Bijgevolg, gut microbiota zijn onlangs uitgegroeid tot een belangrijk doelwit voor ziektepreventie en therapie1. Bovendien, gut bacteriën interactie met gastheer intestinale cellen en reguleren fundamentele gastheer processen, met inbegrip van metabole activiteiten, voedingsstoffen availabiliteiten, immuun systeem modulatie, en zelfs hersenfunctie en besluitvorming van2,3 . Endobiotica hebben aanzienlijke potentie om invloed op de bacteriële samenstelling en de diversiteit van gut microbiota. Zo, interacties tussen endobiotica en menselijke gut microbiota hebben aangetrokken toegenomen onderzoek aandacht4,5,6,7,8,9.
Het is moeilijk om de interacties tussen endobiotica en humane gut microbiota in vivo te evalueren als gevolg van bio-ethiek en economische beperkingen. Bijvoorbeeld, experimenten onderzoeken de interacties tussen endobiotica en menselijke gut microbiota kan niet worden uitgevoerd zonder toestemming van de Food and Drug Administration, en werving van vrijwilligers is duur. Daarom worden voor dergelijke onderzoeken vaak diermodellen gebruikt. Het gebruik van diermodellen is echter beperkt als gevolg van verschillende microbiota-composities en diversiteit in dier-vs. mensen-geassocieerde Gemeenschappen. Een alternatieve in vitro methode om de interacties tussen endobiotica en humane gut microbiota te onderzoeken is door het gebruik van batch cultuur experimenten.
Exopolysacchariden (EPSs) zijn prebiotica die significant bijdragen aan het behoud van de gezondheid van de mens10. Afzonderlijke EPSS die bestaan uit verschillende Monosacharide composities en structuren kunnen verschillende functies vertonen. Eerdere analyses hebben de samenstelling van BIF EPSs bepaald, de representatieve xenobiotische die in de huidige studie11is getarget. Echter, gastheer-geassocieerde metabole effecten zijn niet overwogen met betrekking tot EPS samenstelling en diversiteit.
Het hier beschreven protocol gebruikt de fecale microbiota van 12 vrijwilligers tot gisten BIF EPSS. Thin-layer chromatografie (TLC), 16S rRNA Gene High-throughput sequencing en gaschromatografie (GC) worden vervolgens gebruikt in combinatie om de interacties tussen EPSs en humane gut microbiota te onderzoeken. analysis of interactions between endobiotics and human gut microbiota using in vitro bath fermentation systems

Menselijke intestinale micro-organismen zijn onlangs uitgegroeid tot een belangrijk doelwit van onderzoek bij het bevorderen van de menselijke gezondheid en het voorkomen van ziekten. Bijgevolg zijn onderzoeken naar interacties tussen endobiotica (bijv. drugs en prebiotica) en gut microbiota een belangrijk onderzoeksonderwerp geworden. In vivo experimenten met menselijke vrijwilligers zijn echter niet ideaal voor dergelijke studies als gevolg van bio-ethiek en economische beperkingen. Als gevolg hiervan zijn diermodellen gebruikt om deze interacties in vivo te evalueren. Niettemin, diermodel studies zijn nog steeds beperkt door bio-ethiek overwegingen, naast de verschillende samenstellingen en diversiteit van microbiota bij dieren vs. mensen. Een alternatieve onderzoeksstrategie is het gebruik van batch fermentatie-experimenten die het mogelijk maken de interacties tussen endobiotica en gut microbiota in vitro te evalueren. Om deze strategie te evalueren, werden bifidobacteriële (BIF) exopolysacchariden (EPS) gebruikt als representatief xenobioticum. Vervolgens werden de interacties tussen BIF EPS en humane gut microbiota onderzocht met behulp van verschillende methoden zoals Thin-layer chromatografie (TLC), bacteriële compositorische analyse met 16S rRNA Gene High-throughput sequencing en gaschromatografie van korte-keten vetzuren (SCFAs). Hier gepresenteerd is een protocol om de interacties tussen endobiotica en menselijke gut microbiota met behulp van in vitro batch fermentatiesystemen te onderzoeken. Belangrijk is dat dit protocol ook kan worden aangepast om de algemene interacties tussen andere endobiotica en gut microbiota te onderzoeken.

De productie van mucoïde EPS kan worden waargenomen in B. longum culturen op pyg platen na anaerobe incubatie voor 72 h (Figuur 1a). Centrifugeren van cultuur resten, gevolgd door ethanol precipitatie en drogen, resulteerde in de inzameling van cellulose-achtige EPS (Figuur 1b). Ons protocol kan op grote schaal worden gebruikt voor hoge doorvoer en snelle screening van de interacties tussen endobiotica en menselijke darmmicrobiota. Lage kosten en het vermijden van interferentie-effecten van het metabolisme van de gastheer. Deze procedure heeft het potentieel om te worden gebruikt bij de diagnose.Los eerst BIF EPS en zetmeel individueel op in heet water op een magnetische agitator om zowel de concentratie van acht gram per liter te maken. Voeg in het geprepareerde basiscultuurmedium VI 100 milliliter van de BIF EPS-oplossing toe om cultuur VI BIF-oplossing te maken. Voeg 100 milliliter van de zetmeeloplossing toe om cultuur VI zetmeeloplossing te maken.Autoclave de twee media met koolstofbron, en het medium zonder koolstofbron als een controle, op 121 graden Celsius gedurende 15 minuten. Laat ze afkoelen tot kamertemperatuur en voeg hooimannen en cysteïne toe. Breng een submonster van vijf milliliter van elk medium naar kweekbuizen over.En bewaar in een anaerobe couveuse. Bewaar de resterende media op vier graden Celsius. Om menselijk fecal monster voor te bereiden, breng je een gram vers fecale monster over in 10 milliliter anaërobe PBS bij pH zeven in een glazen bekerglas.Gebruik dan een glazen staaf om het te roeren om een drijfmest te bereiken. Voeg in een anaerobe kamer 500 microliter van de gefilterde fecale drijfmest individueel toe aan de drie bereide fermentatiemedia. Gebruik dan een föhn met hete lucht te drogen.Dompel het ene uiteinde van de bemonsterde platen onder in 20 milliliter mierenzuur:n-butanol:wateroplossing gedurende enkele minuten, totdat de elektroforese zich bijna tot het andere uiteinde uitstrekt. Neem vervolgens de platen met tangen, en droog met een föhn. Vervolgens weken de platen in de orcinol reagens voor twee minuten te verven.En droog weer. Breng de platen op 120 graden Celsius naar een bakoven om drie minuten te verwarmen. Neem vervolgens foto's van de plaat om de afbraak van EPS te evalueren door TLC-banden te meten.Om de effecten van EPS op de productie van SCFA te bestuderen, voeg een milliliter van de gefermenteerde supernatants toe aan twee milliliter centrifugebuizen en voeg 0,2 milliliter van 25 gewicht/volumecentidruk aan elk monster toe. Vortex om de oplossingen grondig te mengen. Centrifugeren de mengsels op 13.000 keer G gedurende 20 minuten.In de tussentijd, oplossingen voor te bereiden van 120 millimolar azijn, propionische, butyrische, isobutyrische, valeriische en isovalerische zuren in buizen. Gebruik een spuit uitgerust met 0,22 micron filter om de supernatants te filteren in nieuwe buizen voor gaschromatografie analyse. In deze studie werd de productie van mucoid EPS waargenomen in bifidobacterium longum culturen op PYG-platen, na anaerobe incubatie gedurende 72 uur.Centrifugatie van cultuurschaafwonden, gevolgd door ethanolneerslag en drogen, resulteerde in de inzameling van cellulose-achtige EPS. Uit TLC-analyse bleek dat de afbraaksnelheid van zetmeel door menselijke fecale microbiota sneller was dan die van BIF EPS. Principal Coordinate Analysis toont cultuur VI BIF, en cultuur VI zetmeel groepen, afzonderlijk van elkaar geclusterd, wat aangeeft dat EPS en zetmeel beschikbaarheid differentieel vorm menselijke fecale bacteriële gemeenschappen.Lineaire discriminant Analyse op effectgrootte toont de genera collinsella, coprococcus, parabacteroides, en rhodopseudomonas waren aanzienlijk overvloediger in de cultuur VI BIF monsters dan in de cultuur VI zetmeel monsters. Bovendien, GC metingen tonen SCFAs die werden gemeten aan de hand van fermentatie culturen opgenomen azijn, propionische, isobutyrische, butyrische, isovalerische, en valeriaanzuren. Na gisting gedurende 24 uur en 48 uur, vijf van de zes SCFA concentraties waren vergelijkbaar en niet statistisch verschillend tussen de cultuur VI BIF, cultuur VI zetmeel, en cultuur VI groepen.Echter, propriche zuurconcentraties waren aanzienlijk hoger in de cultuur VI BIF groep dan in de cultuur VI zetmeel groep, na 48 uur van fermentatie.
