ويبين البروتوكول المعروض استخدام الخلايا الاوليه من الأشخاص المصابين بفيروس نقص المناعة الذاتية كنموذج لدراسة البويضات والضامات. فيروس نقص المناعة الذاتية + المرضي الذين يخضعون لعربه العيش مع العدوى لسنوات متعددة ، ويمكن أيضا ان يكون لها آخر العدوى المشتركة ذات الصلة جهاز المناعة...

تسبب عدوي فيروس نقص المناعة البشرية-1 (HIV-1) خللا مناعيا شديدا ، وهو ما يمكن ان يؤدي إلى الاصابه بالعدوى الانتهازية ومتلازمة عوز المناعة المكتسب (الإيدز). علي الرغم من ان المرضي المصابين بفيروس نقص المناعة التي تمر بالعربة تتميز الأحمال الفيروسية المنخفضة والعد العادية الخلايا التائية + T ، ويمكن المساس بأداء الجهاز المناعي في هؤلاء الافراد ، مما يؤدي إلى استجابه مناعية مختلة التي ارتبطت بزيادة خطر الاصابه بالسرطان1 أليات الخلل المناعي في مرضي فيروس نقص المناعة الذاتية علي السلة لا تزال غير معروفه إلى حد كبير. ولذلك ، فان توصيف الخلايا المناعية المستمدة من المريض والتحقيق في علم الاحياء والوظيفة عنصر حاسم في البحوث الحالية بفيروس نقص المناعة المكتسب.
الخلايا الاحاديه والضامة هي المنظمين الرئيسيين للاستجابات المناعية وتلعب أدوارا أساسيه في الاصابه بفيروس نقص المناعة الذاتية2,3,4,5. غير متجانسة والبلاستيك في الطبيعة ، ويمكن تصنيف الضامة علي نطاق واسع في تنشيط كلاسيكي (M1) أو بدلا من ذلك تنشيط (M2). في حين ان هذا التصنيف العام ضروري عند اعداد الشروط التجريبية ، قد يتم عكس حاله الاستقطاب من الضامة من قبل مجموعه متنوعة من السايتوكينات6،7،8،9. علي الرغم من ان العديد من الدراسات قد حققت في اثار الاصابه بفيروس نقص المناعة الذاتية علي الخلايا الاحاديه والجذعية ، والتفاصيل الجزيئية للاستجابات بوساطة البويضات غير معروفه إلى حد كبير6،7،10،11،12،13،14،15،16،17،18،19 من بين العوامل التي تنطوي عليها تنظيم الخلايا المناعية ووظيفتها ، ميكرورناس (miRNAs) ، قصيرة غير الترميز RNAs ان ما بعد الهيستونات تنظيم التعبير الجيني ، وقد ثبت ان تلعب دورا هاما في سياق المسارات الخلوية الرئيسية (اي النمو ، والتمايز ، والتنمية ، والمبرمج)20. وقد وصفت هذه الجزيئات بأنها منظمات هامه من عوامل النسخ الاساسيه لإملاء الاستقطاب وظيفية من الضامة21. وقد تم التحقيق في الدور المحتمل لmirnas في الخلايا الاحاديه من الافراد المصابين بفيروس نقص المناعة الذين يخضعون لعربه التسوق ، ولكن التقدم في هذا المجال يتطلب المزيد من العمل22،23،24،25،26. تناقش هذه الورقة طريقه محسنه لتحويل miRNAs و siRNAs إلى خلايا بشريه أساسيه من المرضي المصابين بفيروس نقص المناعة البشرية والضوابط.
يعتمد هذا البروتوكول علي الكواشف ومجموعات المواد المتاحة تجاريا ، لان الاستمرارية في الإجراءات التقنية تساعد علي أزاله المتغيرات التجريبية غير الضرورية عند العمل مع العينات السريرية. ومع ذلك ، فان الطريقة توفر نصائح هامه (اي عدد الخلايا المطلية أو الحاضنة القصيرة مع وسائل الاعلام الخالية من المصل لتعزيز التصاق الخلايا بالصفيحة). الاضافه إلى ذلك ، فان شروط الاستقطاب المستخدمة في هذا البروتوكول مستمده من الاعمال المنشورة27و28و29.

ولا يزال فيروس نقص المناعة البشرية أحد الشواغل الصحية الرئيسية علي الرغم من إدخال العلاج المضاد للفيروسات العكوسه في منتصف التسعينيات من القرن العشرين. في حين ان العلاج المضاد للفيروسات الرجعية يخفض بكفاءة الحمل الفيروسي الجهازي ويستعيد العد الطبيعي للخلاياالتائية ، فانه لا يعيد تشكيل جهاز مناعي وظيفي تماما. يمكن ان يتميز جهاز المناعة المختل في الافراد المصابين بفيروس نقص المناعة الذين يخضعون لعربه التسوق بالتنشيط المناعي ، والشيخوخة المبكرة للخلايا المناعية ، أو التهاب المستمر. وتضيف هذه الشروط ، إلى جانب العوامل المصاحبة للاصابه بالفيروس ، تعقيدا إلى المرض ، الذي لا يمكن استنساخه بسهوله في النماذج الخلوية والحيوانية. للتحقيق في الاحداث الجزيئية الكامنة وراء الخلل المناعي في هؤلاء المرضي ، يتم عرض نظام للثقافة والتلاعب بالخلايا الوحيدة الاساسيه البشرية في المختبر هنا. وعلي وجه التحديد ، يسمح البروتوكول للثقافة والانتقال من CD14 الابتدائية + مونوسيدات التي تم الحصول عليها من الافراد المصابين بفيروس نقص المناعة المكتسب الذين يخضعون لعربه التسوق وكذلك من الضوابط السلبية لفيروس نقص المناعة المكتسب. وتشمل هذه الطريقة العزلة والثقافة والانتقال من الخلايا الاحاديه والضامة المشتقة من البويضات. في حين يتم استخدام مجموعات المتاحة تجاريا والكواشف, البروتوكول يوفر نصائح هامه والظروف المثلي للانضمام الناجحة والانتقال من الخلايا الاحاديه مع محاكاة ميرنا ومثبطات وكذلك مع siRNAs.

وتسبب الإصابة بفيروس نقص المناعة البشرية خللاً في المناعة حتى لدى الأفراد الذين يخضعون للعلاج المضاد للفيروسات العكوسة دون وجود فيروس يمكن اكتشافه. هذه الطريقة تسمح لدراسة وظيفة أحادية التي هي المنظمين الرئيسية للاستجابة المناعية. لقد قمنا بتحسين هذه التقنية لعزلة وثقافة وtransfection من أحاديات الإنسان الأولية.نحن نستخدم هذه الطريقة لفهم الآليات الجزيئية للخلل المناعي لدى الأفراد المصابين واستخدام ماصة لرسم محتويات من وسط كل أنبوب مع الحرص على عدم التعرق أكثر من ملليلتر من خلايا الدم الحمراء والاستغناء عن محتويات الأنبوب في واحدة من أربعة أنابيب 50 ملليلتر جديدة. أضف 500 ميكرولترات إضافية من الخرز المغناطيسي الدوامة إلى كل أنبوب وخلط بلطف حل الخلية.ضع الأنابيب مرة أخرى في الاقحام المغناطيسي لمدة خمس دقائق قبل نقل المحتويات بعناية من وسط كل أنبوب إلى واحدة من أربعة أنابيب 50 ملليلتر جديدة. عندما تم نقل جميع تعليق الخلية، ضع كل أنبوب 50 ملليلتر جديد في حاملات المغناطيس لمدة خمس دقائق قبل نقل محتويات الأنبوب بعناية إلى مجموعة ثالثة من أربعة أنابيب 50 ملليلتر جديدة. ثم جمع الأبقار الجنينية، والمضادات الحيوية، وليبوبوليكساكريد، وترفيرفيرون غاما واستبدلوا المستفحلين من ثقافات M2 بمصل بوفين الجنيني، والمضادات الحيوية، و M-CSF، وinterleukin-4. بعد 24 ساعة من الثقافة، اغسل كل لوحة من الضامة المنشطة مرتين مع برنامج تلفزيوني جديد لكل غسل. بالنسبة لتحليلات الحمض النووي الريبي والبروتين، فإن الخلايا المرفقة مباشرة في اللوحات تسمح بجمع محتويات الخلايا المفرج عنها لتحليلها.لتحليل تدفق cytometric، فصل الخلايا مع اثنين من ملليمولار EDTA في برنامج تلفزيوني لمدة 10 دقائق في 37 درجة مئوية قبل كشط الخلايا بلطف من قيعان لوحة للسماح جمعها في أنابيب microcentuge 1.5 ذلك، لا يقلل نقل الدم بشكل كبير من صلاحية الخلايا المشتقة من المريض أو السيطرة عليها بغض النظر عن مرحلة نضوج الخلايا أو ظروف النقل. ينتج عن عملية نقل الدرن تنظيمًا هابطًا فعالًا للجين المستهدف عند التهجين مع الحمض النووي الريبي الصغير المحدد في اليوم الأول واليوم الرابع بعد العزل.وعلاوة على ذلك، تظهر الخلايا التي تُصاب بـ "ميكرورنا" زيادة في التعبير عن ميكرورنا بمقدار 48 إلى 72 ضعفاً على
