このプロトコルは、小穀物作物におけるEMS突然変異誘発による突然変異頻度の高い堅牢な耕作集団の開発を詳述する。TILLING集団は、機能的ゲノミクス、ならびに小さな穀物作物における前方遺伝的空間遺伝子発見に使用することができる。この技術を用いて開発されたTILLING集団は、高い突然変異頻度を有し、このプロトコルは任意の遺伝子型に適用することができる。Cel-1アッセイベースの変異検出は、基本的なラボ機器を使用して行うことができます。最も重要なステップは最適なEMS濃度を決定することです。したがって、EMS投与曲線は、個々の集団のために特別に行われるべきである。まず、50ミリリットルの蒸留水を含む6つの250ミリリットルガラスフラスコのそれぞれに目的の遺伝子型を持つ100種子を浸します。100 RPMで室温で8時間振ってインビビションを行います。次に、ヒュームフードに、EMS液と蒸留水を混合して5つの異なる濃度のEMS溶液を50ミリリットル調製する。インビビションの後、5つのフラスコから水をデカントします。インビベッド種子を含む5つのフラスコのそれぞれに50ミリリットルのEMS溶液を加えます。75 RPMと室温でフラスコを16時間振ります。EMS溶液を空の廃棄物ボトルにデカントし、処理した種子を空の廃棄物ボトルに置いたチーズクロスに注ぎ、各処理のために別々に収集します。種子の注ぎ込みに余分な水を使用してください。また、汚染されたフラスコの壁にEMS不活性化溶液の数ミリリットルをスプレーします。そして、24時間溶液中に使用されるピペットの先端を浸漬する。24時間治療するためにEMS不活性化溶液の1つのボリュームを追加することにより使用EMS溶液を非アクティブ。ひねりを加えたネクタイを使用して、EMS処理した種子をチーズクロスの中に保持し、水道水の下で2時間洗浄します。洗浄後、各種をポッティング土壌を含む根トレーナーに個別に移植します。16時間の光の期間のために摂氏20〜25度で植物を育てます。移植の15日後、植物をチェックし、発芽に失敗した種子の数を数えます。植物の生存のデータを記録します。生存率が40〜60%以内でない場合、変異生成実験後に40〜60%の所望の致死率を達成するまで、改変濃度で第2ラウンドの投薬量最適化を行い、96ウェル組織採取ボックスにM2植物の葉組織を採取する。組織は、自家型M1植物から成長したM2植物から採取される。各M1プラントから1つのM2プラントが示されています。メーカーの推奨に従って、DNA精製システムを用いた植物DNA抽出キットを使用してDNAを抽出します。その後、DNA抽出物の2マイクロリットルを16スロットのLVプレートにロードします。260ナノメートルと280ナノメートルの波長で分光光度計を使用してDNAを定量化します。DNA濃度をヌクレアーゼを含まない水で1マイクロリットル当たり25ナノグラムに希釈する。各サンプルの行と列の同一性を維持しながら、4つの96ウェルブロックの各井戸のDNAをプールプレートに組み合わせることで、200マイクロリットルの4倍のDNAプールを作成します。次に、各チューブPCRバッファー、フォワードおよびリバースプライマー、および原稿に従ったDNAポリメラーゼに添加することにより、遺伝子特異的プライマー用のPCRマスターミックスチューブを調製する。アリコートマスターは96ウェルPCRプレートのウェルに混入する。次に、プールされた DNA テンプレートを追加します。PCR チューブをサーマルサイクラーにロードし、タッチダウン プロファイルを使用してサーマルサイクラーで PCR 反応を実行します。不一致のDNA間のヘテロ二重鎖を生成するには、別のプログラムでサーマルサイクラー中のPCR産物をインキュベートします。次に、ヘテロデュプレックスPCR産物のそれぞれに自家製Cel-1エンドヌクレアーゼを2.5マイクロリットル加えます。摂氏45度で45分間インキュベートします。その後、pH 8で0.5モルEDTAの2.5マイクロリットルを加えてCel-1反応を終了する。各Cel-1処理物の30.5マイクロリットルを3%アガロースゲルに5マイクロリットルの染料と混合し、100ボルトで2時間半稼働します。突然変異プールをデコンボルテするには、最初の反応に2.5マイクロリットルのM2 DNAと2.5マイクロリットルの野生型DNAが含まれ、第2の反応にはM2 DNAの5マイクロリットルしか含まれなかった個々のM2 DNAに対して2つのPCR反応を実行することによって、突然変異体の接合性を決定する。このプロトコルは、小穀物作物のEMS変異体化と変異体の特徴を示す。投薬曲線は、小麦の3つの異なる種のための所望の50%生存率のための最適なEMS用量を示す。M2集団における容易に同定可能な型の存在は、小さい穀物集団における突然変異の有効性を確認する。ここでは、M2 TILLING集団における変異型の4つの例、大麦M2集団におけるアルビノ変異体、大麦M2集団におけるクロリーナ突然変異体、アエギロプスタウシイM2集団におけるピンク色変色を有する可変変異体、およびトリチカムモノコクカムM2集団における低耕耕変異体の4つの例を示す。アガロースゲルプラットフォーム上でCel-1を使用した変異体同定は、ユニークな切断バンドによって12の突然変異プールから同定される潜在的な突然変異プールを示す。変異プールの破壊は突然変異の接合性を決定し、個々のサンプルへの突然変異を追跡するのに役立った。A4プールにはヘテロ接合性突然変異があり、ボックス4とボックス4の両方の野生型DNAサンプルで切断されたバンドによって示された。H5プールには、ユニークな切断バンドがBox 5 H5野生型DNAサンプルにのみ存在していたため、ホモ接合性突然変異が含まれていました。最も重要なステップは、40〜60%の致死率を達成するためにEMSの最適濃度を決定することです。一度高い突然変異頻度を有するTILLING集団が開発された後、それは関心のある任意の遺伝子の機能的特徴付けに使用することができる。さらに、人口は作物改善のための有用な遺伝的変異の源であり得る。TILLINGは、モデルおよび作物植物の機能的ゲノム研究のために広く使用されています。得られる突然変異の範囲は、誤った結果、ノックアウト、サイレント、あるいはミスプリケート形態である可能性があります。EMSはミュータゲンです。そのため、EMSを取り扱う際には、適切な個人用保護具を使用してください。残った溶液を除染し、ピペットの先端を使用した。
