Con este método, podemos estudiar los efectos de la actividad intrínseca en la maduración de los precursores de interneurón cortical. Esta técnica proporciona una manera eficiente de orientar y manipular las propiedades celulares de los progenitores interneurones corticales que es accesible a la mayoría de la comunidad científica. Este protocolo implica el manejo meticuloso de muestras y equipos, y se puede apreciar mejor mediante demostración visual.
Usando una pluma impermeable, dibuja una línea recta a través de la mitad de la parte inferior de la superficie inferior de seis platos Petri de 35 milímetros. Añadir 10 mililitros de líquido 4%baja de agarosa gelificante en PBS en dos de los platos de Petri de 35 milímetros e incrustar de tres a cuatro cerebros diseccionados en la agarosa con las bombillas olfativas mirando hacia abajo por plato a través de la línea recta, dejando de tres a cinco milímetros de espacio entre cada muestra. Cuando todos los cerebros se hayan incrustado, coloque los platos a cuatro grados celsius para permitir que la agaria se solidifique y esculpí los tres cerebros en un solo bloque de agarosa, dejando aproximadamente tres milímetros de gel alrededor de los bordes de las muestras.
Pegue el bloque en la superficie de una base de microtomo y utilice una cuchilla quirúrgica para cortar todo el camino a través de la parte inferior del bloque entre cada muestra de tejido para obtener tres bloques independientes. A continuación, utilice un microtoma de hoja vibratoria para secuerpar los bloques en la solución de Krebs helada en rodajas de 250 micrómetros de espesor, utilizando una microspatula plana doblada para recoger sólo las rodajas que contienen las eminencias gangliónicas mediales o caudales. A continuación, coloque cada sección sobre membranas de filtro individuales de 13 micrómetros de diámetro, de tamaño de poro de ocho micrómetros que flotan en un medio esencial mínimo en platos individuales de cultivo de órganos de centro de poliestireno.
Una vez obtenidas todas las secciones, coloque los platos en una incubadora de cultivo de tejido de dióxido de carbono a 37 grados centígrados durante una hora. Antes de la inyección focal de ADN, coloque columnas de agarosa de 10 milímetros de diámetro y 10 milímetros de largo perforadas con una pipeta de vidrio de vidrio de 225 milímetros de largo y dos mililitros de capacidad en una solución de Krebs helada. Usando una cuchilla quirúrgica, corte una pieza más pequeña de agarosa para caber en la superficie del electrodo de electroporación y una pieza más grande para ser utilizada como base para las inyecciones focales de ADN.
A continuación, coloque los bloques de agarosa en la solución de Krebs helada. Para la inyección, mezcle el ADN vectorial de expresión y control a una concentración de un microgramo por microlitro para cada vector y agregue una solución rápida de stock verde en una dilución de uno a 10o. Llene una micropipeta de vidrio de diámetro exterior de 0,5 milímetros de diámetro interno con 10 microlitros de la mezcla de ADN y cargue la micropipeta en un inyector neumático PicoPump.
Coloque el bloque más grande de agarosa bajo un microscopio estéreo y coloque la rebanada que se va a inyectar en la pieza de agarosa. A continuación, inyecte de 25 a 50 volúmenes de nanter en la región de eminencia gangliónica medial o caudal seleccionada de la rebanada. Inmediatamente después de la inyección, coloque el pequeño bloque de agarosa en el electrodo de la placa Petri y utilice una microspatula plana para fijar la columna de agarosa al electrodo de la cubierta móvil.
Transfiera la rodaja inyectada con su membrana de soporte sobre el bloque de agarosa y coloque el electrodo superior con la columna de agarosa en la parte superior de la región seleccionada de la rebanada. Luego, electropograta la región con dos pulsos de cinco milisegundos de 125 voltios, 500 milisegundos de diferencia. Después de la electroporación, coloque la rebanada con su membrana de soporte de nuevo en su plato de retención y devuelva el plato a la incubadora de cultivo de tejido.
Después de una hora, reemplace el medio esencial mínimo por un medio básico adecuado para cultivos neuronales primarios y devuelva las rodajas a la incubadora durante la noche. En estos experimentos de electroporación, aproximadamente el 50% de las neuronas positivas de la GFP co-expresaron la proteína inyectada positiva RFP y, por lo tanto, la población negativa de RFP positiva de la GFP sirvió como control interno para el efecto de los ligandos de ADN inyectados. La administración de clozapina y óxido aumenta selectivamente la actividad de las células positivas de RFP transfectadas, como lo demuestra la expresión de la proteína dependiente de la actividad, c-Fos, en estas secciones del tejido coronal recién nacido.
El tratamiento de clozapina y óxido también da lugar a un aumento en la proporción de células RFP positivas en la GFP en relación con el número de células negativas RFP positivas en GFP en comparación con los littermatos administrados por vehículos. Este procedimiento se puede utilizar para estudiar los efectos de las interneuronas injertadas moduladas por actividad en la plasticidad circulatoria y la función cerebral del huésped.
