Esta técnica permite estudiar NG, trampas extracelulares de neutrófilos, en tejidos de parafina tanto de ratón como de hombres.
Y también se considera estudiar material archivado o estudio retrospectivo. En primer lugar, coloque los portaobjetos preparados en bastidores y sumerjalos en los medios utilizados para la deshidratación y el desbroce en orden inverso durante cinco minutos cada uno. A continuación, calienta un baño de agua con una placa caliente con temperatura controlada a 70 grados centígrados.
Coloque un frasco lleno de tampón de recuperación de epítopos inducidos por el calor y un 10% de glicerol en el baño de agua. Cuando el búfer haya alcanzado los 70 grados centígrados, coloque el bastidor con las diapositivas en el tarro del búfer. Incubar los toboganes a 70 grados centígrados durante 120 minutos.
Después de esto, retire el frasco del baño de agua y déjelo enfriar a temperatura ambiente. Enjuague las secciones enfriadas tres veces con el agua ionizada y una vez con TBS. Usando papel de filtro enrollado o un hisopo de algodón, retire cuidadosamente cualquier líquido entre las secciones de los portaobjetos, asegurándose de dejar las secciones hidratadas.
A continuación, utilice una pluma de barrera hidrófoba para crear una barrera alrededor de cada sección. Incubar la sección en el tampón de bloqueo a temperatura ambiente durante 30 minutos para evitar la unión inespecífica. Diluir los anticuerpos primarios en el tampón de bloqueo a una concentración de un microgramo por mililitro.
Retire el búfer de bloqueo de las diapositivas y agregue los anticuerpos primarios diluidos, asegurándose de utilizar un volumen suficiente para evitar el secado. Cierre el recipiente húmedo e incubar durante la noche a temperatura ambiente. Al día siguiente, lave las secciones tres veces con TBS con cada lavado que dura cinco minutos.
Preparar una solución de trabajo de anticuerpos secundarios en el tampón de bloqueo. Cubra las secciones de los tejidos con la solución de anticuerpos secundaria y transfiera las diapositivas a un recipiente húmedo. Selle el recipiente húmedo e incubar a temperatura ambiente durante una hora.
Después de esto, lave las secciones tres veces con TBS y una vez en agua con cada lavado que dura cinco minutos. Cubra las secciones lavadas con un medio de montaje y aplique el vidrio de cubierta evitando la formación de burbujas. Después de que el medio de montaje se haya solidificado, utilice un microscopio confocal o un microscopio de campo ancho con filtros de paso de banda adecuados para analizar la inmunofluorescencia.
Para digitalizar las secciones completas con un escáner de diapositivas, establezca la intensidad de la fluorescencia utilizando los controles negativos y positivos. Usando la tinción de laastasa de neutrófilos, busque las áreas ricas en neutrófilos y haga zoom en esas áreas para comprobar si la señal de laastasa de neutrófilos es granular o extracelular. Si la señal es extracelular y se superpone con la histona y la tinción de ADN, se han obtenido las trampas extracelulares de neutrófilos.
En este estudio, los componentes de NET se detectan con éxito en el tejido incrustado en parafina, tanto de origen humano como murino. Si las secciones de tejido tienen un espesor entre dos y tres micrómetros, pueden ser analizadas por microscopía de campo ancho utilizando objetivos 10x o 20x. Para evaluar correctamente la tinción, se han procesado tanto muestras negativas como positivas.
Una sección representativa del tejido de apendicitis humana muestra tejidos manchados para NE, H2B y Hoechst 33342. Las imágenes de la izquierda son de un área de la sección que contiene NETs, mientras que las imágenes de la derecha son de un área diferente de la misma sección que contiene numerosos neutrófilos pero no NETs. Las áreas con formación masiva de NET se pueden encontrar fácilmente incluso con aumentos bajos, ya que los tres componentes de colocación NET se encuentran a menudo en estructuras extracelulares de cadena.
Esto aparece en la superposición de los tres canales como fibras extracelulares blanquecinas que se pueden cuantificar mediante el uso de software de análisis de imágenes para crear una superposición púrpura utilizando píxeles de señales superpuestas que son positivas para verde, rojo y azul. Para una resolución más alta, los microscopios confocales o los microscopios de campo ancho con desconvolución tienen que utilizarse para minimizar el desenfoque fuera de foco. Una proyección máxima de una pila confocal de un área rica en NET a partir de la misma muestra de apendicitis humana muestra que NE se encuentra en gránulos, pero también es abundante extracelularmente donde se coloca con H2B y con ADN.
La colocación extracelular da como resultado una combinación de colores blanquecinos. Los píxeles positivos para verde, rojo y azul se pueden utilizar una vez más para crear una superposición púrpura que presenta NOMINADAs. Un detalle representativo de una sección central de un pulmón de ratón infectado con Mycobacterium tuberculosis proporciona otro ejemplo de la colocación de los tres componentes NET siendo claramente visible como áreas blanquecinas entre neutrófilos que se pueden utilizar para crear una capa púrpura que indica NG.
Durante el procedimiento de tinción, las secciones no deben secarse porque esto causará una tinción falsa positiva o un fondo alto. El análisis del material archivado puede ayudar a comprender el impacto de los NT en las enfermedades. Debido a que el tejido de parafina puede tener un fondo enorme, es importante tener un control positivo y negativo que se puede distinguir entre la tinción y el fondo.
