ここでは、側方のジェラジアル原核を含む急性脳スライスの調製のためのプロトコルと、レチナルジェノレートおよび皮質原性シナプス機能の電気生理学的調査を提示する。レチナルガンギ細胞軸索は、皮質入力から遠く離れた側側の原形核に入る。これは、非常に異なる機能的特性を有するレチナルジェノレートおよび皮質原性シナプスの別々の刺激を可能にする。この手順を開始するには、解剖液の100ミリリットルで満たされた2つのスライスチャンバーと記録溶液の100ミリリットルで他を準備します。2つのビーカーを摂氏37度の水浴に入れ、解剖前に少なくとも15分間カルボーゲンで溶液を泡立てます。次に、プラスチックビーカーに氷冷に近い解剖液を250ミリリットル充填し、使用前に少なくとも15分間カルボゲンで泡立てます。その後、脳解剖のための冷たい解剖溶液でプラスチックペトリ皿を充填します。ペトリ皿の蓋にフィルターペーパーを入れ、少量の解剖液で満たします。その後、ビブラートメの解剖室を冷やす。頭の皮膚を尾骨から鼻に切り、指で開けて頭蓋骨を露出させます。前脳を得るために、最初に後部/前正線に沿って頭蓋骨を切断する。その後、コロナ縫合糸とラムドイド縫合糸をさらに2回カットします。冠状縫合糸とラムドイド縫合糸の間の頭蓋骨を取り除き、大脳を露出させる。嗅球を切り、前脳を細かい刃で中脳から分離する。薄いへらで頭蓋骨から脳を取り除き、ペトリ皿の蓋のフィルターペーパーの上に置きます。脳スライス中の側根核への感覚および皮質入力の完全性を維持するために、2つの半球を3〜5度の角度で切断したパラサジタルカットで分離する。その後、2つの半球の内側側面をフィルターペーパーに置いて乾燥させます。次に、水平面から10〜25度の角度で切断段階に接着します。金属バッファートレイの中央にステージを置き、残りの氷冷解離液を緩やかにバッファートレイに注ぎます。あまりにも強く注ぐと、貼り付けた脳が取り除かれる可能性があるため、このステップを慎重に実行してください。次に、液中トレイを氷で満たされたトレイに入れ、解剖時の低温を維持します。250マイクロメートルの切片は、1秒あたり0.1ミリメートルの速度と1ミリメートルの振幅でカミソリの刃を有する。34°Cで酸素化解剖液を充填した保持チャンバーに30分程度移し、さらに34°Cで記録溶液中でスライスを回復させます。回収後、水浴からスライス保持室を取り出し、実験に使用するまでスライスを室温に保ちます。この手順では、ホウケイ酸ガラスキャピラリーとフィラメントプーラーを使用して記録ピペットを引き出します。次に、同じプロトコルを使用して刺激ピペットを引っ張りますが、直径を大きくするために引っ張った後、先端を少し壊します。続いて記録ピペットを細胞内溶液とバイオシチンで満たし、刺激ピペットを記録溶液で満たします。次に、記録チャンバーにスライスを入れ、室温で酸素化記録溶液を継続的に浸透させます。IR-DICビデオ顕微鏡を搭載した直立した顕微鏡でスライスを視覚化します。すべてのスライスを確認し、無傷の視路を表示するものを選択します。細胞に記録ピペットをパッチする前に、刺激ピペットをスライスに置きます。レチナル・ジェノレート・シナプスを調べるには、刺激ピペットを、眼球神経節細胞の軸索繊維が束なっている光学路に直接置きます。皮質原発性シナプスを解析するには、刺激電極を視床核に置き、これは側核に隣接する星質起源である。記録ピペットを記録溶液に浸漬したら、ピペット抵抗を監視するために5ミリボルトのステップを適用します。保持電位をゼロミリボルトに設定し、保持電流がピコインゼロになるようにオフセット電位をキャンセルします。正圧を加えながら、記録ピペットでセルに接近します。ピペットが細胞膜に直接接触する場合は、陽圧を放出し、保持電位をマイナス70ミリボルトに設定する。その後、ギガオームシールが形成されるように、細胞膜がガラスピペットに付着できるようにわずかな負圧を適用します。ピペット容量を補償し、負圧パルスを印加してセルを開きます。シナプス機能を調べるには、刺激ピペットを介して0.1ミリ秒の電流パルスを適用します。5ミリボルトのステップを適用して、連続抵抗を継続的に監視します。直列抵抗は、5ミリボルトを呼び起流電流のピーク振幅で割ることによって推定することができる。分析には、20メガオームより小さい直列抵抗を持つセルのみを使用します。バイオシチン標識の場合、少なくとも5分間全細胞構成を維持し、遠位樹状へのバイオシチンの拡散を可能にする。記録後、細胞からピペットをそっと取り出して、ソーマが破壊されないようにします。24ウェル細胞培養プレートを準備します。4%PFAの300マイクロリットルで各井戸を充填します。記録されるスライスと接触するPFAで何かを汚染しないように注意してください。記録チャンバーからゴム製ピペットでPFA含有プレートにスライスを移し、一晩固定します。翌日、PFAをPBSに置き換えます。今後の処理のためにPBSでスライスを摂氏4度に保ちます。細胞を染色するには、スライスを新鮮なPBSで3回洗います。ブロック非特異的結合部位は、ブロック溶液中のスライスを軌道シェーカー上の室温で2時間インキュベートすることによって行う。次いでブロッキング溶液を廃棄し、ブロック溶液で一晩4度のブロッキング溶液で希釈したストレプトアビジンAlexa 568でスライスを軌道シェーカーでインキュベートします。翌日、抗体溶液を廃棄し、オービタルシェーカー上の室温でそれぞれ10分間PBSでスライスを3回洗浄します。その後、取り付ける前に水道水でスライスを洗います。1 つのマウスのスライスを 1 つのガラス スライドに置きます。染色されたセルがスライスの上面に存在することを確認します。スライスの周りの水をティッシュで吸収し、スライスを約15分間乾燥させます。その後、各スライスに取り付け媒体を2滴塗布し、気泡を生成することなくスライドにカバースリップを取り付けます。ラベル付きスライスを共焦点顕微鏡で見た後、4°Cで保管します。このIR-DIC画像は、パッチピペットの先端を有する後端横型核リレーニューロンのソーマを示す。バイオシチン染色は、記録されたニューロンのビューを得ることを可能にします。双極性形態を有するインターニューロンとは異なり、リレーニューロンは3つ以上の一次樹状突起を含む多極樹状を有する。その後、バイオシチン標識リレーニューロンの3次元再構成が生成され、これは放射対称樹状アーキテクチャを示す。同じ脳スライス内のレチナルジェノレート入力と皮質原質入力を維持することが、このプロトコルにおいて最も重要です。この手順に従って、例えば、核網状核から後方横方向の神経細胞に対するインプヒターのインプットを調べることもできます。PFAは危険であるため、PFAで脳スライスを固定する際は手袋を着用してください。
