免疫組織化学および診断組織病理学に使用される組織は、10%リン酸緩衝液ホルマリンに日常的に固定されている。ホルマリンで固定された脳組織における狂犬病ウイルス抗原の検出は、独特の課題を提示し、IHC試験は狂犬病ウイルス抗原検出のための敏感かつ特異的な方法であることが判明した。生検または他の検出方法の主な利点は、抗原が組織および細胞形態に関連して検出できることである。IHCでは、脳内および非脳組織における狂犬病抗原の局在化により、定期的なヘマトキシリンおよびエオシン染色片を介して得ることができなかった追加情報を提供する。狂犬病診断検査は、狂犬病ウイルスに曝露されたヒトにおける曝露後予防の開始に重要である。IHCは、ホルマリン固定組織に関する最も一般的に行われるアッセイの1つです。特異的抗体を用いて、それぞれの病原体の検出に使用することができる。この手順を開始するには、壊死後に採取した3〜5ミリメートルの脳組織を、24〜72時間10%緩衝ホルマリン溶液に入れる。おおよその組織重量、組織タイプ、ホルマリンの体積を記録する。ホルマリン固定後および処理前に、より長い組織貯蔵のために組織を70%エタノールに入れる。標本の固定に続いて、脳幹、小脳、海馬の断面などの重要な脳領域を含むように組織を解剖する。それぞれ、3ミリメートルから5ミリメートルの間の厚さにカットします。組織を処理カセットに入れます。パラフィンワックス浸潤のための組織カセットを処理します。パラフィンブロックに埋め込み、ミクロトームに切り離します。まず、テキストプロトコルの図1に示すように、染色皿を設定します。各皿に250ミリリットルの溶液を充填します。AEC基板ストック溶液を調製するために、ガラスピペットを使用して、1つの20ミリグラム錠剤のAECを5ミリリットルのN、N-ジメチルホルムアミドに溶解させる。プロテアーゼストック溶液を調製するために、プロテアーゼの7ミリグラムをPBSの200ミリリットルに溶解する。リンスバッファーを調製するには、100 ミリリットルの Tween 80 ~ 990 ミリリットルの PBS を加え、よく混合して PBT-T の均質溶液を形成します。ミクロトームを使用して、5マイクロメートルのパラフィンセクションを作ります。38°Cの水浴にセクションをロードし、ガラスのスライドにそれを収集します。試薬耐性ペンでスライドにラベルを付けます。スライドをトレイに置き、オーブンで55~60度で1時間溶かします。その後、スライドをオーブンから取り出し、5分間の3つの連続したキシレンリンスを使用して、すぐにそれらを脱パラフィン化します。この後、テキストプロトコルで概説されているように、脱イオン水へのエタノールの希釈を減少させる順次浸漬によってスライド上のセクションを水分補給します。プロテアーゼで30分間スライドを処理し、タンパク質分解抗原を取り出し、PBS-Tでスライドを10分間リンスし、3%過酸化水素で10分間処理します。その後、PBS-Tでスライドを10分間洗い直します。まず、バッファから1つのスライドを取り外し、一度に1つのスライドのみを処理し、他のスライドは水没したままにし、ペーパータオルを使用して組織セクションの周りから余分なバッファを消します。スライドを湿度チャンバーに入れ、室温で通常のヤギの血清で15分間インキュベートします。次に、最適な所定希釈原発性抗狂犬病抗体と共に湿度チャンバー内のスライドをインキュベートし、陰性対照抗体を、その間に洗剤を全く入れずに室温で60分間培養する。この後、PBS-Tでスライドを10分間洗浄し、室温でビオチン化抗体を用いて湿度チャンバーで15分間インキュベートします。PBS-Tでスライドを10分間洗い直します。スライドと湿度チャンバーをストリップ除細分とHRB錯体を室温で15分間インキュベートし、PBS-Tを10分間洗浄します。次いで、室温で10分間、湿度チャンバ内でAECでインキュベートする。スライドを脱イオン水で10分間洗い、ギルのヘマトキシリンでカウンターステインし、脱イオン水で1~2分希釈します。この後、滑り水を浸して余分なヘマトキシリンをすすぎま。スコットの水道水のスライドを30秒間洗い流し、脱イオン水で10分間洗います。スライドを1つずつ取り出し、水溶性の取り付け媒体で取り付けます。次に、軽い顕微鏡を使用してスライドを読み取ります。本研究では、ホルマリン固定組織の代替診断試験として狂犬病ウイルス抗原の検出に免疫特性化学試験が実証された。脳幹、小脳細胞、プルキンエ細胞、海馬など、異なる脳組織における陽性および陰性対照サンプルの代表的な免疫細胞化学染色結果がここに示されている。マゼンタ赤染色は、AEC基質を用いて青色の対部背景に対して発色を示し、狂犬病抗原に対する抗体の反応性に起因する。免疫組織化学の陽性結果は、組織切片におけるマゼンタ赤染色に相当する。細胞質含有物の染色と様々なサイズの粒状の含み物は、RABV感染に陽性のサンプルを示しています。試料は、ヘマトキシリンによる特定の赤色染色または青色の背景のみが観察されなかった場合に陰性とみなされる。陽性染色に加えて、含包物の分布は、サンプル中の狂犬病抗原のレベルを間接的に定量化することができ、これは組織サンプルのウイルス負荷に対応する可能性がある。分布のレベルに関係なく、任意の特定の染色は、RABV抗原検出のために陽性としてサンプルを分類します。組織はホルマリンで適切に固定し、このプロセス中に組織を凍結しないようにする必要があります。狂犬病抗原はIHCによって検出され、次に、分子技術は、RNA配列情報に基づいてリッサウイルス変異体を決定するパラフィンセクション上で行うことができる。キシレンやホルマリンなどの試薬は、ヒュームフードで取り扱う必要があります。発がん性物質であるため、AECの取り扱い中は注意が必要です。
