原発性正常上皮細胞の使用により、細胞内遺伝子および腫瘍抑制遺伝子に特異的な変異を導入することにより、細胞間のクラスタリング制御を用いて、細胞形質転換に必要なゲノム変化を再現的に誘導することが可能になる。マウスにおける短いパリンドロミック反復(CRISPR)ベースのゲノム編集技術。この技術により、マウスを用いてヒトがんに生じる遺伝的変化を正確に模倣することができます。マウスの一次細胞を遺伝子組み換えすることで、がんの発症、進行、治療、診断をより良く研究することができます。本研究では、クレ誘導性Cas9マウス舌上皮細胞を用いて、アデノ関連ウイルス(AAV)をインビトロでゲノム編集することを可能にした。具体的には、正常舌上皮細胞でKRAS、p53、APCを改変することにより、同系マウスの腫瘍性であるプリン頭頸部癌(HNC)細胞株をインビトロで生成した。ここで提示する方法は、特定のゲノム改変を有するHNC細胞株を生成する方法を詳細に説明し、同系マウスにおける腫瘍進行を予測するための適合性を説明する。我々は、この有望な方法がHNCの腫瘍生物学と治療を研究するのに有益かつ有用であろうことを想像する。

このプロトコルは、特定のゲノム操作を伴う頭頸部癌モデルの開発を可能にし、新生物の過程における特定の遺伝子変異の役割に対する我々の理解に大きな影響を与えた。この技術の主な利点は、細胞株が事実上任意の器官から開発することができる容易さである。手順を実証することは、私の研究室の卒業生であるマヌ・プラサドです。まず、手術用はさみを使用して、生後6週間の男性または女性のB6トランスジェニックマウスから舌を収穫することから始めます。メスを使用して組織を非常に小さな断片にミンチし、血清なしで4.5ミリリットルのRPMIプレーン培地を含む15ミリリットルのチューブに入れ、その部分を採取します。チューブに200マイクロリットルのトリプル酵素ミックスを加え、サンプルを37°Cで30分間、10分ごとにチューブをタップしてインキュベートし、組織の酵素解離を強化します。インキュベーションの終わりに、PBSで5%FBSで反応を停止し、70マイクロメートルメッシュストレーナーを介して細胞懸濁液を濾過し、より大きな組織断片から細胞を分離する。遠心分離によって細胞を収集し、完全な培地の3ミリリットルにペレットを再中断します。その後、16ミリメートル皿あたり完全な媒体の3ミリリットルで細胞をプレートします。フィルターの上に保持された細胞凝集体を別の16ミリメートル培養皿に移し、3ミリリットルの完全な培地を加える。異なる細胞コロニーが形成されるまで、両方の料理をインキュベーターに保管してください。1週間培養した後、線維芽細胞汚染の存在について一次細胞培養を顕微鏡的に調べ、02%EDTAで25%のトリプシンを摂氏37度で1分間治療し、線維芽細胞を除去する。培養10日目には、02%EDTAで25%トリプシンを摂氏37度で3〜5分間使用し、細胞懸濁液または細胞凝集培養物から細胞を収穫し、6ウェルプレートの各ウェルに2ミリリットルの培地で2倍の10から5番目の初代細胞を見る。翌日、1ミリリットル当たり10回から12個のウイルスゲノムで細胞をトランスデュースし、ウイルス粒子含有培地中の細胞を摂氏37度で48時間インキュベートする。インキュベーションの終了時に、培養上清を1ウェルあたり2ミリリットルの新鮮な完全培地に置き換え、細胞を細胞培養インキュベーターに戻します。培養の2週間後に、検証および生体内腫瘍化実験のために細胞を播種する。AAV CAS9ベクターを用いて、1ミリリットルのウイルスにつき10〜12個のウイルスゲノムが、実証されたプロトコルを用いて原発性メス細胞を腫瘍化マップに効率的に変換することが決定された。トランスデューシングされた細胞は、エクスプレスクレ、CAS9および緑色蛍光タンパク質である。NOD SCIDマウスの舌、唇および皮膚への5回の10回の注射は、腫瘍を誘発しない一次マップの注入とは異なり、これらの動物における腫瘍形成をもたらす。原発性舌上皮細胞の腫瘍形成は、免疫蛍光およびウェスタンブロッティングを用いて確認することができる。ゲノム解析は、DNAの深いシーケンシングによって、形質転換された細胞から単離され、AAV CAS9系による腫瘍タンパク質53およびAPC遺伝子の有効な遺伝子編集およびフレームシフトを確認する。さらに、形質転換された舌細胞は、野生型C57黒6匹のマウスにおいて効率的に腫瘍を形成する。新生腫瘍細胞の免疫組織化学的解析は、E-カドヘリンおよびケラチン14の細胞質発現を明らかにする。この手順を試みるとき、ミンチが多すぎたり、酵素反応が多すぎたりすると、細胞の生存率が低下し、ウイルストランザクション効率が低下することを常に覚えておいてください。この手順を使用して、細胞株を遺伝子組み換えして、対象となるあらゆる組織から癌細胞の腫瘍化および転移性ポテンシャルに関する洞察を得ることができる。
