このプロトコルは、アキソロテルでの遺伝子スクリーニングに使用できる変異原性ハプロイド四肢の産生を可能にする。ハプロイドでは、低レベルの突然変異誘発でさえ、細胞内の機能性の喪失を生じ得る。この移植技術はまた、胚性致死的な型を救うために使用することができる。この方法は、体外受精および移植に適した両生類種の再生を研究するために使用することができる。ハプロイド胚をドナーとして使用する前に、GFP陽性および陰性二倍体胚の間の移植片の成功を練習し、確認することをお勧めします。女性のテインジドナー調製のために、性的に成熟した白または白のRFP女性アキソロテルを麻酔し、ホテ供与ドナーとして働く。疼痛シミュレーションに対する応答の欠如を確認した後、30ゲージのインスリン注射器を使用して、後肢に戻る筋肉性の後部に0.15ミリリットルのヒト絨毛性ゴナドトロピンを注入する。脊髄との接触を避けるために、正中線に45度の角度で注射器を挿入します。注入した動物を新鮮な40%ホルトフレッターの溶液に入れ、アショロトルを摂氏8~12度の冷蔵庫に48時間移します。その後、室温に雌を戻します。メスが卵を産んでいる間、完全に麻酔をした雄を解剖顕微鏡の下で湿ったペーパータオルの上に仰向けの位置に置き、P1000ピペットの先端を支配的な手でクロアカの基部に置きます。もう一方の人差し指と親指を骨盤に2〜3センチメートルのロストラルを置き、後ろ足に向かって指を動かしながら動物を静かに絞り、各サンプルを採取した精子尿サンプルを新しいマイクロ遠心チューブに洗い流します。次に、各サンプルから5マイクロリットルをペトリ皿に移し、逆顕微鏡を使用して精子の質を検査します。カウント後、精子を0.1X滅菌MMRで1ミリリットル当たり4番目の細胞に8倍10倍の濃度に希釈し、卵子1個あたり0.5マイクロリットルの精子細胞を新しいペトリ皿に移す。次に、ピペットチップを使用して1ミリメートルの厚い層に懸濁液を広げ、プラスチックリフトを使用して、254ナノメートルのクロスリンカーの電球からサンプルを4センチメートル配置し、8倍の10倍のマイクロジュールから5番目のマイクロジュールに照射します。健康な精子サンプルを採取した後、湿ったペーパータオルの積み重ねの上に女性をサピインの位置に置き、同様の紅潮運動を使用して雌のアキソロトルから未受精卵を抽出する。その後、湿った鉗子を使用して卵を10センチメートルのペトリ皿に移します。収集から15分以内に、P10ピペットを使用して、照射された精子を未受精卵に分配し、各卵子に0.25〜0.5マイクロリットルの不活性化精子を塗布します。卵を室温で30分間座させた後、卵を0.1X MMRであふれさせる。水分補給から30分後、鋭利な鉗子を使って卵をゼベゼし、卵を摂氏18度にして7時間以内にマイクロインジェクションします。ハプロイド・ディプロイド・キメラを生成するには、1つまたは2つの段階で1つまたは2つのステージを持つ1つの健康なハプロイドドナーを、摂氏10度または低冷却段階で外科培地を含む冷やされた手術皿のトラフ内にGFP陽性ディプロイド宿主胚と一致させた。2つの超微細オートクレーブストレートチップ鉗子を使用して、四肢の芽全体と周囲の外皮および中皮組織層を取り除き、宿主組織を脇に置きます。ハプロイドドナーから同等のサイズの組織シースを取り除き、ハプロイドドナー組織シースを宿主胚の対応する領域に置く。破砕された顕微鏡カバーグラスからオートクレーブ長方形のガラス破片で組織を固定し、組織を宿主胚の体内にそっと押し込みます。カバーグラスの破片を剥がすために鉗子を使用する前にガラスが滑っていないことを確認するために、20分ごとに60〜75分間アンカーを所定の位置に置いたままにしておきます。生着した胚を新鮮な外科媒体に移し、12または24ウェルプレートに個別に生まれた胚を収容する前に、一晩で摂氏8〜12度で治癒することができます。卵母細胞の38.7%が正常に発達したハプロイド胚に発達した。移植片の段階では、ハプロイド胚は前部後軸に沿って減少した曲率および黄身栓の不完全な囲いを示す。また、蛍光顕微鏡を用い、ハプロイド胚が父親由来のGFP発現を含まないのを確認することができる。失敗し、不純な移植片は、様々な型の型を生成します。移植片が清潔で通常開発される場合、GFP発現は主に、宿主脊髄に由来するニューラルネットワークである腕神経叢に限定されるべきである。Punctate GFP発現は、発達中の四肢に移行する血液由来宿主細胞の感覚ニューロンと思われる単一細胞にも存在する。RFP陽性ドナーが使用される場合、ハプロイド移植片肢はRFPの普遍的発現を示す。開発を通じて、非変異原性ハプロイド肢は、キメラ動物の対向性ジプロイド前肢よりも有意に短い。非変異原性ハプロイド四肢は完全に再生するが、二倍化と比較してデジタル成長段階に到達するわずかな遅れを示す。滅菌技術を使用し、きれいに移植された四肢を生成するために、胚宿主から完全な中胚および外胚組織層を完全に除去して交換することを忘れないでください。突然変異肢は、再生型の切断およびスコア付けが可能である。変異対立体は次世代シーケンシングで定量することができる。CRISPR-Cas9変異誘発と組み合わせることで、当社の移植技術により、ハプロイド四肢を再生する際に標的遺伝子の陰性選択スクリーンを実施することができました。
