Kwantitatieve methoden voor de evaluatie van verhoogde GSK-eiwit concentraties zijn nuttige hulpmiddelen bij de karakterisering van de fysiologische en pathologische toestand van het CZS. Afgezien van een betrouwbare kwantificering van het CSF-eiwitgehalte, vereist de opsporing van CSF-eiwitten echter een uitdrukking van de resultaten die discrimineert tussen in bloed en CNS afgeleide fracties in het CSF. Cerebrospinale vloeistof (CSF), een heldere en kleurloze vloeistof rond de hersenen en het ruggenmerg, houdt grote klinische en fundamentele wetenschappelijke betekenis. De CSF behoudt de elektrolytische omgeving van het centrale zenuwstelsel (CNS), balanceert de systemische zuur-base status, levert voedingsstoffen aan neuronale en gliale cellen, functioneert als een lymfatisch systeem voor het CZS, en vervoert hormonen, neurotransmitters, cytokines en andere neuropeptiden in het CZS1. Dus, aangezien de CSF-samenstelling de activiteit van het CZS weerspiegelt, biedt deze vloeistof een waardevolle, hoewel indirecte, toegang om de fysiologische en pathologische toestand van het CZS te karakteriseren.
CSF is gebruikt voor de diagnose van aandoeningen die invloed hebben op het CNS voor meer dan honderd jaar, en voor de meeste van deze tijd, het werd voornamelijk bestudeerd door clinici als een diagnostisch instrument. In de afgelopen jaren hebben neuro biologen echter het potentieel van CSF erkend voor het bestuderen van de pathofysiologie van het CZS. In het bijzonder zijn er verschillende eiwitanalyse hulpmiddelen met hoge doorvoer geïntroduceerd in het neurowetenschaprijk, waardoor een gedetailleerde studie van de eiwit samenstelling van het CSF mogelijk is, met de verwachting dat deze analyse kan helpen om inzicht te geven in de dynamische veranderingen die zich binnen het CZS voordoen.
Technologische ontwikkelingen in multiplex immunoassay technieken zoals luminex en simoa Technologies2,3, bieden onderzoekers vandaag de mogelijkheid om honderden eiwitten te detecteren bij zeer lage concentraties. Bovendien maken deze zelfde technologieën het gebruik van kleine monsterhoeveelheden mogelijk, waardoor studies bij kleine dieren, waaronder muizen, worden bevorderd, waarbij beperkte monstervolumes van CSF tot voor kort de gedetailleerde kenmerken van de vloeistof hebben uitgesloten.
Niettemin is een voorbehoud dat eiwitten gemeten in CSF kunnen voortvloeien uit intrathecale synthese en/of transudatie uit het serum als gevolg van een beschadigde bloed-hersen interface (BBI). Helaas, eiwitanalyse van CSF alleen kan alleen de som van deze twee componenten te bepalen. Om onderscheid te maken tussen transudaat en intrathecaal geproduceerde eiwitten, moeten de CSF-eiwit metingen met behulp van een beschikbare proteïne-Analysetool worden aangepast voor individuele variabiliteit in serumconcentraties en barrière-integriteit. Hoewel deze aanpassing vaak wordt gebruikt in de klinische praktijk, bijvoorbeeld de CSF IgG-index, die een hoge gevoeligheid heeft voor het opsporen van intrathecale IgG-synthese4,5,6, hebben tot op heden zeer weinig onderzoeken de CSF-eiwit concentraties gecorrigeerd voor de serumconcentratie en barrière-integriteit7,8.
Momenteel is de Reibergram aanpak de beste manier om de barrièrefunctie en intrathecale synthese van eiwitten te bepalen. Het is een grafische evaluatie in CSF/serum quotiënt diagrammen die, op een geïntegreerde manier, zowel de barrière (dys) functie en intrathecale eiwitsynthese analyseert, verwijzend naar een uitsluitend bloed-afgeleid eiwit9,10. De zeer overvloedige eiwit albumine wordt meestal gekozen als referentie-eiwit omdat het alleen in de lever wordt geproduceerd en omdat de grootte, ongeveer 70 kDa, intermediair is tussen kleine en grote eiwitten11. Het analyseschema werd voor het eerst gedefinieerd door Reiber en Felgenhauer in 1987 voor de belangrijkste klassen van immunoglobulinen (IGS)11, empirisch gebaseerd op de resultaten verkregen uit de analyse van duizenden menselijke monsters9. De aanpak werd vervolgens bevestigd door de toepassing van de twee Fick-wetten van diffusie in de theorie van de Moleculaire diffusie/debiet12. Een dergelijke theorie toont aan dat de diffusie van een eiwit door de barrière een hyperbolische verdeling heeft en de dynamiek van eiwitten in het CZS9,13kwantificatief kan verklaren. Over het algemeen is het voordeel van het gebruik van het Reibergram voor het aantonen van intrathecale eiwitsynthese dat het gelijktijdig de eiwitfractie identificeert die de CSF uit het serum binnenkomt, evenals de hoeveelheid eiwit die in het CSF wordt aangetroffen vanwege lokale productie.
Het huidige artikel en het bijbehorende protocol beschrijven de hele procedure, van CSF en bloedafname tot de uiteindelijke berekeningen die de GSK-eiwit niveaus corrigeren, voor de kwantitatieve meting van intrathecale eiwitsynthese in muismodellen van neurologische Aandoeningen. Deze procedure vormt een basis voor de beoordeling (1) van de pathofysiologische oorsprong van elk CSF-eiwit en (2) de stabiliteit en de functionele betekenis van de barrière-integriteit. quantitative measurement of intrathecally synthesized proteins in mice

Cerebrospinale vloeistof (CSF), een vloeistof gevonden in de hersenen en het ruggenmerg, is van groot belang voor zowel de basis-als de klinische wetenschap. De analyse van de CSF-eiwit samenstelling levert cruciale informatie in fundamenteel neurowetenschappelijk onderzoek en neurologische ziekten. Een voorbehoud is dat eiwitten gemeten in CSF kunnen voortvloeien uit intrathecale synthese en transudatie uit serum, en eiwitanalyse van CSF kan alleen de som van deze twee componenten bepalen. Om onderscheid te maken tussen eiwit transudatie uit het bloed en intrathecaal geproduceerde eiwitten in diermodellen en bij mensen, de metingen van het CSF-eiwit profiel met behulp van conventionele eiwitanalyse hulpmiddelen moeten de berekening omvatten van het albumine CSF/serum quotiënt (Q-albumine), een marker van de integriteit van de bloed-hersen interface (BBI) en de eiwit index (q-eiwit/q-albumine), een schatting van intrathecale eiwitsynthese. Dit protocol illustreert de gehele procedure, van CSF en bloedinzameling tot quotiënten en indices berekeningen, voor de kwantitatieve meting van intrathecale eiwitsynthese en de beperking van BBI in muismodellen van neurologische aandoeningen.

Dit protocol maakt csf en bloedverzameling in de kwantitatieve correctie van CSF-eiwitniveaus mogelijk om te meten in een thecale eiwitsynthese in muismodellen van neurologische aandoeningen. Deze procedure biedt een basislijn, waartegen de pathofysiologische oorsprong van CSF-eiwitten van belang, en de stabiliteit en functionele betekenis van het bloed CSF gunst integriteit kan worden beoordeeld. De analyse van interthecale eiwitsynthese en barrièreintegriteit kan worden toegepast op andere diermodellen en menselijke studies.Bijvoorbeeld om te controleren op ziekten van het centrale zenuwstelsel. Het verzamelen van aanzienlijke hoeveelheden schone CSF kan technisch uitdagend zijn bij muizen, dus het beoefenen van de techniek totdat grote hoeveelheden niet-verontreinigd monster kan worden verkregen, wordt geadviseerd. Neem de chirurgische plaats met 30%chlorhexidine diacetaat, en maak een sagittale huid incisie inferieur aan de occiput om de spieren boven de cisterna magna bloot te stellen. Met behulp van tangen ontleedt stompe het onderhuidse weefsel en de spieren en gebruikt u micro-oprolmechanismen om de spieren uit elkaar te houden om de dura mater meningeal laag over de cisterna magna bloot te leggen.Was het weefsel voorzichtig met steriele PBS om eventuele bloedverontreiniging te verwijderen en gebruik een steriel wattenstaafje om de dura mater droog te deppen. Het plaatsen van de eerste punctie in de cisterna magna is essentieel voor het verkrijgen van overvloedige, niet-verontreinigde CSF. Met behulp van een naald van 30 meter prik je voorzichtig het membraan dat de koektuur magna bedekt en steek snel en voorzichtig een kleine glazen capillaire buis in de punctie.Wanneer vijf tot 12 microliter csf zijn verzameld, verwijder dan voorzichtig de buis van het membraan en gebruik een stuk polyethyleenbuizen om de buis aan te sluiten op een spuit van drie milliliter. Injecteer de verzamelde CSF in een gelabelde 500 microliter buis op ijs, en gebruik een wegwerpnaald en gevarieerde polydioxanon hechtingen om de incisie te sluiten. Reinig het gebied van een gedroogd bloed of weefsel, en plaats de muis in een schone, warme kooi met controle tot volledige recumbency.Verzamel de CSF door centrifugatie, en visueel inspecteren van de pellet en supernate het voor eventuele tekenen van bloedverontreiniging. Breng vervolgens met behulp van schone pipettechniek de CSF-bevattende supernaat over in een nieuwe 200 microliterbuis en verdun de CSF bij een één-drie-verhouding met PBS voor onmiddellijke opslag bij 80 graden Celsius. Voor serumverzameling door hartpunctie, onmiddellijk na de csf-verzameling, zoals zojuist is aangetoond, plaatst u de muis in de supinepositie.Watstaaf de buikhuid met 70% alcohol. Gebruik een schaar om de borstholte te openen, het hart bloot te stellen en een 25-meter naald in te voegen die aan een 3 milliliter spuit in de linkerventrikel zit. Breng vervolgens voorzichtig negatieve druk uit op de spuitzuiger en trek de naald terug nadat het bloed is verzameld.Druk de zuiger om het verzamelde bloed uit te werpen in een 1,5 milliliter flesje. Laat het bloed 30 tot 60 minuten stollen bij kamertemperatuur voordat u het serum scheidt door centrifugatie. Breng het serum vervolgens met behulp van een schone pipettechniek over in een nieuw gelabeld 500 microliter flesje voor onmiddellijke opslag bij 80 graden Celsius.Om de doeleiwitten en albumen te kwantificeren in overeenkomende serum- en CSF-exemplaren, gebruikt u een standaard eiwitkwantificeringstest, waarbij standaardeiwitten worden gebruikt om een standaardcurve voor elk eiwit van belang te bereiden. Na de analyse exporteer je de ruwe gegevens naar een geschikt softwaregrafiekprogramma en grafiek de intensiteit van de detectiesignaalfluorescentie ten opzichte van de standaard eiwitconcentraties om een standaardcurve te creëren voor elk eiwit van belang. Gebruik vervolgens de standaardcurven om de concentraties van elke analyt van belang in de monsters te berekenen.Ten slotte gebruikt u albumen en richt u op analytconcentraties om Q-waarden en intrathecale index te berekenen. De werkelijke niveaus van de totale IgG zijn aanzienlijk verhoogd in het CSF van twee geteste knaagdieren modellen van multiple sclerose, in vergelijking met de overeenkomstige leeftijd afgestemd sham controles. R-EAE muizen tonen aanzienlijk verbeterde albumen quotiënt waarden, wat wijst op een verhoogde doorlaatbaarheid van de bloedherenbarrière in deze muizen.Nonversely, geen verschillen in albumen quotiënt bestaan tussen TMEV-IDD en sham muizen, bevestiging van eerdere bevindingen van een intacte barrière in TMEV-IDD muizen. Bovendien worden bij TMEV-IDD-dieren aanzienlijk hogere IgG-indexwaarden en dus intrathecale IgG-productie waargenomen. De berekening van een eiwitindex vergemakkelijkt de identificatie van nieuwe eiwitbiomarkers die nuttig zijn voor vroegtijdige diagnose, uitkomstvoorspelling en ziektecursusmonitoring voor zowel neuro-inflammatoire als neurodegeneratieve ziekten.
