Den flowcytometri teknikken for måling av ADCP-aktivitet beskrevet her ble først beskrevet i 201130 og har siden blitt påvist robust og sitert i mer enn 80 studier. Morsmelk (BM) består av mors celler som er > 90% levedyktig1. Celle sammensetning er påvirket sterkt av stadium av amming, helsetilstand av mor og spedbarn, og individuell variasjon, som fortsatt er dårlig forstått1,2,3,4. Gitt at BM inneholder ~ 103− 105 leukocytter/ml, kan det anslås at ammet spedbarn svelges ~ 105− 108 mors leukocytter daglig5. Various in vivo studier har vist at mors leukocytter gir kritisk immunitet til spedbarnet og er funksjonell langt utover disse nettstedene for innledende svelging5,6,7,8 ,9,10,11. Alle moderskap celler inntatt av spedbarnet har potensiale til å utføre immun funksjoner ved siden av eller for å kompensere for spedbarnets egne leukocytter12.
Mor-til-barn-smitte (MTCT) av humant immunsviktvirus (HIV) er fortsatt en krise i ressurs-begrensede land. Som diaré og luftveissykdommer er ansvarlig for betydelige utbredelsen av dødelighet blant spedbarn i ressurs-begrensede land, og disse sykdommene er betydelig redusert med eksklusiv amming, fordelene til HIV-smittede mødre av amming langt oppveier risikoen13,14,15. Med mindre tilgang til rent vann og passende morsmelkerstatning er pålitelig, WHO anbefaler ikke opphør av amming for HIV-smittede mødre16. Omtrent 100 000 MTCTs via BM forekommer årlig. Likevel, bare ~ 15% av spedbarn ammet av deres HIV-smittede mødre blir smittet, noe som tyder på en sterk beskyttende effekt av BM17,18,19,20,21. Fluorescerende perler belagt med HIV-konvolutt (env) epitope ble brukt som mål for analyse av ADCP. Celler ble farget med CD45 overflate markør for å identifisere leukocytter. Det ble funnet at ADCP aktivitet var betydelig over kontroll eksperimenter og reproduserbar målbare ved hjelp av en HIV-spesifikk antistoff 830A.

Isolation of Leukocytes from Human Breast Milk for Use in an Antibody-dependent Cellular Phagocytosis Assay of HIV Targets

Vi utviklet en streng metode for å isolere celler fra morsmelk for å studere antistoffavhengig cellulær fagocytose, eller ADCP, ved morsmelkfagocytter mot HIV-mål. Denne teknikken forenkler relativt rask og enkel isolering av brystmelkceller for å evaluere kapasiteten til leukocytterpopulasjoner for å utføre ADCP. Å demonstrere prosedyren vil være Alisa Fox, en tekniker fra laboratoriet mitt.Etter å ha valgt et relevant målantigen, bruk et kommersielt sett til å biotinylere proteinet av interesse i henhold til produsentens protokoll. Og deponer den biotinylererte prøven i det øvre kammeret i en spin-kolonne. Tilsett PBS opptil 400 mikroliter og hette kammeret før du setter kolonnen inn i et oppsamlingsrør for sentrifugering.Etter å ha kastet strømmen gjennom, legg PBS til 400 mikroliter for en ny sentrifugering. For alle CD45 positive celler, eller for hver leukocytt undergruppe av interesse, måle antistoffavhengig cellulær fagocytose aktivitet ved gating med en markør på perlen positive celler i et histogrammet av riktig fluorophore kanal for konjugert perler.Deretter beregner antistoffavhengige cellulære fagocytose score som median for fluorescensintensiteten av perlen positive celler med prosentandelen av den totale CD45 pluss cellene i den positive befolkningen. Her vises gatingstrategien for å eliminere dobler, rusk og døde celler. Mellom en til 12% av totale levende celler er vanligvis CD45 positive leukocytter med det meste av den påståtte side scatter lav til middels CD45 lave monocytter ser ut til å være forløpere, eller til umodne celler, basert på side scatter versus CD45 gating.De påståtte granulocytter er definert som side scatter høy, CD45 mellomliggende celler. Måling av ADCP-aktiviteten til nyisolerte morsmelkceller ved hjelp av et HIV-spesifikt monoklonalt antistoff ved en måned etter fødsel viser at behandling med aktinhemmer Cytochalasin D og eller Fc-reseptorblokkerende antistoffer før inkubasjon med antistoffbundne antigenkobler resulterer i ADCP-aktiviteten på eller under som observeres i nærvær av kontrollantistoff. ADCP-poengsummen som bestemmes for totalt CD45-positive celler, er vanligvis mellom 25 og 35 ganger over bakgrunnsnivåene, definert ved hjelp av en negativ kontroll, anti-miltbrann monoklonalt antistoff.Hver morsmelkprøve er unik og noen pellets kan være vanskelig å se. I tillegg kan leukocyttpopulasjonene, og derfor ADCP-skårene, variere enormt fra utvalg til utvalg.
