Live Cell Imaging to Assess the Dynamics of Metaphase Timing and Cell Fate Following Mitotic Spindle Perturbations

Live celleavbildning muliggjør observasjon av dynamiske cellulære prosesser over tid i en enkelt celle. Disse tilnærmingene er kraftige for å vurdere forholdene som påvirker den dynamiske prosessen med celledeling og avslører virkningene som defekter i mitose har på dattercellespredning og levedyktighet. Fototoksisitet som følge av lyseksponering under bildebehandling representerer en betydelig utfordring knyttet til denne protokollen.Derfor bør eksponeringstider og bildeintervall og varighet optimaliseres. Bruk aseptisk teknikk og et biosikkerhetsnivå to sikkerhetsskap, bruk et sterilt engangsglass Pasteur pipette for å aspirere mediet fra kulturplaten som inneholder cellelinjen som bærer uttrykket konstruere av interesse. Ikke mer enn 30 minutter før avbildning, tilsett en relevant konsentrasjon av et mitotisk legemiddel til en eller flere av brønnene og legg til et likt volum av inhibitordiluent til samme antall brønner som kontrollene.For å forberede mikroskopet for avbildning, plasser platen på scenen av et omvendt epifluorescencemikroskop utstyrt med et høyoppløselig kamera, et miljøkammer forvarmet til 37 grader Celsius og et leveringssystem for fuktet 5% karbondioksid. Velg et 20 ganger luftmål med en numerisk blenderåpning på 0,5 og utstyrt for høykontrastfluorescens og fasekontrast, eller lys feltavbildning og se cellene for å justere kurset og finne fokus for å bringe cellene i fokus. Åpne det genererte poengkontrollpanelet, og velg Begrenset og Kantlinje under Arbeidsområde for å begrense koordinatvalgområdet for å utelate grensene for brønnen.Velg Hele området på rullegardinmenyen Områdebegrensning, og velg en tilfeldig punktplassering. Angi antallet til seks, og klikk Randomize for å velge antall og fordeling av punktene som skal registreres per brønn. Klikk deretter på og skriv inn verdiene i tidssekvenskontrollpanelet for å velge og skrive inn tidsintervallet og varigheten for å samle bildene.Hvis du vil visualisere RFP-H2B merket kromatin, velger du bildene som er tatt med RFP-filterkuben på plass og identifiserer en celle som kommer inn i mitose som angitt av den første kromatinkomprimeringen og atomkonvoluttsbrytningen for å bestemme mitotisk timing av metafasejusteringen og anafaseinnsettelsen i individuelle celler. Spor cellen av interesse gjennom påfølgende tidspunkter i den oppkjøpte filmen for å bestemme antall tidspunkter eller minutter fra mitotisk oppføring til RFP-H2B merket kromatin fullfører justeringen på celleequator under metafasen. For å overvåke mitotisk timing, mititotisk troskap og celleskjebne, fortsett å spore cellen gjennom påfølgende tidspunkter for å identifisere tidskoordinaten der anaphase kromosomsegregering er tydelig og / eller når kromatindekompensasjon og kjernefysisk konvoluttreformasjon har skjedd.Deretter visualiserer DU RFP-H2B for å identifisere celler i hver populasjon som viser mititotiske defekter, inkludert lagging kromosomer og kromatinbroer under anaphase kromosom segregering. Ved å visualisere alfa tubulin-EGFP, kan det observeres at celler som opplever spindelavbrudd gjennomgår dynamiske endringer som spindeltrekkfokus er oppnådd og en bipolar mitotisk spindel dannes som forberedelse til celledeling. Samtidig med spindelenhet kan kromosombevegelse visualiseres med RFP-H2B for å vurdere kromosomjusteringen og segregeringsmogivelsen.Ved hjelp av live cellebildetilnærmingsmetoder viser disse representative resultatene at celler med normalt centrosome innhold er i stand til å fortsette fra kjernefysisk konvoluttsammenbrudd gjennom metafasejustering og anafaseinnsett for å oppnå en bipolar divisjon på under 30 minutter. I nærvær av ekstra centrosomer er nesten 50% av cellene i stand til å overvinne en forbigående multipolar mitotisk spindel og danne en bipolar spindel i fullstendig celledeling. De resterende cellene er ikke i stand til å oppnå en bipolar spindel, og som et resultat, avslutt mitose gjennom en multipolar divisjon.Uansett om spindel bipolaritet oppnås, viser celler med ekstra centrosomer en betydelig økt varighet av mitose sammenlignet med celler med to centrosomer. Indikerer at dynamikken i mitotisk progresjon kan endres selv når endringer i mititotisk utfall ikke er tydelig. Når du definerer eksponeringstider for fluorescerende kanaler, optimaliser eksponeringstidene for å begrense fototoksisitet.Også kamerapikselbinning kan velges slik at lavere eksponeringstider kan brukes. Denne prosedyren har anvendelser i farmakologiske screeninger, invasjon og migrasjonsanalyse. Disse tilnærmingene kan også brukes til å undersøke effekten av narkotikabehandlinger eller dynamikken i cellulær motilitet.
