転写におけるタンパク質とタンパク質の相互作用は、従来、Por透析を用いて研究されてきた。しかし、Por透析は純粋に定量的です。この問題を克服するために、バイオレイヤ干渉法(BLI)を使用しています。BLIは、Por danと比較して、ボンディングパートナー間のリアルタイムの関連付けと解離を検出します。また、相互作用メカニズムを示す定量的な運動パラメータを生成します。BLI技術は威圧的に聞こえるかもしれませんが、この技術を使用するには、BLI機器と関連ソフトウェアへのアクセス権を持っている必要があります。このビデオを使用すると、BLIテクノロジーに簡単に適応できます。アッセイ開始の約10分前に、PCRチューブに200マイクロリットルのBLIバッファーをピペット。手袋をした手でバイオセンサーの広い部分を保持することにより、元の包装からニッケルNTAバイオセンサーを取り除きます。バイオセンサーのガラス先端のみがBLIバッファーに沈むようなPCRチューブの上にバイオセンサーを置きます。バイオセンサーチップを少なくとも10分間水没させ、完全な水分補給を確実にします。ブリッツマシンをオンにします。マシンの背面にある USB データ出力ポートを介して、コンピューターに接続されていることを確認します。コンピュータで、関連するソフトウェアを開き、画面の左側にある[高度なキネティクス]をクリックします。ソフトウェアで、それぞれの見出しの下に実験に関する適切な情報をすべて入力します。[バイオセンサータイプ]をクリックし、ドロップダウンメニューからニッケルNTAを選択します。各ステップの期間は、必要に応じてデフォルトから変更できます。最適な結果を得るには、初期ベースラインとベースラインに最低 30 秒、関連付けと解離に 120 秒を使用します。水分補給ニッケルNTAバイオセンサーをPCRチューブから取り出し、バイオセンサーの広い部分をマウントにスライドさせて機械のバイオセンサーマウントに貼り付けます。0.5ミリリットルの黒いマイクロ遠心分離チューブを機械のチューブホルダーに入れ、400マイクロリットルのBLIバッファーを入れます。マイクロ遠心チューブ内のバッファーにバイオセンサーチップが沈むような機械のカバーを閉じます。ソフトウェアの [次へ] をクリックして、初期ベースラインの記録を開始します。最初のベースラインステップの記録が完了したら、マシンのカバーを開きます。スライダーを右に動かし、ドロップ ホルダが黒い矢印の前に配置されます。透析したヒスタグ付きリガンドのピペット4マイクロリットルをドロップホルダーに取り付け、機械のカバーを閉じ、自動的にローディングステップの記録を開始します。積み込みステップの記録が完了したら、マシンのカバーを開きます。スライダーを左に動かし、チューブホルダーが再び黒い矢印の前に置くようにします。機械の蓋を閉め、バイオセンサーチップがチューブホルダーのチューブのBLIバッファに沈み込まないようにします。コンピュータとソフトウェアは自動的にベースライン ステップの記録を開始します。ベースライン ステップの記録が完了したら、マシンのカバーを開きます。ドロップホルダーを取り除き、タンパク質をピペットアウトし、合計5回二重脱イオン水で洗い流して洗浄します。最後の洗浄後に、ティッシュワイプを使用して、落下ホルダーの表面を洗浄します。ドロップホルダーをマシンに戻します。マシンのスライダーを右に動かし、ドロップホルダーが再び黒い矢印の前に置くようにします。透析した検地のピペット4マイクロリットルをドロップホルダーに取り付け、機械のカバーを閉じて、自動的に関連ステップの記録を開始します。関連付けステップの記録が完了したら、マシンのカバーを開きます。チューブホルダーが再び黒い矢印の前に配置されるように、マシン上のスライダーを右に移動し、自動的に解離ステップの記録を開始するマシンのカバーを閉じます。解離ステップの記録が終了したら、機械のカバーを開き、ドロップホルダーとチューブホルダーを取り外します。両方を二重脱イオン水で十分に洗い流し、タンパク質を洗い流します。バイオセンサーを取り外し、安全に廃棄します。異なる検体濃度を用いて、同じリガンド検体対に対してこれらのステップを繰り返す。すべての実行が完了したら、画面左側にある [ファイル] と [実験を名前を付けて保存] をクリックして、データをソフトウェアに保存します。[データの実行] 見出しで、[ステップ補正] と [1 対 1 にフィット] を選択し、[分析] をクリックして運動データを生成します。定量的データをワークシートに抽出してグラフを生成するには、[CSVにエクスポート] をクリックして、記録したデータを CSV ファイルとして保存します。スプレッドシート ソフトウェアを使用して CSV ファイルを開きます。フル長GrgAは288アミノ酸から成っている。ここに示すように、28アミノ酸中間領域はシグマ28を直接結合する。ここでエンド末端Hisタグでタグ付けされた中間領域をリガンドとして使用し、これはまずニッケルNTAバイオセンサの先端に固定化した。リガンド結合の30秒前に開始し、最後の洗浄開始の2分後に終了する3つの異なる検体濃度を有する実験の記録が示されている。リガンドの抽出物の結合および解離の増強された可視化は、ベースラインの最初の2段階における値の除去およびリセット後に示される。非結合および末端Hisタグ付きGrgA 138〜165をバイオセンサーから洗浄した後、Sigma28の添加に続いて検体とのリアルタイムの関連付けを記録した。最後に、洗浄後にリアルタイム解離を記録した。BLIアッセイの前に、グリセロールはリガンドおよび検体から除去される。テキストで説明されているように、透析後すぐにBLIを実行することをお勧めします。BLIとの転写におけるタンパク質とタンパク質の相互作用を特徴付けた後、その相互作用が転写開始、伸長、または終了にどのように影響するかを調べることができます。
