Dieses Manuskript beschrieb ein Protokoll von MEC und Perizyten-Isolierung von LG und SMG. Dieses Verfahren basierte auf der genetischen Kennzeichnung von SMA, dem einzigen zuverlässigen Biomarker von MECs und Periicytes.

Myoepitheliale Zellen (MECs) sind in vielen exokrinen Drüsen vorhanden, darunter Tränendrüsen, Speichel, Harderian, Schweiß, Prostata und Säugetiere. MECs sind ein einzigartiger Zelltyp, der einen Epithel-und einen glatten Muskelphenotyp kombiniert. MECs drücken α-smooth Muskelaktin (SMA) aus und haben eine kontraktile Funktion1,2. Die Tränendrüse (LG) und die submandibuläre Drüse (SMG) enthalten neben MECs auch SMA + Gefäßzellen, die Periicytes genannt werden, also Zellen endodermalen Ursprungs, die die Oberfläche von Gefäßrohren 3 umhüllen. Obwohl MECs und Periicytes viele Marker ausdrücken, ist SMA der einzige Marker, der nicht in anderen LG und SMG-Zellen1,3 ausgedrückt wird.
In den letzten 40 Jahren berichteten mehrere Labore von Untersuchungen zur Dissoziation verschiedener exokriner Drüsengewebe, in denen nicht-enzymatische und enzymatische Ansätze angewandt wurden. In einem der ersten Berichte, die 1980 veröffentlicht wurden, beschrieben Fritz und Co-Autoren ein Protokoll zur Isolierung von Katzenparotid acini mit sequenzieller Verdauung in einer Collagenase/Trypsin Lösung4. Im Jahr 1989 haben Hann und Coauthors dieses Protokoll für die Isolierung von Rat LGs mit einer Mischung aus Collagenase, Hyaluronidase und DNase5angepasst. 1990 veröffentlichten Cripps und Kollegen die Methode der nicht-enzymatischen Dissoziation der Tränendrüseacini 6. Später, im Jahr 1998, kehrten Zoukhri und Coautoren zu einem enzymatischen Dissoziationsprotokoll zurück, weil sie Ca2 +-Bildgebung auf LGundSMG isolierten acini 7 nachstellten. In den letzten zehn Jahren haben die Forscher ihren Fokus auf die Isolierung von Stamm/-Vorläuferzellen aus exokrinen Drüsen gerichtet. Pringle und Coauthors beschrieben 2011 ein Protokoll zur Isolierung von Mäuse-SMG-Stammzellen8. Diese Methode basierte auf der Isolierung von stammzellhaltigen Salisphären, die in der Kultur beibehalten wurden. Die Autoren behaupteten, dass sich vermehrende Zellen, die Stammzellmarker ausdrücken, von diesen Salisphären8isolieren könnten. Shatos und Coautoren veröffentlichten das Protokoll zur Vorläuferzellenisolierung von ungeschädigten erwachsenen Ratten-LGs mit enzymatischer Verdauung und dem Sammelnvon"befreiten" Zellen 9. Später, im Jahr 2015, passten Ackermann und Mitautoren dieses Verfahren an, um mutmaßliche "Murine-Tränendrein-Stammzellen" ("mLGSCs") zu isolieren, die sich als Einschichtkultur über mehrerePassagen 10 verbreiten ließen. Allerdings ist keines der oben genannten Verfahren erlaubt, um zelluläre Subtypen und einzelne Populationen isolierter Epithelzellen zu unterscheiden. Im Jahr 2016 veröffentlichten Gromova und Coautoren ein Verfahren zur Isolierung von LG-Stem/Vorläuferzellen von erwachsenen Murin-LGs mit FACS11. Dieses Protokoll war jedoch nicht dazu gedacht, MECs zu isolieren.
Vor kurzem haben wir gezeigt, dass wir in der Lage sind, SMA +-Zellen von 3 Wochen alten SMA-GFP-Mäuse 12 zu isolieren. Zum jetzigen Zeitpunkt haben wir jedoch keine verschiedenen Populationen von SMA +-Zellen getrennt. Hier haben wir ein neues Verfahren zur direkten Isolierung von differenzierten MECs und Pericytes von erwachsenen LGs und SMGs eingeführt.

Die Tränendrüse (LG) ist eine exokrine Tubuloacinardrüse, die eine wässrige Schicht Tränenfilm abscheidet. Der LG-Epithelbaum besteht aus Akinar-, duktalen Epithel-und Myoepithelzellen (MECs). Dieses Protokoll ermöglicht den Vergleich der Funktion und der regenerativen Fähigkeiten gesunder und kranker Myoepithelzellen und die Verarbeitung dieser Zellen für Genexpressionsstudien.Myoepithelzellen existieren in anderen exokrinen Drüsen wie Brust, Speichelundschaft und Bauchspeicheldrüse, wodurch dieses Protokoll durch Isolierung von Myoepithelzellen und Pericyten aus jedem anderen Gewebe angepasst werden kann. Mit einer Breitbohrpipettenfilterspitze das gehackte Gewebe in ein zwei Milliliter Rundbodenrohr übertragen und das Volumen im Rohr mit Verdauungsmedium auf zwei Milliliter bringen.Invertieren Sie das Rohr zu mischen, und legen Sie das Rohr in einem 37 Grad Celsius Schüttel-Inkubator für 90 Minuten bei 100 bis 120 Umdrehungen pro Minute. Alle 30 Minuten trituieren die Drüsenstücke 20 bis 30 Mal mit 1000 Mikroliter Filterspitzen mit abnehmender Bohrungsgröße. Am Ende jeder Trituration, inspizieren Sie ein 10 Mikroliter Aliquot unter dem Mikroskop für Cluster.Nach 90 Minuten die Probe zwei- bis dreimal durch eine Insulinspritze Nadel passieren, um die Zellen weiter in Suspension zu geben und die Zelllösung in eine 15 Milliliter Tube zu übertragen. Übergeben Sie die Zellen durch ein 70-Mikrometer-Zellsieb in ein 50-Milliliter-Rohr und waschen Sie das Sieb mit einem Milliliter Blocking Medium Type 1, bevor Sie die Zellen ein einziges Mal durch den Filter belasten.Als nächstes sammeln Sie die Zellen durch Zentrifugation und setzen Sie das Pellet in zwei Millilitern blockierender Blockierung Medium Typ 2 wieder auf. Übertragen Sie die Zellen in ein Zwei-Milliliter-Mikrozentrifugenrohr und sammeln Sie die Zellen durch Zentrifugation. Setzen Sie die Zellen in einem Milliliter Accutase-Lösung für eine zwei- bis dreiminütige Inkubation bei 37 Grad Celsius und 100 bis 120 Umdrehungen pro Minute aus.Übertragen Sie die Zellen am Ende der Inkubation in ein 50-Milliliter-Rohr und addieren Sie bis zu 10 Milliliter Blocking Medium Type 1. Nach der Zentrifugation setzen Sie die Zellen in sechs Millilitern Erholungsmedium für eine 30-minütige Inkubation bei Raumtemperatur wieder auf. Überprüfen Sie am Ende der Inkubation eine 10-Mikroliter-Probe unter dem Mikroskop, um eine vollständige Zelldissoziation zu gewährleisten.Nach dem Zählen die Zellen durch Zentrifugation sammeln. Um die Zellen für fluorescence-activated Cell Sorting (FACS) zu färben, addieren Sie bis zu fünf mal 10 zellen pro Zwei-Milliliter-Röhre, die 400 Mikroliter FACS-Puffer enthält, und fügen Sie fünf Mikroliter Brilliant Violet 421 Anti-Maus-CD326 und 0,5 Mikroliter Ghost Red 780 Viability Dye hinzu. Nach gründlichem Mischen die Rohre mit Folie für eine 45-minütige Inkubation bei 4 Grad Celsius mit Rotation wickeln.Am Ende der Inkubation die Zellen durch Zentrifugation sammeln und die Pellets in einem Milliliter FACS-Puffer pro Probe wieder aufsetzen. Übertragen Sie die Zellproben auf einzelne fünf Milliliter FACS-Röhren auf Eis und passen Sie die Kompensation mit einfarbigen Kontrollen an. Sortieren Sie dann die Zellen mit 20 Pfund pro Quadratzoll durch eine 100-Mikrometer-Düse.Mikrovaskuläre Pericyten entwickeln sich um die Wände von Kapillaren und zeigen eine quadratische Form. Myoepithelzellen umgeben die lakrymal ensekretorischen Acini, haben lange Prozesse und besetzen eine relativ große Fläche. Hier werden Zellen gezeigt, die sich von einer lakrymalen Drüse in Vorbereitung auf die fluorescence-aktivierte Zellsortierung dissoziierten, wie gerade gezeigt.Für die zytometrische Sortierung von Myoepithelzellen und Pericyten sollten die Zellen zunächst durch ihre vorderen und seitlichen Streubereiche abgezäut werden, bevor Doublets ausgeschlossen werden. Nach dem Gating, um die abgestorbenen Zellen auszuschließen, werden Kontrollproben verwendet, um das Hintergrundrauschen zu bestimmen und eine angemessene Kompensation für eine optimale Trennung zwischen Signalen zu etablieren.
