De studie stelde vast dat de epidermis van de glabroushuid van de achterpoot van ratten gemakkelijk van dermis kon worden gescheiden met behulp van thermolysine (0,5 mG/ml) in PBS met 1 mM calciumchloride bij 4 °C gedurende 2,5 uur. Histologische evaluatie wees uit dat de epidermis gescheiden was van de dermis op het keldermembraan en dat de epidermale reteruggen intact waren. Evaluatie van inflammatoire mediatoren en neurotrofe factoren van de huid kan worden beperkt als gevolg van de heterogeniteit van celtypen gevonden in de ontstoken dermis en epidermis1,2,3. Verschillende enzymen, chemische, thermische of mechanische technieken waarbij de twee lagen worden gescheiden of voor het uitvoeren van celdissociatie voor evaluatie zijn onlangs herzien4. Zuur, alkali, neutraal zout en warmte kunnen de epidermis van dermis snel verdelen, maar cellulaire en extracellulaire zwelling treedt vaak op5,6. Trypsine, pancreatin, elastase, keratinase, collagenase, pronase, dispase en thermolysine zijn enzymen die zijn gebruikt voor epidermale-dermale scheiding4,7. Trypsine en andere proteolytische enzymen op grote schaal zijn actief bij 37-40 °C, maar moeten zorgvuldig worden gecontroleerd om dissociatie van epidermale lagen te voorkomen. Dispase splijt de epidermis bij de lamina densa, maar vereist 24 uur voor scheiding in de koude4,8 of kortere tijdspunten bij 37 °C4,9. Een beperkend kenmerk van al deze technieken is de mogelijke verstoring van weefselmorfologie en verlies van integriteit van mRNA en eiwitten.
Om de integriteit van mRNA en eiwitten te behouden, moet een huidscheidingsmethode gedurende een korte periode in de kou worden uitgevoerd. Bij het evalueren van huidscheidingstechnieken voor ontstekingsstudies is thermolysine een effectief enzym om de opperhuid van dermis te scheiden bij koude temperaturen4. Thermolysine is actief bij 4 °C, splijt epidermale hemidesmosomen van de lamina lucida en scheidt de epidermis van dermis binnen 1–3 h4,8,10. Het doel van dit rapport is om het gebruik van thermolysine te optimaliseren voor de scheiding van ontstoken rattenepidermis van dermis om mRNA- en eiwitniveaus voor ontstekingsmediators en neurotrofe factoren te detecteren. Er zijn verschillende voorlopige verslagen gepresenteerd11,12,13,14,15. Het doel van dit manuscript is om een optimale huidscheidingstechniek met thermolysine te beschrijven en de detectie van 1) markers van ontsteking, 2) interleukine-6 (IL-6) mRNA, en 3) zenuwgroeifactor (NGF) mRNA en eiwit in de epidermis van ratten met carrageen-geïnduceerde ontsteking (C-II)16,17. Een voorlopig rapport met behulp van het volledige Freund's adjuvansmodel geeft aan dat de NGF mRNA- en eiwitspiegels vroeg toenemen tijdens ontsteking15. Bij muizen veroorzaakt huidsensibilisatie met de topische toepassing van oxazolone een vroege stijging van het IL-6 mRNA met behulp van in situ hybridisatie36. Zowel IL-6 als NGF zijn betrokken bij C-II18,19, maar er zijn geen rapporten die mRNA- of eiwitniveaus voor IL-6 of NGF specifiek beschrijven vanuit de epidermis tijdens de acute stadia van C-II.
De thermolysinetechniek is goedkoop en eenvoudig uit te voeren. Bovendien maakt thermolysinescheiding van de epidermis van dermis mRNA, westelijke vlek en immunohistochemische analyse van ontstekingsmediators en neurotrofe factoren tijdens het ontstekingsprocesmogelijk 15. Onderzoekers moeten deze techniek gemakkelijk kunnen gebruiken in zowel preklinische als klinische studies naar huidontsteking.

Eenvoudig te gebruiken en goedkope technieken zijn nodig om de site-specifieke productie van ontstekingsmediators en neurotrofinen tijdens huidletsel, ontsteking en / of sensibilisatie te bepalen. Het doel van deze studie is om een epidermale-dermale scheidingsprotocol te beschrijven met behulp van thermolysine, een proteïnase dat actief is bij 4 °C. Om deze procedure te illustreren, worden Sprague Dawley-ratten verdoofd en worden rechterachterpoten geïnjecteerd met carrageen. Zes en twaalf uur na injectie worden ratten met ontstekingen en naïeve ratten geëuthanaseerd en een stukje achterpoot, glabroushuid wordt in het gemodificeerde adelaarsmedium van koude Dulbecco geplaatst. De epidermis wordt vervolgens in het keldermembraan van de dermis gescheiden door thermolysine in PBS met calciumchloride. Vervolgens wordt de dermis vastgezet door microdissectie-tang en wordt de epidermis voorzichtig weg geplaagd. Toluidine blauwe kleuring van weefselsecties tonen aan dat de epidermis netjes wordt gescheiden van de dermis op het keldermembraan. Alle keratinocytcellagen blijven intact en de epidermale reteruggen samen met inkepingen van dermale papillen worden duidelijk waargenomen. Kwalitatieve en real-time RT-PCR wordt gebruikt om zenuwgroeifactor en interleukine-6 expressieniveaus te bepalen. Westerse blotting en immunohistochie worden eindelijk uitgevoerd om hoeveelheden zenuwgroeifactor te detecteren. Dit rapport illustreert dat koude thermolysinevertering een effectieve methode is om epidermis van dermis te scheiden voor evaluatie van mRNA en eiwitveranderingen tijdens ontsteking.

Carrageeninjectie in de achterpoot van de rat veroorzaakte klassieke symptomen van ontsteking zoals roodheid en oedeem16,17. De zwelling van de achterpoot werd gemeten met mechanische remklauwen20. Hier wordt een protocol gepresenteerd voor de scheiding van epidermis van dermis om de productie van ontstekingsmediators te evalueren. Na ontsteking wordt de epidermis van de achterpoot van de rat gescheiden door thermolysine bij 4 °C. Ons protocol is een gebruiksvriendelijke en goedkope epidermale-huidscheidingstechniek voor het bepalen van de plaatsspecifieke productie van ontstekingsmediators en neurotrofinen tijdens huidontsteking. Het belangrijkste voordeel van deze techniek is dat enzymatische scheiding van de epidermis van de dermis wordt uitgevoerd bij vier graden Celsius, waardoor de integriteit van mRNA en eiwit behouden blijft. Deze methode geeft inzicht in de soorten mediatoren die primaire afferente terminals sensibiliseren bij acute en chronische ontstekingen voor de ontwikkeling van nieuwe therapeutische interventies.Wondgenezing, huidaandoeningen, chronische wonden en brandwonden kunnen baat hebben bij dit onderzoek. En deze methode kan worden toegepast op andere epitheeloppervlakken, zoals het GI-kanaal en het hoornvlies. De optimale scheidingstijd kan enigszins verschillen tussen labs.Een voorlopig experiment waarbij verschillende tijdspunten worden vergeleken in combinatie met een 2D-morfologie-evaluatie wordt aanbevolen om de kwaliteit van de scheiding te bepalen. Na het bevestigen van een gebrek aan reactie op pedaalreflex bij een verdoofde acht tot negen weken oude Sprague Dawley rat, subcutaan injecteren van de rechter glabrous achterpoot met 100 microliters van 1X Lambda carrageen verdund in PBS. Gebruik remklauwen om de achterpootmetatarsale dikte vlak voor en op 6 en 12 uur na injectie te meten om het volume oedeem als reactie op de stimulus te bepalen.Gebruik op het juiste experimentele eindpunt een scherpe scalpel om een stuk glabrousachterpoothuid van één bij twee millimeter van de geïnjecteerde poot te oogsten en gebruik microdissectie-tang om de huid gedurende 15 tot 60 minuten in een microcentrifugebuis op ijs over te brengen in één milliliter koude DMEM. Terwijl het weefsel aan het broeden is, voeg je een milliliter vers bereide geactiveerde thermolysine toe aan 10 putten van een 24-put celkweekplaat op ijs. Gebruik aan het einde van de incubatie de microdissection forceps om één huidmonster over te brengen in elke put van geactiveerd thermolysine, stratum corneum kant naar boven zonder de monsters in de oplossing onder te dompelen.Het is van cruciaal belang dat de huid niet wordt ondergedompeld in de thermolysine en dat het stratum corneum naar boven kijkt om een effectieve scheiding te bereiken. Gebruik na twee tot drie uur de microdissection forceps om één huidmonster in zeven tot acht milliliter van vier graden Celsius DMEM onder te dompelen in een Petrischaaltje om meer ruimte te geven voor de opperhuid om zich te scheiden van de dermis. Gebruik een tang om de opperhuid voorzichtig rond de omtrek van de huid te borstelen totdat de bijna doorschijnende epidermis aan de randen van het monster wordt waargenomen.Wanneer de opperhuid merkbaar van de dermis scheidt, pak dan zorgvuldig elke huidlaag met een enkele tang vast en trek langzaam de epidermis uit de dermis, evalueer vervolgens de doorschijnendheid van de geïsoleerde epidermis door middel van lichte microscopie om ervoor te zorgen dat deze optisch consistent is. Als de lagen goed zijn gescheiden, plaatst u de epidermale en huidweefsels gedurende 30 minuten in afzonderlijke containers van vijf millimolaire EDTA in DMEM bij vier graden Celsius. Na thermolysinedeactivering een deel van de epidermis gedurende een uur bij kamertemperatuur fixeren met agitatie.Plaats aan het einde van de incubatie het vaste weefsel gedurende een uur in PBS bij kamertemperatuur met agitatie, voordat u het monster bevriest in een geschikte weefselinbeddingsmatrix voor doorsnede. Gebruik een cryostaat om 14 micrometer dikke doorsneden te verkrijgen en monteer de secties op met gelatine bedekte glazen microscoopglaasjes. Bevlek de monsters na het drogen op een schuifwarmer gedurende 90 seconden met een werkende oplossing van toluidineblauw.Was daarna voorzichtig toluidineblauw van de dia's met PBS en verzet u tegen afdeklips met een waterig montagemedium en observeer vervolgens de epidermis met Brightfield-microscopie bij een vergroting van 50 tot 250x. Carrageeninjectie in de achterpoot van de rat veroorzaakt klassieke symptomen van ontsteking, zoals roodheid en oedeem bij zowel 6 als 12 uur na injectie. Brightfield microscopie kan worden gebruikt om de effectiviteit van thermolysinescheiding van de epidermis en dermis van de achterpoothuid van de rat glabrous te evalueren.Inderdaad, toluidine blauwe kleuring van de epidermale doorsneden toont aan dat de epidermis is gescheiden van de dermis op het basismembraan, maar dat de epidermale Rete-ruggen en de inkepingen van de huidpapilla intact blijven. Verder worden alle keratinocytcellagen waargenomen. Westerse blotting van de thermolysine gescheiden epidermis produceert consistente resultaten, wat wijst op stabiele eiwitniveaus tijdens de techniek bij vier graden Celsius.Zeer weinig NGF-eiwit wordt gedetecteerd in de naïeve rattenepidermis, maar de NGF-eiwitniveaus zijn aanzienlijk geherreguleerd na zes uur carrageen-geïnduceerde ontsteking. 12 uur na de injectie worden de NGF-spiegels verlaagd in vergelijking met de NGF-spiegels die na zes uur worden waargenomen, maar blijven ze verhoogd ten opzichte van de controles. Zoals verwacht uit de westerse vlekanalyse, wordt NGF-immunoreactiviteit niet gedetecteerd bij naïeve controle-epidermisweefselmonsters, maar na zes uur na carrageen-geïnduceerde ontsteking wordt NGF-immunoreactiviteit waargenomen in de meeste keratinocyten van het stratum granulosum en stratum lucidum.Een paar cellen in het stratum spinosum zijn ook NGF immunoreactief. Bij het gebruik van thermolysine om de intacte epidermis te isoleren, is het belangrijk om te onthouden dat de optimale scheidingstijd empirisch moet worden bepaald door de eindgebruiker. Verschillende proteomische en moleculaire technieken kunnen worden gebruikt om de expressie van specifieke eiwitten en/of primaire gentranscripties te valideren om te bevestigen dat deze procedure de basale expressie niet verandert.Deze techniek van epidermale-dermale scheiding stelt onderzoekers in staat om aanvullende vragen te beantwoorden met betrekking tot de site-specifieke expressie van neurotrofinen en ontstekingsmediators in verschillende cutane ontstekingsmodellen. Picrinezuur en paraformaldehyde zijn gevaarlijk. Werk met beide chemicaliën in een zuurkast, gebruik persoonlijke beschermende kleding en handschoenen en was je gezicht en handen na het hanteren.
