選択プロセス全体を通じて、細胞の母集団全体が単一のグループとして維持されます。クローン作成またはその他の分離方法は使用されません。細胞は、最初に薬物の低レベルに連続的に露出されます。続いて、細胞が薬物の存在下で増殖するように適応した後、薬物の用量は、薬物10、11の最終最適用量にゆっくりと増加する。あるいは、パルス薬物投与または変異発生は、薬剤治療12、13の前に行われる抵抗細胞の選択に使用することができる。残念ながら、薬剤耐性が発症しない症例は一般的に報告されていない。選択戦略は、臨床的に関連する抵抗を再構築するための癌患者の状態を模倣することを目的として開発されています。時には、薬剤耐性のメカニズムに関連する分子変化を同定するために、高い薬物濃度が使用される。このモデルは臨床的に関連性が低くなる。

肺腺癌PC-9細胞からアファチニブ耐性細胞株を確立する方法が開発され、耐性細胞が特徴付けられた。耐性細胞は、非小細胞肺癌患者に適用可能な表皮成長因子受容体チロシンキナーゼ阻害剤耐性機構を調製するために使用することができる。

新たに獲得した耐性機構の研究は、がん剤耐性および分剖可能癌患者におけるより効果的かつ安全な治療戦略の開発に寄与する。残念ながら、薬剤耐性が発現しなかった症例は一般的に報告されていません。この段階的な用量のエスカレーション方法は、取得した抵抗細胞を得るための最も信頼性の高い技術と考えられている。適切なアファチニブ耐性濃度を決定するために、10センチメートルの細胞培養処理皿中の培養培地中のPC-9細胞を摂氏37度および5%の二酸化炭素で培養した。PC-9細胞を成長培地濃度のミリリットル当たり4倍の10〜4個の細胞で再懸濁し、96ウェルマイクロプレートでウェルあたり50マイクロリットルの細胞懸濁液で細胞を座る。細胞培養インキュベーターで一晩インキュベートした後、細胞をウェルあたり50マイクロリットルのアファチニブ溶液で処理し、示された濃度の6回の反復で、およびカリング培養インキュベーターで96時間のインキュベーションの後、各ウェルに15マイクロリットルのMTT染料を加える。細胞培養インキュベーターで4時間後、各ウェルに100マイクロリットルの可溶化/停止溶液を加え、プレートを一晩インキュベーターに戻します。翌朝、マイクロプレートリーダー上の570ナノメートルの光学密度を測定します。連続的なアファチニブ曝露のために、細胞が亜合流に達するまで10ミリリットルの増殖培地を含有するP-100ディッシュ中のPC-9細胞を培養する。サブコンフルエント培養物を、初期培養皿毎に9ミリリットルの増殖培地の3つの新しいP-100皿に移し、細胞を培養インキュベーターに戻す。翌朝、3つの培養皿の各セットに0.1ナノモル、またはアファチニブの半最大阻害濃度の1/10を加える。アファチニブ処理細胞がサブコンフルエントになったら、1ミリリットルのピペットを使用して培養物を完全に混合し、各皿から新しいP-100皿の新鮮な成長培地の9ミリリットルに1ミリリットルの細胞を移す。新しい培養物に10%〜20%高い濃度のアファチニブを加え、10〜12ヶ月間に培養が亜合流に達するたびに段階的な用量エスカレーションによってアファチニブ濃度を増加させる。1マイクロモルのアファチニブ濃度に達した場合、細胞がアファチニブ耐性を発症したことを確認するために実証されたMTTアッセイを行う。細胞株の増殖曲線を決定するために、培養親PC-9および3つのアファチニブ耐性細胞株を細胞培養インキュベーター中で24時間培養培地で培養する。各培養物の細胞を、培地濃度の1ミリリットル当たり5倍10〜3番目の細胞で再懸濁し、96ウェルマイクロプレートで各細胞集団を12個100マイクロリットルずつ複製する。次に、MTTアッセイを示されているように、0日目、1日目、2日目、3日目、5日目、7日目に実行し、適切な統計解析ソフトウェアプログラムを使用して結果をプロットします。表皮成長因子受容体、またはEGFRゲノムDNA発現における変化を同定するために、逆転写酵素ポリメラーゼ鎖解析により、製造業者の指示に従ってDNA精製キットを用いて目的の細胞培養物からゲノムDNAを単離する。分光光度計上の分離されたゲノムDNAの25ナノグラム-マイクロリットル濃度の濃度を測定する。次にSYBRグリーンマスターミックスを用いて50ナノグラムのゲノムDNAを増幅し、蛍光ベースの逆転写酵素ポリメラーゼ連鎖反応検出システムを用いて結果を解析した。シーケンシングによるEGFRゲノムDNA発現の変化を同定するために、EGFRエキソン19〜21に特異的なプライマーを用いてゲノムDNAを増幅する。次に、メーカーの指示に従ってPCR精製キットを使用して増幅PCR製品を精製し、アンプリコンを配列します。ウェスタンブロット分析による表皮成長因子受容体タンパク質発現の変化を同定するために、細胞をアファチニブで24時間治療する。24時間後、細胞培養物を1回の培養ごとに5ミリリットルの氷冷PBSで2回洗浄する。細胞を放射性免疫沈降アッセイバッファーにリーゼし、1%プロテアーゼ阻害剤カクテルおよびホスファターゼ阻害剤をカクテル2と3を摂氏4度で30分間添加する。インキュベーションの終了時に、遠心分離によってライセートを収集する。ライゼートサンプルのタンパク質濃度を決定するために、ビチンチオン酸アッセイを使用します。4-Xサンプルバッファーで0.5、またはマイクロリットル当たり1マイクログラムにタンパク質サンプルを調整し、サンプルを摂氏96度で5分間沸騰させます。サンプルのウェスタンブロット分析のために、エタノール洗浄ガラス板およびスペーサーを組み立て、適切な実験用サプリメントを用いて8%ポリアクリルアミドゲルで金型をロードします。ゲルが重合している間、適切な金型にスタックゲル溶液を加え、櫛を挿入し、積層ゲルを室温で20〜30分間重合させます。両方のゲルが重合したら、それらを電気泳動装置に入れ、タンクにランニングバッファを充填します。その後、各タンパク質サンプルの20〜30マイクロリットルの体積を各ウェルに1つロードし、180ボルトでゲルを実行します。約60分後に電気泳動を停止し、染料フロントがゲルから流れ出たら、ツイーンを補ったトリスバッファー生理食音でゲルを1〜2分間洗浄します。300ミリアンペアの一定の電流で1時間と1/2時間半乾燥ブロッティングによってポリビニリデンフッ化物膜にタンパク質を移します。トリスバッファー食塩水とトゥイーン溶液で希釈した5%ノンファットドライミルクを室温で1時間使用して膜をブロックします。次に、摂氏4度で目的の適切な抗体を一晩で膜を探る。翌朝、洗浄ごとに新鮮なトリス緩衝生理食糸を3回洗浄して膜を洗浄した後、膜を適切な二次抗体に1~1時間、室温で1/2時間曝露します。次いで、洗浄ごとに新鮮なトリス緩衝生理生理液で膜を5回洗浄し、膜を使用してシグナル可視化のための強化された化学発光溶液に膜を露出させます。ここで、アファチニブの濃度の増加に応答して親のPC-9細胞の細胞増殖において観察される減少が図示され、PC−9細胞がアファチニブ曝露に敏感であることを確認する。しかし、3つのアファチニブ耐性細胞株のいずれも、アファチニブ曝露下での細胞増殖の抑制を示す。アファチニブ耐性細胞株は、親のPC-9細胞よりも著しく遅い成長曲線を示す。アファチニブ耐性細胞は、親のPC-9細胞よりもEGFRゲノムDNAおよびEGFRタンパク質発現の有意に高いレベルを発現する。EGFRシーケンシングは、PC−9細胞がエキソン19において15塩基対欠失を示し、かつ野生型EGFRがエキソン20に存在することを示す。しかし、アファチニブ耐性の1および2つの細胞株細胞は、野生型EGFRエキソン19の増幅を示す。アファチニブ耐性細胞株3細胞は、PC−9細胞と同様の15塩基対欠失をEGFRエキソン19に含むが、EGFRエキソン20ではT790Mの点突然変異が認められる。細胞増殖は、インヒビター濃度がIC-50値に近いとかなり遅くなることがある。細胞が徐々に増殖し続けるので、培養について議論しないでください。これらの戦略は、がん患者が臨床的に関連する耐性を開発し、取得した抵抗メカニズムを評価し、安全で効果的な治療戦略を開発する条件を模倣するために開発されました。
