Den inneholdt metoden er enkel å bruke og tilbyr en rask, nøyaktig og reproduserbar måling av DNA-skaden respons inkludert Double-strand Break (DSB) induksjon og reparasjon, celle syklus effekter og apoptotisk celle død. Målet med onkologi er å drepe eller deaktivere kreftceller uten å skade normale celler. Mange terapier enten direkte eller indirekte induserer gentoksisk stress i kreftceller, men også til noen forlenge i normale celler. Kjemoterapi eller målrettede legemidler er ofte kombinert med strålebehandling for å forsterke radiosensitivity av bestrålt tumor1, 2,3,4,5, som gir mulighet for en reduksjon av strålingsdosen for å minimere normal vevsskade.
Ioniserende stråling og andre gentoksisk midler induserer ulike typer DNA skade, inkludert base modifikasjoner, strand krysskoblinger og Single-eller Double-strand pauser. DNA dobbel-strand pauser (DSBs) er de mest alvorlige DNA lesjoner og deres induksjon er nøkkelen til cellen drepe effekten av ioniserende stråling og ulike cytostatiske narkotika i radiochemotherapy. DSBs ikke bare skade integriteten til Genova, men også fremme dannelsen av mutasjoner6,7. Derfor forskjellige DSB reparasjon trasé, og mekanismer for å eliminere ugjenkallelig skadede celler som apoptose har utviklet seg i løpet av evolusjon. Hele DNA-skade respons (DDR) er regulert av et komplekst nettverk av signalering trasé som kommer fra DNA-skade anerkjennelse og celle syklus arrestert for å tillate for DNA-reparasjon, til programmert celle død eller inaktive i tilfelle reparasjon svikt8.
Den presenterte flyten analytiske metoden er utviklet for å undersøke DDR etter gentoksisk stress i en omfattende analysen som dekker DSB induksjon og reparasjon, samt konsekvenser av reparasjons feil. Den kombinerer måling av mye brukt DSB markør γH2AX med analyse av celle syklus og induksjon av apoptose, ved hjelp av klassisk subG1 analyse og mer spesifikk evaluering av ordtak-3 aktivering.
Kombinasjonen av disse endepunktene i en analyse reduserer ikke bare tid, arbeid og kostnads utgifter, men muliggjør også celle syklus spesifikk måling av DSB induksjon og reparasjon, samt ordtak-3-aktivering. Slike analyser vil ikke være mulig med uavhengig gjennomførte analyser, men de er svært relevante for en helhetlig forståelse av DNA-skaden respons etter gentoksisk stress. Mange anti-kreft narkotika, for eksempel cytostatiske forbindelser, er rettet mot å dele celler og deres effektivitet er sterkt avhengig av celle syklus scenen. Metoden har blitt brukt til å sammenligne effekten av ulike radiotherapies for å overvinne motstands mekanismer i glioblastom13 og å analysere samspillet mellom ioniserende stråling og målrettede stoffer i osteosarcoma14,15 og atypisk teratoid rhabdoid kreft16. I tillegg har den beskrevne metoden blitt mye brukt til å analysere bivirkninger av radio-og kjemoterapi på Mesenchymal stamceller17,18,19,20,21, 22,23,24, som er avgjørende for reparasjon av behandling-indusert normal vevsskade og har en potensiell anvendelse i regenererende medisin.

cell cycle-specific measurement of &#947;h2ax and apoptosis after genotoxic stress by flow cytometry

Den presenterte metoden eller litt modifisert versjoner har blitt utviklet for å studere konkrete behandlingstiltak og bivirkninger av ulike anti-kreft behandlinger som brukes i klinisk onkologi. Det gjør det mulig for en kvantitativ og langsgående analyse av DNA-skaden respons etter gentoksisk stress, som indusert av strålebehandling og en rekke anti-kreft narkotika. Metoden dekker alle stadier av DNA-skaden respons, gir endepunkter for induksjon og reparasjon av DNA dobbel-strand pauser (DSBs), celle syklus arrest og celle død av apoptose i tilfelle reparasjon svikt. Kombinere disse målingene gir informasjon om celle syklus-avhengige behandlingseffekter og dermed gir en grundig studie av samspillet mellom cellulære spredning og mestring mekanismer mot DNA skade. Som effekten av mange kreftlegemidler, inkludert kjemoterapeutiske midler og ioniserende stråling er begrenset til eller sterkt varierer i henhold til spesifikke celle syklus faser, correlative analyser stole på en robust og gjennomførbar metode for å vurdere behandlingen effekter på DNA i en celle syklus-spesifikk måte. Dette er ikke mulig med enkelt endepunkt analyser og en viktig fordel med den presenterte metoden. Metoden er ikke begrenset til en bestemt cellelinje og har blitt grundig testet i en rekke tumor og normal vev cellelinjer. Metoden gir informasjon om cellesyklusavhengige behandlingseffekter og tillater og grundig studie av samspillet mellom cellulær spredning og mestringsmekanismer mot DNA-skade. Denne metoden kombinerer viktige endepunkter fra DNA skade respons inkludert dobbel-strand frag anerkjennelse og reparasjon, celle syklus effekter, og eventuell celle død av apoptose i en omfattende analyse. Vi har brukt denne teknikken hovedsakelig til å undersøke spesifikke behandlingsresponser og bivirkninger av radiokjemoterapeutiske behandlinger i klinisk onkologi.Analysen er svært nyttig for korrelative studier innen radiologi og strålebehandling, men det kan også brukes på ulike andre områder av onkologi. Etter å ha samlet cellene fra kulturen i henhold til standard celleinnsamlingsprosedyrer, gjenbruker du pelleten i en milliliter PBS. Og overfør cellene til to milliliter fikseringsløsning i en hette.Etter en 10 minutters inkubasjon ved romtemperatur, samle cellene ved sentrifugering og dekantere supernatanten. Det er viktig å fikse cellene som en enkelt celle suspensjon, og for å holde fikseringstiden konstant for alle prøvene. Gi tiltalende celler nok tid til å løsne for å oppnå pent avrundede celler for analyse.Løsne deretter pelleten ved å trykke og gjenbruke cellene i tre milliliter med 70% etanol. For å vaske cellene før farging, sediment cellene ved sentrifugering, og løsne pellet ved å trykke. Deretter gjenbruker cellene med to påfølgende vasker med tre milliliter vaskeløsning og en vask med en milliliter vaskeløsning.Kast det overnaturlige mellom hver vask. Karbonioner induserer høyere gamma H to AX toppnivåer i glioblastomceller som avtar saktere og forblir betydelig forhøyet i 24 til 48 timer sammenlignet med fotonstråling ved samme fysiske dose. For både karbonioner og fotonstråling var gamma H to AX-nivåene høyest i G enfaseceller sannsynligvis fordi DNA dobbel trådbrudd reparasjon er begrenset til banen for ikke-morderisk slutt sammenføyning på dette stadiet i cellesyklusen.I tråd med høyere dobbel tråd bryte induksjonshastighet og langsommere reparasjon kinetikk, karbonioner indusere en sterkere og lengre varig celle syklus arrest i G to fase og en høyere hastighet av apoptose enn fotoner. Avhengig av styrken av behandlingseffekten, kan oppløsningen av de forskjellige cellesyklusfasene være utfordrende. I noen cellelinjer har DAPI-konsentrasjonen stor innvirkning på kvaliteten på cellesyklusprofilen og må justeres.Lysbilder kan tilberedes fra de gjenværende prøvene etter strømningscytometri for å evaluere antall, størrelse og form av gamma H to AX foci og å skille mellom fokal og parnukleær gamma H to AX farging. Ved hjelp av vår metode kan vi vise at mesenchymale stamceller er relativt motstandsdyktige mot irioniserende stråling og topoisomerase hemming, men følsomme for gliomyacin. Siden PFA-røyk er giftig, må du alltid bruke en røykhette når du klargjør fikseringsløsningen og under fikseringstrinnet.Husk også å bruke hansker når du håndterer DAPI.
