Le microbiote intestinal joue un rôle important dans le fonctionnement des intestins humains et dans la santé de l'hôte. Par conséquent, le microbiote intestinal est récemment devenu une cible importante pour la prévention et la thérapie des maladies1. En outre, les bactéries intestinales interagissent avec les cellules intestinales hôtes et régulent les processus fondamentaux de l'hôte, y compris les activités métaboliques, les disponibilités nutritives, la modulation du système immunitaire, et même la fonction cérébrale et la prise de décision2,3 . Les endobiotiques ont un potentiel considérable pour influencer la composition bactérienne et la diversité du microbiote intestinal. Ainsi, les interactions entre les endobiotiques et le microbiote intestinal humain ont attiré l'attention croissante de la recherche4,5,6,7,8,9.
Il est difficile d'évaluer in vivo les interactions entre les endobiotiques et le microbiote intestinal humain en raison de la bioéthique et des contraintes économiques. Par exemple, des expériences sur les interactions entre les endobiotiques et le microbiote intestinal humain ne peuvent être réalisées sans la permission de la Food and Drug Administration, et le recrutement de volontaires coûte cher. Par conséquent, les modèles animaux sont souvent utilisés pour de telles enquêtes. Cependant, l'utilisation de modèles animaux est limitée en raison de différentes compositions de microbiotes et de la diversité dans les communautés associées aux animaux par rapport à l'homme. Une autre méthode in vitro pour explorer les interactions entre les endobiotiques et le microbiote intestinal humain est l'utilisation d'expériences de culture par lots.
Les exopolysaccharides (EPS) sont des prébiotiques qui contribuent de manière significative au maintien de la santé humaine10. Les EPS distincts qui se composent de différentes compositions et structures monosaccharides peuvent présenter des fonctions distinctes. Des analyses antérieures ont déterminé la composition des EPS Bif, qui sont le xénobiotique représentatif visé dans la présente étude11. Cependant, les effets métaboliques associés à l'hôte n'ont pas été pris en considération en ce qui concerne la composition et la diversité de l'EPS.
Le protocole décrit ici utilise le microbiote fécal de 12 volontaires pour fermenter les EPS Bif. La chromatographie à couches minces (TLC), le séquençage à haut débit du gène 16S rRNA et la chromatographie gazeuse (GC) sont ensuite utilisés en combinaison pour étudier les interactions entre les SPE et le microbiote intestinal humain. Les avantages distincts de ce protocole par rapport aux expériences in vivo sont son faible coût et l'évitement des effets d'interférence du métabolisme de l'hôte. Les micro-organismes intestinaux humains sont récemment devenus une cible importante de la recherche dans la promotion de la santé humaine et la prévention des maladies. Par conséquent, les études sur les interactions entre les endobiotiques (p. ex., les médicaments et les prébiotiques) et le microbiote intestinal sont devenues un sujet de recherche important. Cependant, les expériences in vivo avec des volontaires humains ne sont pas idéales pour de telles études en raison de la bioéthique et des contraintes économiques. En conséquence, des modèles animaux ont été utilisés pour évaluer ces interactions in vivo. Néanmoins, les études de modèles animaux sont encore limitées par des considérations de bioéthique, en plus de compositions et de diversités différentes du microbiote chez les animaux par rapport aux humains. Une autre stratégie de recherche est l'utilisation d'expériences de fermentation par lots qui permettent d'évaluer les interactions entre les endobiotiques et le microbiote intestinal in vitro. Pour évaluer cette stratégie, les exopolysaccharides bifidobactériens (Bif) ont été utilisés comme xénobiotique représentatif. Ensuite, les interactions entre Bif EPS et le microbiote intestinal humain ont été étudiées à l'aide de plusieurs méthodes telles que la chromatographie à couches minces (TLC), l'analyse compositionnelle de la communauté bactérienne avec le séquençage à haut débit du gène rRNA 16S et la chromatographie gazeuse. d'acides gras à chaîne courte (SCFA). Notre protocole peut être largement utilisé pour le criblage élevé et rapide des interactions entre l’endobiotics et le microbiote humain d’intestin. Cette procédure a le potentiel d’être employée dans le diagnostic.Tout d’abord, dissoudre BIF EPS et amidon individuellement dans l’eau chaude sur un agitateur magnétique pour faire à la fois la concentration de huit grammes par litre. Dans le milieu de culture de base préparé, VI, ajouter 100 millilitres de la solution BIF EPS pour faire de la culture VI solution BIF. Ajouter 100 millilitres de la solution amidon pour faire de la culture VI solution d’amidon.Autoclave les deux médias avec source de carbone, et le milieu sans source de carbone comme un contrôle, à 121 degrés Celsius pendant 15 minutes. Dans une chambre anaérobie, ajouter 500 microlitres de boue fécale filtrée dans les trois supports de fermentation préparés, individuellement. Prenez ensuite des photos de la plaque pour évaluer la dégradation de l’EPS en mesurant les bandes TLC.Pour étudier les effets de l’EPS sur la production de SCFA, ajoutez un millilitre des supernatants fermentés aux tubes de centrifugeuse de deux millilitres et ajoutez 0,2 millilitres de 25 poids/volume pour cent d’acide métaphosphorique à chaque échantillon. Vortex pour bien mélanger les solutions. Centrifuger les mélanges à 13 000 fois G pendant 20 minutes.En attendant, préparez des solutions d’acides acétiques, propioniques, butyriques, isobutyriques, valérianes et isovaleriques de 120 millimolaires dans des tubes. Ajouter ensuite un millilitre de chaque acide préparé à un tube contenant 1,2 millilitres d’acide métaphosphorique de 25 poids/volume pour cent. Et chargez-les dans la bouteille d’échantillon de l’instrument de chromatographie gazeuse comme cocktails standard.Sortez les tubes de la centrifugeuse et transférez les supernatants dans des tubes frais. Utilisez une seringue équipée d’un filtre de 0,22 micron pour filtrer les supernatants dans de nouveaux tubes pour l’analyse gaz-chromatographie. Dans cette étude, la production d’EPS mucoïde a été observée dans les cultures de longum de bifidobacterium sur des plaques de PYG, après incubation anaérobie pendant 72 heures.La centrifugation des éraflures de culture, suivie des précipitations d’éthanol et du séchage, a entraîné la collecte d’EPS de cellulose. L’analyse de TLC a indiqué que le taux de dégradation de l’amidon par microbiote fécal humain était plus rapide que celui du BIF EPS. L’analyse principale des coordonnées montre que la culture VI BIF et les groupes d’amidon de culture VI, regroupés séparément les uns des autres, indiquent que la disponibilité de l’EPS et de l’amidon façonne différemment les communautés bactériennes fécales humaines.L’analyse discriminante linéaire sur la taille d’effet montre que les genres collinsella, coprococcus, parabactérides, et rhodopseudomonas étaient sensiblement plus abondants dans les échantillons de BIF de culture VI que dans les échantillons d’amidon de culture VI. En outre, les mesures gc montrent SCFAs qui ont été mesurés à partir des cultures de fermentation inclus acétique, propionique, isobutyrique, butyrique, isovaleric, et les acides valeric. Après fermentation pendant 24 heures et 48 heures, cinq des six concentrations de SCFA étaient semblables et non statistiquement différentes entre la culture VI BIF, l’amidon de culture VI, et les groupes de culture VI.Cependant, les concentrations d’acide proprionique étaient significativement plus élevées dans le groupe de la culture VI BIF que dans le groupe d’amidon de culture VI, après 48 heures de fermentation. Faites cette expérience dans des conditions anaérobies, puis transférez les échantillons fixes dans un incubateur anaérobie dès que possible.
