Langlaufbiofilms bestaande uit zowel schimmel- als bacteriële soorten zijn betrokken bij verschillende pathologische aandoeningen in de mondholte. Candida spp. zijn vaak geïsoleerd van endodontische infecties1 en van parodontale laesies2,3. Bij slijmvliesinfecties is aangetoond dat streptokokkensoorten uit de mitisgroep de vorming van schimmels biofilm, weefselinvasie en verspreiding in zowel in vitro als murinemodellen4,5,6,7verbeteren. Het meest interessant, mondelinge vervoer van Candida spp. is bewezen te worden geassocieerd met de prevalentie van cariës bij kinderen8. Zoals blijkt uit knaagdiermodellen, verhoogt een symbiotische relatie tussen Streptococcus mutans en Candidas albicans de productie van extracellulaire polysachariden en leidt tot de vorming van dikkere en meer cariogene biofilms9,10.
In alle bovengenoemde omstandigheden, met name voor jonge kinderen, is de pH van de biofilm van belang voor de progressie van de ziekte, en de eminente rol van de biofilmmatrix voor de ontwikkeling van acidogene microomgevingen11 vraagt om methodologieën die het bestuderen van pH-veranderingen in cross-kingdom biofilms mogelijk maken. Eenvoudige en nauwkeurige confocale microscopiegebaseerde benaderingen om pH in bacteriële12 en schimmel13 biofilms te monitoren zijn ontwikkeld. Met de ratiometrische kleurstof C-SNARF-4 en op drempels gebaseerde beeldnabewerking kan extracellulaire pH in real-time worden bepaald in alle drie de dimensies van een biofilm14. In vergelijking met andere gepubliceerde technieken voor op microscopie gebaseerde pH-monitoring in biofilms is pH-ratiometrie met C-SNARF-4 eenvoudig en goedkoop, omdat het niet de synthese vereist van deeltjes of verbindingen die een referentiekleurstof15 of het gebruik van tweefotonexcitatie16bevatten . Het gebruik van slechts een kleurstof voorkomt problemen met sonde compartimentering, fluorescerende bleed-through, en selectief bleken16,17,18 terwijl nog steeds rekening houdend met een betrouwbare differentiatie tussen intra- en extracellulaire pH. Ten slotte wordt incubatie met de kleurstof uitgevoerd na de groei van de biofilm, waardoor zowel laboratorium als in situ-gekweekte biofilms kunnen worden bestudeerd.
Het doel van het huidige werk is om het gebruik van pH-ratiometrie uit te breiden en een methode te bieden om pH-veranderingen in cross-kingdom biofilms te bestuderen. Als proof of concept wordt de methode gebruikt om pH te monitoren in biofilms van twee soorten bestaande uit S. mutanen en C. albicans die aan glucose zijn blootgesteld.

monitoring extracellular ph in cross-kingdom biofilms using confocal microscopy

Langlaufbiofilms bestaande uit zowel schimmel- als bacteriële cellen zijn betrokken bij een verscheidenheid aan orale ziekten, zoals endodontische infecties, parodontitis, mucosale infecties en, met name, vroege kindercariës. In al deze omstandigheden beïnvloedt de pH in de biofilmmatrix microbe-host interacties en dus de progressie van de ziekte. Het huidige protocol beschrijft een confocale microscopiegebaseerde methode om de pH-dynamiek te monitoren in cross-kingdom biofilms bestaande uit Candida albicans en Streptococcus mutanen. Het pH-afhankelijke dual-emissiespectrum en de kleureigenschappen van de ratiometrische sonde C-SNARF-4 worden benut om druppels pH te bepalen in extracellulaire gebieden van de biofilms. Het gebruik van pH-ratiometrie met de sonde vereist een zorgvuldige keuze van imaging parameters, een grondige kalibratie van de kleurstof, en zorgvuldige, drempel-gebaseerde nabewerking van de beeldgegevens. Bij correct gebruik maakt de techniek een snelle beoordeling van extracellulaire pH mogelijk in verschillende gebieden van een biofilm en dus de monitoring van zowel horizontale als verticale pH-gradiënten in de tijd. Hoewel het gebruik van confocale microscopie Z-profilering beperkt tot dunne biofilms van 75 μm of minder, is het gebruik van pH-ratiometrie bij uitstek geschikt voor de niet-invasieve studie van een belangrijke virulentiefactor in cross-kingdom biofilms.

Na 24 uur en 48 uur ontwikkelden robuuste cross-kingdom biofilms zich in de putplaten. C. albicans vertoonden verschillende mate van filamenteuze groei en S. mutanen vormden dichte clusters tot 35 μm hoog. Enkele cellen en ketens van S. mutanen gegroepeerd rond schimmel hyphae, en grote intercellulaire ruimten aangegeven de aanwezigheid van een volumineuze matrix (Figuur S1).

Het protocol presenteert een eenvoudig uit te voeren methode voor de teelt van cross-kingdom biofilms bestaande uit StreptococcuS. mutans en Candida albicans. De daaropvolgende confocale microscopiegebaseerde pH-ratiometrie maakt nauwkeurige monitoring van de pH-ontwikkelingen in de extracellulaire matrix van die biofilms mogelijk.De zorgvuldige keuze van de parameters voor het verkrijgen van afbeeldingen is van het grootste belang om beeldgegevens te verkrijgen met voldoende contrast voor latere analyse. pH ratiometrie is een snelle, nauwkeurige en goedkope methode om zowel horizontale als verticale pH-gradiënten in cross-kingdom biofilms in real time te bestuderen. pH ratiometrie kan ook worden uitgevoerd in puur schimmel- en bacteriële biofilms.De methode draagt bij aan het vergroten van ons begrip van microbiële stofwisseling in biofilms. Het aantonen van de procedure voor biofilm groei zal Anette Aakjaer Thomsen, een technicus van mijn laboratorium. Grow S.mutans en C.albicans op bloed agar platen op 37 graden Celsius onder aërobe omstandigheden.Breng vervolgens enkele kolonies van elk organisme over naar reageerbuizen gevuld met vijf milliliter hersenhartinfusie en laat ze nog eens 18 uur groeien. Op de volgende dag, centrifuge de culturen op 1, 200 keer g gedurende vijf minuten en gooi de supernatant. Resuspend de cellen in fysiologische zoutoplossing en pas de OD 550 aan op 0,5 voor zowel C.albicans als S.mutans.Verdun vervolgens de S.mutans suspensie 1 tot 10 om gelijkwaardige concentraties te bereiken. Pipette 50 microliters van steriele speekseloplossing in de putten van een optische bodem 96-put plaat voor microscopie. Incubeer de plaat gedurende 30 minuten bij 37 graden Celsius, was de plaat vervolgens drie keer met 100 microliter steriele fysiologische zoutoplossing en leg de putten leeg.Voeg 100 microliters van C.albicans suspensie toe aan elke put, broed de plaat gedurende 90 minuten op 37 graden Celsius en was de put vervolgens drie keer met zoutoplossing. Voeg vervolgens 100 microliters van warmte-geïnactiveerd foetaal runderserum toe aan elke put. Broed de plaat twee uur uit en was hem vervolgens drie keer met zout.Leeg de putten, maar laat een 20 microliter reservoir om overmatige afschuifkrachten te voorkomen. Voeg 100 microliter S.mutans suspensie en 150 microliter BHI met 5% sacharose toe aan elke put. Dan incubeer de plaat op 37 graden Celsius voor 24 uur of langer.Vervang bij het kweken van oudere biofilms dagelijks het medium. Aan het einde van de groeifase van het kruisrijk, was de plaat vijf keer met steriele fysiologische zoutoplossing. Voor ratiometrische pH-beeldvorming, gebruik maken van een omgekeerde confocale laser scanning microscoop met een 63X olie of water onderdompeling lens, een 543 nanometer laserlijn, en een spectrale imaging systeem.Gebruik een couveuse om de microscoop fase warm tot 35 graden Celsius. Stel de detector voor gelijktijdige detectie van groene fluorescentie van 576 tot 608 nanometer en rode fluorescentie van 629 tot 661 nanometer. Kies vervolgens een geschikte laser kracht en krijgen om te voorkomen dat over en onderbelichting.Stel de pinhole grootte in op een luchtunit of een optisch segment van ongeveer 0,8 micrometer. Stel vervolgens de afbeeldingsgrootte in op 512 bij 512 pixels en de scansnelheid op twee. Kies een regelgemiddelde van twee met de optie gemiddelde.Bereid 100 microliter van steriele fysiologische zoutoplossing met 0,4% glucose getitreerd tot pH zeven. Maak vervolgens een voorraadoplossing van C-SNARF-4 en voeg de kleurstof toe aan een uiteindelijke concentratie van 30 micromolar. Leeg een van de putten met de cross-kingdom biofilm waardoor een 20 microliter reservoir en voeg de zoutoplossing met glucose en ratiometrische kleurstof.Plaats de plaat op de microscoop fase en start beeldvorming. Bereid een reeks van 50 millimolar MES buffer getitreerd tot pH vier tot 7,8 in de stappen van 0,2 pH-eenheden bij 35 graden Celsius. Pipette 150 microliters van elke bufferoplossing in de putten van een optische bodem 96-put plaat.Voeg de kleurstof toe aan de met buffer gevulde putten bij een concentratie van 30 micromolar en laat deze gedurende vijf minuten in evenwicht brengen. Verwarm de microscoopfase tot 35 graden Celsius en kies dezelfde instellingen als voor ratiometrische pH-beeldvorming die eerder werd beschreven. Plaats de 96-well plaat op de microscoop fase en focus op de bodem van de putten.Verkrijg groene en rode kanaalafbeeldingen voor alle bufferoplossingen. Maak regelmatig foto's met de laser uitgeschakeld om de detector te corrigeren. Voer het kalibratieexperiment uit in drievoud en exporteer alle afbeeldingen als TIFF-bestanden.Sla de groene en rode afbeeldingen op in afzonderlijke mappen en hernoemen beide reeksen bestanden met opeenvolgende getallen. Importeer de afbeeldingen in speciale beeldanalysesoftware zoals ImageJ. In de foto's genomen met de laser uit, klik analyseren en histogram om de gemiddelde fluorescentie intensiteit te bepalen.Trek deze waarde af van de biofilmafbeeldingen door op proces, wiskunde en aftrekken te klikken. Importeer vervolgens de twee afbeeldingsreeksen in daime en voer een op drempels gebaseerde segmentatie van de rode kanaalafbeeldingen uit door op segment, automatische segmentatie en aangepaste drempelwaarde te klikken. Stel de lage drempel boven de fluorescentie intensiteit van het schimmelcytoplasma en de hoge drempel onder de intensiteit van de schimmelcelwanden en de bacteriën.Breng vervolgens de objectlaag van de gesegmenteerde afbeeldingsreeks over naar de afbeeldingsreeks van het groene kanaal door op segment te klikken en objectlaag over te brengen. Niet-objectpixels verwijderen in de rode en groene kanaalreeks. Nu worden de biofilmbeelden gewist uit bacteriële en schimmelcellen en kunnen ze worden geëxporteerd als TIFF-bestanden.Importeer de afbeeldingsreeks terug in ImageJ en verdeel de rode beeldreeks zelf. Vermenigvuldig vervolgens het resultaat met de oorspronkelijke afbeeldingsreeks. De resulterende beeldreeks is identiek aan het origineel, behalve dat alle pixels met nulintensiteit worden verwijderd.Herhaal het proces met de groene afbeeldingsreeks. Gebruik vervolgens het gemiddelde filter op beide beeldreeksen om detectorruis te compenseren. Verdeel het groen door de rode beeldreeks die vervolgens de groen-rode verhouding voor alle objectpixels oplevert.Robuuste cross-kingdom biofilms ontwikkeld in de put platen na 24 en 48 uur. Enkele cellen en ketens van S.mutans gegroepeerd rond schimmelhyphae en grote intracellulaire ruimten wezen op de aanwezigheid van een volumineuze matrix. De pH in de extracellulaire ruimte werd gevisualiseerd met behulp van een opzoekttafel.De extracellulaire pH in de biofilms daalde snel in de eerste vijf minuten na blootstelling aan glucose. Daarna vertraagde de verzuring meestal het bereiken van waarden van 5,5 tot 5,8 na 15 minuten. Als gevolg van de lokale pH-veranderingen veranderde de fluorescentieintensiteit in de biofilms in de loop van de tijd.Tijdens beeldsegmentatie werden hoge en lage drempels gekozen om alle gebieden die door bacteriële en schimmelcellen worden bestreken adequaat te elimineren. De blauwe en groene gebieden werden geëlimineerd door respectievelijk de lage en hoge drempels. De zorgvuldige keuze van beeldverwervingsparameters is cruciaal om een goed contrast te verkrijgen tussen bacteriële cellen, schimmelcellen en de biofilmmatrix.Als cross-kingdom biofilms worden gekweekt in stroomcellen, kunnen pH-ontwikkelingen worden gevolgd onder stroomconditie die die in de mondholte nabootsen. pH ratiometrie is gebruikt om lokale gebieden in tandheelkundige biofilms te identificeren met een bijzonder lage pH, de zogenaamde acidogene hotspots.
