El genoma contiene distintos tipos de dominios de cromatina, que se correlacionan con la actividad transcripcional y la sincronización de replicación2,3. Estos dominios de cromatina varían en tamaño de unas pocas kilobases (kb) a más de 100 kb y se caracterizan por un enriquecimiento en distintas modificaciones histonas4. Las preguntas centrales son: ¿cómo se forman estos dominios y cómo se propagan?
Uno de los dominios de cromatina más bien caracterizados se fomenta a través de la actividad del complejo represivo 2 (PRC2). PRC2 es un complejo multisubunidad compuesto por un subconjunto del Grupo Polycomb (PcG) de proteínas5,6, y cataliza la mono-, di- y trimetilación de lisina 27 de histona H3 (H3K27me1/me2/me3)7,8, 9,10. H3K27me2/me3 están asociados con un estado de cromatina represiva, pero la función de H3K27me1 no está clara6,11. Uno de los componentes principales de PRC2, el desarrollo embrionario de ectodermos (EED), se une al producto final de la catálisis PRC2, H3K27me3, a través de su jaula aromática y esta característica da como resultado la estimulación alostérica de PRC212,13. La actividad enzimática PRC2 es crucial para preservar la identidad celular durante el desarrollo, ya que la expresión inapropiada de ciertos genes del desarrollo que están contraindicados para un linaje específico, sería perjudicial5,6 . Todos los sistemas experimentales anteriores diseñados para investigar la formación de dominios de cromatina, incluidos los dominios de cromatina mediados por PRC2, se realizaron en condiciones de estado estacionario, que no pueden rastrear los eventos de formación de dominios de cromatina en Células. Aquí, presentamos un protocolo detallado para generar un sistema celular inducible que monitorea el reclutamiento inicial y la propagación de dominios de cromatina. a method to study de novo formation of chromatin domains

La organización y la estructura de los dominios de cromatina son exclusivas de los linajes celulares individuales. A pesar de los enormes esfuerzos, nuestra comprensión de la formación y propagación de dominios de cromatina sigue siendo limitada.
