内源性心脏再生是一个动态过程，涉及多种细胞类型协同工作，以进行强大的再生反应。通过清除的心脏的3D成像对损伤进行窥视到再生组织中，可以帮助重建指导这种心脏再生反应的细胞动力学。我们的协议是专门设计用于跟踪细胞在开发和再生期间。我们预计，在将我们的清除、染色和成像技术广泛应用于其他器官类型方面没有限制。对于LAD诱导的冠状动脉闭塞，首先用防腐剂解对麻醉一天大的新生儿幼崽的手术区进行消毒。在胸部区域做一个横向皮肤切口。要将皮肤与肌肉分开，请使用一双敷料钳小心地抬起皮肤，同时将剪刀轻轻按在闭合位置的间肌肉上。找到第四个成本间空间，并使用小钳子，使一个小的表面穿刺注意避免内脏器官。使用钝解剖和敷料钳，扩大腹间肌肉之间的区域，直到手指可以应用到腹部的左侧，同时用修整钳将腹间空间打开，轻轻地将心脏从胸腔中引导出来。一旦心脏被提取，让心脏休息在成本间肌肉和定位LAD作为心脏区域与较少汇集的血液。为了诱发心肌梗塞，使用6-0缝合线将一个方结紧紧地系在血管周围两次，而不会切断动脉。应立即观察顶点时发白。对于类似的梗塞尺寸，在沿 LAD 的缝合位置上保持一致非常重要。当血液仍然流经动脉时，通过快速访问心脏，最好实现此目的。缝合后，让心脏滑回胸腔。用一个外科医生和一个方结缝合肋骨，用钝钳抬起上一组肋骨，同时通过上部和下部的肋骨进行 6-0 缝合。将少量皮肤胶涂抹在上腹部，并使用细钳将下腹部的皮肤放在裸露的胸部区域。然后把小狗放在加热垫上，对下一只动物进行结扎。当所有的手术都进行了，把温暖和移动的幼崽还到母亲的笼子里。为了被动地清除收获的心脏组织，21天后进行手术后，确认对手趾捏没有反应，将鼠标放在一个干净的手术区域，在上部位置，用胶带固定四肢。使用组织钳子，抓住西腓工艺正下方的毛皮，并作出跨越肋骨宽度的切口。沿着肋骨的侧部切割，并使用钳子提起西腓过程以暴露隔膜。用弯曲的钳子分离隔膜，将组织颅骨拉扯，直到跳动的心脏被接近。使用弯曲的钳子抓住底部的心脏，并使用虹细胞切除术剪刀切割大奥和优越的维纳卡瓦。将跳动的心脏放入 PBS 的培养皿中，以便心脏泵出任何保留的血液。心肌梗塞可以通过LAD结扎的正确解剖位置来证实。当心脏停止跳动时，用钳子轻轻挤压器官以排出残余血液，将标本转移到含有两毫升 PBS 的一次性 2.5 毫升玻璃壳瓶中。将小瓶放在摇床上至少 30 分钟，在室温下每 10 分钟更换一次 PBS 溶液，直到溶液保持血液。当PBS是明确的，更换PBS与两毫升冷4%对甲醛在室温下孵育4小时，然后三个10分钟的洗涤在摇床与两毫升新鲜PBS每次洗涤。最后一次洗涤后，用两毫升4%丙烯酰胺加满小瓶，并辅以0.5%VA-044溶液，在4摄氏度下进行过夜孵化。第二天，把小瓶放在37摄氏度的高温块里三个小时。在聚合结束时，将心脏转移到一个新的玻璃壳瓶中，并在两毫升新鲜PBS中清洗心脏组织，每次孵化结束时刷新PBS。当 PBS 清除时，用两毫升的清除溶液更换洗涤缓冲液。在37摄氏度下孵育标本数周，直到心脏清晰，每两到三天刷新一次清除溶液。当器官清晰时，在摇摇器上用 PBS 清洗试样，如演示，然后用新鲜的 PBS 重新填充小瓶，在 37 摄氏度下进行 24 小时孵育。第二天，用RIMES代替PBS，在37摄氏度下进行24小时孵育。在孵育结束时，组织完全清除。几周后，组织应该变得透明。在心肌梗塞诱导后，为已清除的产后小鼠心脏进行成像，用PBS填充定制凹陷滑梯的半室，并使用弯曲钳子小心地将清除的心脏转移到腔室中。用 PBS 填充腔室的剩余体积。然后，在腔室中填充 PBS，直到液体表面在腔室顶部形成圆顶，然后安装盖幻灯片，然后根据标准成像方案通过共声显微镜对组织进行成像。如果穿刺和钝解剖离胸骨太近，心脏可能无法离开胸腔。如果心脏不轻松离开腔腔，向左腹部施加压力，以方便手术。将心脏放在心间肌肉上可能会出现并发症。因此，在水平方向上将钝解剖保持最小大小非常重要，以便实现LAD的清晰可视化和可访问性。请注意，表面结扎在最终缝合放置中调整的空间较小，并且应通过可控的稳定运动执行围绕 LAD 的缝合。到清除步骤结束时，整个心脏应始终不透明，中心内无变色。在室温下 RIMS 溶液中几天后，心脏应完全清除，并可能观察到一些组织扩张。正如tdTomato记者观察到的蛋白质表达，心脏神经主要是肤浅的，一些人群居住在史诗层。重要的是，记者蛋白表达确认保留在经过清晰协议后，如证明。3D心脏的清理和成像为心脏细胞群的位置和形态提供了视角。这使我们能够识别细胞模式和细胞-细胞相互作用的新方面。
