能性があります。ヒトアデノウイルス(HAdV)、または特定の病原体などの指標ウイルスのサーベイランスは、ウイルス病原体への曝露を定義し、ヒト感染の潜在的な原因を特定するのに役立つだろう7,8, 9,10および衛生対策の有効性を検証する 11.
これまで、これらのシナリオでのウイルスの検出は、熟練したスタッフと複雑なロジスティクスに依存していました。VirWaTest(virwatest.org)は、使用時点で水サンプルからのウイルスの濃度およびその後の検出のためのシンプルで手頃な価格の、ポータブルな方法の開発を目的としています。
ウイルス濃度は、10L水サンプルの有機凝集の原理に基づいており、それによってウイルスは小さな体積12、13で回収される。フロッハを採取し、ウイルスを分解し、2週間以内に室温で保存すると核酸の劣化を防ぐバッファーに添加されます。
核酸抽出法は、核酸が吸着する磁性粒子の使用に基づいている。それらは、粒子が付着する磁気ピペットを使用して、ある洗浄バッファーから別の洗浄バッファーに移され、最後に溶出バッファーに移すことができます。得られたウイルス核酸懸濁液は、PCRに基づく分子法を用いて検出を行うことができる基準実験室に出荷することができる。各核酸抽出について、2つの異なる量が試料によって生じた酵素阻害を排除するために試験される。また、機器の可用性を最小限に抑えながら、使用時点で PCR テストを実行することもできます。プロセス全体は、電源装置とは無関係に実行するように設計されています (図1)。
ヒトによって排泄され、高濃度の排水サンプル中に見られるHAdVを検出する定量的なPCRアッセイは、使用時に実行されるように適合されている。HAdV は、ヒトの fecal ウイルスインジケーターとして使用されます。.MS2バクテリオファージの定量化のためのPCRは、MS2がプロセス制御としてVirWaTestで使用されているので、適応されています。この方法は、目的の任意のウイルスの検出のためにカスタマイズすることができます。
開発後、VirWaTestメソッドは、中央アフリカ共和国(RCA)とエクアドルの2つの異なる設定でユーザーによって適用され、実際の状況でプロトコルの適用に関するフィードバックを提供しています。
私たちの知るところによれば、これは、電源、大型機器、凍結/冷却条件に依存しない、使用時点でのウイルスの濃度と検出を可能にする最初の手順です。堅牢な結果を得るために、各水サンプルの2つの反復を収集することをお勧めします。

virwatest, a point-of-use method for the detection of viruses in water samples

人間や動物によって排泄されるウイルスは、水源を汚染し、この水を飲み物、食品灌漑、洗濯などに使用すると、人間の健康に危険をもたらす可能性があります。古典的な大腿菌の指標は、常にウイルス病原体の存在をチェックしないので、ウイルス病原体とウイルス指標の検出は、特に人道的なシナリオや地域でリスク軽減の対策を採用するために関連しています水媒介ウイルスの発生が頻繁に発生する場所。
