Este protocolo permite o estudo do câncer de pâncreas no local do tecido nativo, minimizando a inflamação durante a injeção de células tumorais e fornece um método de alta produtividade para gerar grandes coortes de camundongos. Por meio de orientação de ultrassom, a necessidade de cirurgia abdominal é eliminada. Em nossas mãos, o UG-OTIM tem uma taxa muito menor de semeadura de parede peritoneal em comparação com a implantação ortotópica tradicional.
O UG-OTIM produz tumores que recapitulam características histológicas e imunobiológicas características do micro ambiente tumoral pancreático e, portanto, podem ser usados para investigar novas combinações terapêuticas em um cenário clinicamente relevante. Desenvolvemos este protocolo para terapias de imuno-oncologia e biologia do câncer, mas ele poderia ser usado em outras áreas de pesquisa, incluindo investigações de função pancreática normal ou estados de doenças como diabetes. A imagem de ultrassom em tempo real da inserção e injeção da agulha é parte integrante deste método.
Assim, a demonstração visual do passo é fundamental para a técnica adequada. Antes de iniciar o procedimento, suspendemos a linha de interesse da célula adenocarcinoma do ductile pancreático em um volume apropriado de PBS estéril e frio no gelo. Coloque as gaiolas em uma placa de aquecimento Celsius de 37 graus e limpe completamente o armário de segurança biológica, a câmara de indução e o estágio de ultrassom com um esterilizador apropriado.
Defina a função de aquecimento do estágio de ultrassom para 37 graus Celsius e após a indução da anestesia, aplique pomada aos olhos do primeiro animal. Coloque o rato em recumbência dorsal no estágio de ultrassom e proteja suavemente as extremidades superior e inferior ao palco com fita. Use um aplicador de ponta de algodão estéril para aplicar uma generosa camada de creme depilatório no quadrante superior esquerdo do abdômen sobre a região do baço até a linha média.
Após um minuto, use uma almofada de gaze seca para remover suavemente o cabelo antes de remover o excesso de creme depilatório com uma gaze úmida salina. Em seguida, coloque o mouse em uma das novas gaiolas limpas no aquecedor. Para implantação de células tumorais guiadas por ultrassom, ajuste a plataforma de ultrassom para que a superfície seja paralela ao chão, e fique no lado esquerdo do animal com a cabeça do rato voltada para a direita.
Ajuste a posição do transdutor para que uma imagem abdominal transversal seja obtida e fixe os membros do mouse na plataforma com fita. Abaixe suavemente o transdutor para entrar em contato com o abdômen do rato e ajuste o transdutor até que o pâncreas esteja claramente visível. Localize o rim esquerdo e o baço para fornecer uma orientação precisa na cavidade abdominal.
Quando o local da injeção estiver localizado, carregue uma seringa de insulina de 29 gage de 1/2 polegada com 25 micro litros da suspensão da célula tumoral, e limpe a ponta da agulha com uma almofada de preparação de álcool estéril. Use fórceps de borda sem corte para agarrar a pele do rato e a parede peritoneal para aumentar a tensão no local da injeção. Segurando a seringa em um ângulo de aproximadamente 25 a 45 graus para a superfície da plataforma de ultrassom, avance lentamente a agulha através da pele e da parede peritoneal.
Confirme se a agulha perfurou a parede peritoneal antes de utilizar a visualização do ultrassom para guiar a agulha diretamente para o pâncreas. Quando a agulha estiver no lugar, injete lentamente as células tumorais. A formação de um bolus fluido dentro do pâncreas será evidente em um pâncreas injetado corretamente por ultrassom.
Uma vez que o volume total de suspensão tenha sido injetado, e o bolus fluido possa ser observado por ultrassom, mantenha a seringa muito parada por vários segundos antes de retirar lentamente a agulha do abdômen do rato, tomando cuidado para não perturbar as células injetadas. Em seguida, coloque o mouse em uma gaiola nova e limpa no aquecedor com monitoramento até a recuperação completa. A colocação adequada da agulha no pâncreas, incluindo profundidade e ângulo da agulha, é um passo crítico neste protocolo.
A visualização do bolus fluido ajuda a confirmar uma injeção tumoral bem sucedida. Controlar a profundidade da injeção é o aspecto mais difícil. Meu conselho é praticar com injeções de tripamboo e confirmar o bolus fluido tanto pelo ultrassom quanto pela necropsia.
O monitoramento da implantação e taxa de crescimento do tumor por ultrassom semanal revela tumores que estão contidos nas bordas do pâncreas durante todo o período experimental. Injeções tumorais de alto nível resultam em uma maior proporção de animais portadores de tumores três semanas após a injeção em comparação com a baixa coorte de titer. Apesar da demora no aparecimento do tumor, a taxa global de crescimento do tumor não é significativamente diferente entre as duas doses.
Da mesma forma, embora a taxa de sobrevivência entre as duas coortes não seja significativamente diferente, elas tendem a tendência para uma sobrevivência ligeiramente melhorada na baixa coorte de titer. A coorte de alto título também produz uma maior proporção de camundongos com tumores inlroláveis em estudos pré-clínicos, em quatro semanas após a injeção. Após o sacrifício, a anatomia bruta dos tumores modelo de implantação de tumores ortotópicos guiados por ultrassom é semelhante à dos tumores Espontâneos de KPC e a análise histológica demonstra um padrão de estruturas ductais anormais semelhantes em ambos os modelos e que recapitula a morfologia da doença humana.
Em ambos os modelos, os tumores são mal infiltrados por Células T, mas altamente infiltrados por macrófagos. Lembre-se de segurar a seringa no ângulo apropriado para permitir uma entrada suave no pâncreas, e para confirmar que a agulha está no pâncreas antes de prosseguir com a injeção. O crescimento tumoral pode ser monitorado e os tumores podem ser colhidos para análise por pho citometria ou imunohistoquímica para determinar o impacto de uma intervenção.
